Bifidobacterium pseudolongum enriched by arctigenin ameliorates cough variant asthma via the acetate-aryl hydrocarbon receptor-interleukin-33 axis.

Gut microbiota dysbiosis is significantly associated with the onset and progression of cough variant asthma (CVA). Targeting and improving gut microbiota dysbiosis may serve as a promising strategy for CVA. Suhuang antitussive capsule (SH) is the sole clinically approved traditional Chinese patent medicine specifically indicated for managing CVA. Herein, we initially identified arctigenin (AG) as one of the effective components detected and preliminarily screened from SH for its potential in treating CVA. AG significantly ameliorates ovalbumin (OVA)-induced CVA symptoms, alleviates gut dysbios

Apple derived extracellular vesicles as positive modulators of glial inflammation and gut-brain axis signaling.

Plant-derived extracellular vesicles (PDEVs) are emerging as bioactive dietary particles with the capacity to modulate mammalian physiology. Here, we characterize the structure and functional properties of apple-derived extracellular vesicles (ADEVs) and evaluate their relevance to neuroimmune and gut-brain communication. ADEVs exhibit canonical PDEV features and elicit rapid IP₃-dependent Calcium (Ca²⁺) signaling in fibroblasts while preserving blood-brain barrier integrity. Neural assays reveal marked cell-type specificity: ADEVs are efficiently internalized by glial cells and activate glial

A newly discovered Aerococcus urinae mediates transfer of the pCF10 plasmid via SPI-WT regulation.

INTRODUCTION: Pheromone-regulated horizontal transfer serves as the core mechanism for horizontal gene transfer of antibiotic resistance genes, playing a pivotal role in driving the spread of resistance. Given the strict species-specific constraints of this regulatory system, it is imperative to determine whether novel regulatory signal peptides and cross-genus receptors responsive to these signals exist, thereby elucidating its potential for disseminating resistance across broader microbial communities. METHODS: This study isolated and screened a Gram-positive coccus, Aerococcus urinae Ae1,

Molecular Mechanisms Associated with Metabolic Dysfunction: Contributions of Nutritional Genomics.

Nutritional genomics has expanded our understanding of how dietary exposures interact with genetic and epigenetic mechanisms involved in metabolic dysfunction. In this context, metabolic dysfunction associated with excessive visceral adiposity arises from a multifaceted interaction between systemic inflammation, insulin resistance, and inter-individual biological susceptibility. Obesity, particularly when driven by diets rich in saturated fatty acids, disrupts intestinal homeostasis, thereby triggering metabolic endotoxemia and contributing to low-grade systemic inflammation and adipose tissue

A deformable Bacillus subtilis with gut homing for tumor treatment via bacterial metabolism-facilitated synergistic approach for starvation, chemotherapy, and immunotherapy.

Cancer treatment remains a critical global health challenge, which often requires multimodal and targeted therapeutic strategies, yet integrating these modalities typically demands complex formulations. Oncolytic bacterial therapy offers unique opportunities for targeted drug delivery and immune modulation, yet clinical translation has been hindered by safety concerns and tedious engineering modification. Here, we report a deformable Bacillus subtilis (DBS), constructed through one-step cisplatin-induced morphological transformation that elongates the bacterium while simultaneously incorporati

Targeting efferocytosis for tissue regeneration: From microenvironment reprogramming to clinical translation.

Efferocytosis, phagocytic clearance of apoptotic cells (ACs), is an essential biological process that resolves inflammation and regulates tissue regeneration in various organ systems. Through removal of apoptotic cell debris, efferocytosis attenuates secondary necrosis and dampens the release of damage-associated molecular patterns (DAMPs). More importantly, it can reprogram phagocytes towards a pro-reparative phenotype via the secretion of anti-inflammatory mediators, metabolic rewiring, and the production of growth factors. There are four closely regulated stages in the entire process: "find

Innate immune recognition of Debaryomyces hansenii requires Dectin-1-Card9 signaling.

Innate immune signaling plays a key role in host response to infection, yet the pattern recognition receptors that detect non-model gut-associated yeasts remain poorly defined. Here, we investigated macrophage sensing of Debaryomyces hansenii, a food-derived yeast that we found to be enriched within intestinal ulcers of Crohn disease (CD) patients. Using a cell surface receptor antibody screen of bone marrow-derived macrophages infected with a CD patient isolate of D. hansenii, we showed that D. hansenii-induced macrophage activation characterized by increased expression of co-stimulatory mole

Response of the mTOR signaling pathway to the reproductive dysfunction of dysbiosis-induced male mice.

The mammalian target of rapamycin (mTOR) signaling pathway plays a key role in coordinating several cellular activities in response to environmental signals, thereby influencing metabolism and reproduction. This study aimed to determine the role of mTOR signaling pathway in testicular dysfunction caused by intestinal microbiome imbalance. An antibiotic mixture containing ampicillin, neomycin sulfate, vancomycin, and metronidazole was administered orally to mice for four weeks, and subsequently serum, and tissues were obtained following sacrifice. Glucose, cholesterol, and sex hormone levels we

The Duodenal Mucosa Plays a Key Role in Metabolic Regulation: Emerging Physiology and New Opportunities for Cell-Directed and Barrier-Related Interventions for the Treatment of Diabetes and Obesity.

