Prevalence and progression of monoclonal gammopathy of undetermined significance and light-chain MGUS in Germany.

We determined the prevalence and progression rate of monoclonal gammopathy of undetermined significance (MGUS) and light-chain MGUS (LCMGUS) in Germany utilizing the biobank of the population-based Heinz Nixdorf Recall Study. The Heinz Nixdorf Recall Study comprises 4,814 men and women aged 45-75 years. To detect monoclonal proteins, standard serum electrophoresis was combined with parallel screening immunofixation using pentavalent antisera. Additionally, free light chains (FLC) were measured in all samples. Definition of MGUS included M-protein concentration, laboratory results, and disease

High rates of Rickettsia parkeri infection in Gulf Coast ticks (Amblyomma maculatum) and identification of "Candidatus Rickettsia andeanae" from Fairfax County, Virginia.

The Gulf Coast tick, Amblyomma maculatum, is a vector of Rickettsia parkeri, a recently identified human pathogen that causes a disease with clinical symptoms that resemble a mild form of Rocky Mountain spotted fever. Because the prevalence of R. parkeri infection in geographically distinct populations of A. maculatum is not fully understood, A. maculatum specimens collected as part of a tick and pathogen surveillance system in Fairfax County, Virginia, were screened to determine pathogen infection rates. Overall, R. parkeri was found in 41.4% of the A. maculatum that were screened. Additional

Developing an integrated proteo-genomic approach for the characterisation of biomarkers for the identification of Bacillus anthracis.

Bacillus anthracis is the causative agent of anthrax, an acute and often fatal disease in humans. Due to the high genomic relatedness within the Bacillus cereus group of species it is a challenge to identify B. anthracis consistently. Alternative strategies such as proteomics coupled with mass spectrometry (MS) provide a powerful approach for biomarker discovery. However, validating and evaluating these markers, particularly for genetically homogeneous species such as B. anthracis are challenging. The objective of this study is to develop a robust biomarker discovery and validation pipeline, u

GPCR antitarget modeling: pharmacophore models for biogenic amine binding GPCRs to avoid GPCR-mediated side effects.

G protein-coupled receptors (GPCRs) form a large protein family that plays an important role in many physiological and pathophysiological processes. However, the central role that the biogenic amine binding GPCRs and their ligands play in cell signaling poses a risk in new drug candidates that reveal side affinities towards these receptor sites. These candidates have the potential to interfere with the physiological signaling processes and to cause undesired effects in preclinical or clinical studies. Here, we present 3D cross-chemotype pharmacophore models for three biogenic amine antitargets

Salidroside inhibits extracellular matrix deposition and oxidative stress in human trabecular meshwork cells by suppressing the TNF signaling pathway and upregulating MMP3 expression.

BACKGROUND: The functional integrity of human trabecular meshwork cells (HTMCs) is essential for regulating intraocular pressure (IOP). In primary open-angle glaucoma (POAG), pathological stimuli induce oxidative damage and promote excessive extracellular matrix (ECM) deposition in HTMCs, thereby hindering aqueous humor outflow. Salidroside (Sal), a compound with diverse pharmacological activities, has been reported to inhibit oxidative stress and reduce ECM accumulation. This study aimed to investigate whether Sal exerts its antioxidant effects by regulating matrix metalloproteinase 3 (MMP3)

CancerHERVdb: Human Endogenous Retrovirus (HERV) Expression Database for Human Cancer Accelerates Studies of the Retrovirome and Predictions for HERV-Based Therapies.

In this study, we sought to create a database summarizing the expression of human endogenous retroviruses (HERVs) in various human cancers. HERVs are suitable therapeutic targets due to their abundance in the human genome, overexpression in various malignancies, and involvement in various cancer pathways. We identified articles on HERVs from PubMed and then prescreened and automatically categorized them using the portable document format (PDF) data extractor (PDE) R package. We discovered 196 primary research articles with HERV expression data from cancer tissues or cancer cell lines. HERV RNA

A Viral Mutation Profiling and Discovery Strategy for Sensitive Multiplex Detection of Viruses and Variants in Saliva by Proteomics.

BACKGROUND: Detecting multiple viruses in biofluids such as saliva remains challenging because of rapid mutation and low abundance presenting in the complex matrix. Proteomics analysis provides a complementary method by directly measuring viral peptides and resolving protein-level sequence variation in rapidly evolving viruses such as influenza and SARS-CoV-2. However, the ongoing viral evolution challenges search sensitivity by introducing numerous mutations. Here, we develop a saliva viral proteomics strategy that integrates mutation profiling of clinically relevant viral sequences with sing

Effect of Perioperative Glutamine-Enhanced Parenteral Nutrition on Short-Term Outcomes in Colorectal Cancer Patients Undergoing Radical Resection: A Systematic Review and Meta-Analysis of Randomized Controlled Trials.

BackgroundParenteral glutamine (Gln) supplementation can enhance the immune function of patients with colorectal cancer(CRC), regulate inflammatory response, nitrogen balance and protein synthesis, reduce the morbidity of postoperative complications and shorten the length of hospitalization. However, some guidelines and clinical studies have questioned the rationality of using glutamine-containing immunonutrition support in the perioperative period of CRC. Therefore, we conducted a meta-analysis of the effects of perioperative glutamine-enriched parenteral nutrition on short-term postoperative

Penicillin-resistant Streptococcus dysgalactiae subsp. equisimilis and the association with penicillin-binding protein mutations: a population genotypic and phenotypic study.

Streptococcus dysgalactiae subspecies equisimilis (SDSE) is increasingly recognized as a cause of human disease. Despite decades of penicillin use, SDSE is near universally penicillin-susceptible. Here we report and characterize by whole genome sequencing (WGS) three penicillin-resistant SDSE isolates (minimum inhibitory concentration [MIC] ≥0.256 mg/L) isolated from blood and skin in an individual with recurrent cellulitis on penicillin prophylaxis. The isolates carried 11 penicillin-binding protein (PBP) PBP2x, five PBP1a, and one PBP2b mutations that were distinct from the only previously r

Computational discovery of co-expressed antigens as dual targeting candidates for cancer therapy through bulk, single-cell, and spatial transcriptomics.

MOTIVATION: Bispecific antibodies (bsAbs) that bind to two distinct surface antigens on cancer cells are emerging as an appealing therapeutic strategy in cancer immunotherapy. However, considering the vast number of surface proteins, experimental identification of potential antigen pairs that are selectively expressed in cancer cells and not in normal cells is both costly and time-consuming. Recent studies have utilized large bulk RNA-seq databases to propose bispecific targets for various cancers. However, co-expressed pairs derived from bulk RNA-seq do not necessarily indicate true co-expres

Correlation between metastasis-associated gene 1 expression and tumor-associated macrophages in non-small cell lung cancer.

The role of metastasis-associated gene 1 (MTA1) in the metastasis of non-small cell lung cancer (NSCLC) has been proved, but its role in the tumor microenvironment is still insufficient. The study was performed to explore the correlation between MTA1 and tumor-associated macrophages (TAMs) in NSCLC. The expression profile data of lung adenocarcinoma (LUAD) and lung squamous cell carcinoma (LUSC) were downloaded from TCGA database. The tumor-infiltrating immune cells in each LUAD and LUSC patient were estimated using the CIBERSORT method. Then, the online TIMER database containing multiple algo