The duodenum has historically been regarded as a passive digestive conduit for nutrient transit and absorption. However, emerging experimental and clinical evidence supports a revision of this view. In a contemporary model, the proximal small intestine also has roles as a metabolic sensor and endocrine signalling hub that exerts a powerful influence over systemic metabolism. The duodenum contains the highest concentration of enteroendocrine cells in the gastrointestinal tract, secreting a variety of hormones that regulate digestive processes and metabolic networks. In addition, a dense bidirec

Evaluation of the immunomodulatory effects of novel phosphinic acid derivatives in vitro and in a murine colitis model.

T cells and cytokines are key contributors to the pathogenesis of inflammatory bowel disease (IBD), where dysregulated immune activation drives chronic intestinal inflammation. Phosphinic acid compounds are biologically active molecules that function as enzyme inhibitors and have been investigated for diverse pharmacological applications, including their potential therapeutics in inflammatory diseases. In this study, we investigated the immunomodulatory effects of seven novel phosphinic acid derivatives, including PAC-tryptophan (PAC-Trp), PAC-leucine (PAC-Leu), PAC-proline (PAC-Pro), PAC-phen

DEV infection induces endoplasmic reticulum stress and activates inflammatory responses in duck intestinal epithelial cells.

Duck enteritis virus (DEV), a member of the Herpesviridae family, causes an acute and highly contagious disease in waterfowl characterized by vascular injury and severe gastrointestinal lesions. Although DEV infection is known to disrupt cellular homeostasis, the interplay between DEV-induced endoplasmic reticulum (ER) stress and host inflammatory responses in intestinal epithelial cells (IECs) remains poorly understood. We developed an in vitro model using DEV infection in primary duck IECs to study the activation of the unfolded protein response (UPR) and its association with downstream immu

Gut-engineered Bacillus subtilis-mediated BAMBI delivery for the treatment of thioacetamide-induced liver fibrosis through mechanotransduction inhibition.

Liver fibrosis, driven by chronic injury and excessive extracellular matrix (ECM) deposition, lacks effective clinical therapies. This study pioneers a strategy employing genetically engineered Bacillus subtilis (strain Bs-BAMBI-8) to constitutively secrete bone morphogenetic protein and activin membrane-bound inhibitor (BAMBI), which is subsequently delivered to the liver via the gut-liver axis to therapeutically antagonize liver fibrosis. Methodologically, liver fibrosis was induced in mice via 6-week intraperitoneal injections of thioacetamide (TAA), followed by a 19-week daily oral gavage

Harnessing dental stem cell-derived synovial cells for IBD therapy: dual modulation of gut immunity and crypt repair.

BACKGROUND: Inflammatory bowel disease (IBD) remains a therapeutic challenge due to its chronic relapsing nature and limited long-term remission strategies. Here, we propose a therapy using stem cells from human exfoliated deciduous teeth (SHEDs). METHODS: We evaluated SHEDs in murine colitis models (DSS/TNBS) and human IBD tissues. Single-cell RNA sequencing (scRNA-seq) and organoid cocultures were used to characterise SHED differentiation and therapeutic mechanisms. FINDINGS: SHEDs migrate to inflamed intestines, differentiate into TGM2+TFPI2+ quiescent fibroblast-like synovial cells (QFLS

Two-dimensional Porcine Intestinal Organoids Reflecting the Physiological Properties of Native Gut.

The gastrointestinal tract (GIT) serves both in the digestion of food and the uptake of nutrients but also as a protective barrier against pathogens. Traditionally, research in this area has relied on animal experiments, but there's a growing demand for alternative methods that adhere to the 3R principles-replace, reduce, and refine. Porcine organoids have emerged as a promising tool, offering a more accurate in vitro replication of the in vivo conditions than traditional cell models. One major challenge with intestinal organoids is their inward-facing apical surface and outward-facing basolat

Molecular Genetics and Probiotic Mechanisms of Saccharomyces cerevisiae var. boulardii.

Saccharomyces cerevisiae var. boulardii (Sb) is a S. cerevisiae (Sc) strain that has been widely used in the treatment of gastrointestinal diseases due to its unique probiotic properties. The key genomic differences that distinguish Sb from Sc include the tetrasomy of chromosome XII, the absence of intact transposon-yeast (Ty) elements, and variations in the copy number of specific genes. These genomic variations may contribute to enhanced thermotolerance, increased acid resistance, and elevated acetate production, collectively supporting its probiotic functions. The probiotic mechanisms of Sb

Sequential activation of conventional and plasmacytoid dendritic cells in autoimmune pancreatitis and systemic lupus erythematosus: similarities and dissimilarities.

Type 1 autoimmune pancreatitis (AIP) and systemic lupus erythematosus (SLE) are caused by type I IFNs secreted by plasmacytoid dendritic cells (pDCs). Our understanding of the immune consequences before and after pDC activation in SLE is expanding, whereas knowledge on those in AIP are insufficient. In this article, we summarize the similarities and dissimilarities in pDC activation between AIP and SLE. In SLE, neutrophil extracellular traps containing self-DNA, anti-microbial peptides, and endogenous alarmins form anti-DNA antibody complexes, promoting type I IFN production by pDCs. Type I IF
