The clinical spectrum of anti-Ro-positive cutaneous neonatal lupus erythematosus.

BACKGROUND: Cutaneous neonatal lupus erythematosus (NLE) is an uncommon disease described mainly through isolated case reports. OBJECTIVE: Our purpose was to examine the cutaneous spectrum, clinical associations, and course of disease in babies with anti-Ro-positive NLE. METHODS: This is a retrospective case series evaluation of newborns with anti-Ro-positive NLE seen at a single ambulatory care university center over a 20-year period. Cases were drawn from a population of 3.2 million. Follow-up was at least 3 years. RESULTS: Four boys and 14 girls were included in our evaluation. Distribut

Biochemistry of copper.

Copper, as a component of numerous cuproenzymes, plays a vital role in many physiologic functions in man and animals. From the stand-point of human health there are at least three functional areas of prime importance. Copper is involved in the development and maintenance of cardiovascular and skeletal integrity, central nervous system structure and function, and erythropoietic function including iron metabolism. Although there is no evidence for widespread copper deficiency in the human population, it does occur, owing to genetic defects and other precipitating factors. A clear understanding o

In-vivo staging of pathology in REM sleep behaviour disorder: a multimodality imaging case-control study.

BACKGROUND: Accumulating evidence suggests that α-synuclein aggregates-a defining pathology of Parkinson's disease-display cell-to-cell transmission. α-synuclein aggregation is hypothesised to start in autonomic nerve terminals years before the appearance of motor symptoms, and subsequently spread via autonomic nerves to the spinal cord and brainstem. To assess this hypothesis, we investigated sympathetic, parasympathetic, noradrenergic, and dopaminergic innervation in patients with idiopathic rapid eye movement (REM) sleep behaviour disorder, a prodromal phenotype of Parkinson's disease. Strebluses A and B as tyrosinase inhibitors from the stems of Streblus ilicifolius (S. Vidal) Corn.

Tyrosinase inhibitors from natural sources are promising skin-whitening agents for treating pigment disorders. The phytochemical analysis of Streblus ilicifolius stems identified two new compounds, strebluses A (cp1) and B (cp2), with strong tyrosinase inhibitory activity. Their structures were determined using NMR, mass spectrometry, and absolute configurations by circular dichroism (CD) combined with ab initio calculations. In the tyrosinase inhibitory activity test, both compounds display more potent inhibitory activity (i.e., IC50 = 6.8 and 22.9 µM, respectively) than the positive control

Engineered tRNA reduces vision loss in a mouse model of Leber congenital amaurosis.

Premature termination codons (PTCs) are a major class of pathogenic variants that underlie rare inherited disorders, including forms of childhood blindness. Therapeutic suppression of these "nonsense mutations" offers a gene- and position-agnostic strategy to restore protein function. Our previous work established that the W53X PTC in the KCNJ13 gene causes Leber congenital amaurosis type 16 (LCA16) by disrupting the inwardly rectifying potassium channel Kir7.1, leading to retinal pigment epithelium (RPE) dysfunction. Here, we present a proof-of-concept approach using anticodon-engineered tran

Treatment of Refractory Centrofacial Melasma in a 64-Year-Old Female Using the NewMelan Depigmentation System: A Case Report.

Melasma is a chronic acquired hyperpigmentation disorder that commonly affects sun-exposed areas of the face and may significantly impact quality of life. Treatment can be challenging because of frequent recurrence and variable responses to conventional therapies. This case report describes the management of refractory facial melasma in a 64-year-old patient with a history of prolonged ultraviolet exposure and persistent facial hyperpigmentation. The patient previously underwent multiple treatment modalities, including chemical peels, mechanical peels, and laser-based therapies, without achiev

Prime editing for ocular gene therapy and disease modeling: a narrative review of advances, delivery, and translational readiness.

Prime editing is a versatile "search-and-replace" genome-editing technology that enables precise and flexible genome correction of genetic sequences by reverse-transcribing an RNA template encoded at the 3' end of a prime editing guide RNA (pegRNA). It supports the introduction of nucleotide substitutions, and insertions or/and deletions (indels) in living cells without requiring double-stranded DNA breaks or exogenous donor templates. Since its introduction in 2019, prime editing has advanced rapidly-from the first-generation prime editor (PE1) to PE7 and other next-generation variants-with e

Partial Unilateral Lentiginosis: A Systematic Review of 159 Reported Cases and Diagnostic Considerations.

Partial unilateral lentiginosis is a rare pigmentary disorder characterized by multiple lentigines confined to one side of the body and is frequently misdiagnosed as nevus spilus or segmental neurofibromatosis type 1. Despite its benign nature, its distinctive distribution, often involving visible areas such as the face and neck, may result in cosmetic concern and diagnostic uncertainty. A systematic review was conducted to synthesize all published cases of partial unilateral lentiginosis. PubMed and Embase were searched from 1983 to 2026 for case reports and case series describing clinically

Large-scale Whole-Exome Sequencing Defines the Protein-Coding Architecture of Retinal Structure, Visual Function, and Major Blinding Diseases.

The retina is an accessible extension of the central nervous system, yet the protein-coding architecture linking retinal structure, visual function, and major blinding diseases remains poorly defined. Here, using large-scale whole-exome sequencing data from 356,982 UK Biobank participants, exome-wide gene-based tests of rare coding variants and single-variant analyses of common coding variants are performed. A total of 22 significant rare-variant gene-based associations involving 16 genes are identified, including 12 novel genes, 10 of which are independently supported in the All of Us cohort

Prolonged survival in Menkes disease with a novel ATP7A variant: A case report.

BACKGROUND: Menkes disease (MD) is an uncommon, X-linked recessive neurodegenerative disorder caused by mutations in the copper-transporting ATP7A gene, leading to defective copper metabolism. Progressive neurological dysfunction, connective tissue abnormalities, characteristic skeletal and hair findings are defining features of the disease. The prognosis of the classical variant is poor since the majority of patients expire by the age of 3 years. This report presents the first documented case of MD in the Kingdom of Bahrain with a novel ATP7A variant. CASE SUMMARY: We report a 45-day old mal

Human retinal organoids: from developmental biology to translational applications.

Human retinal organoids (ROs), derived from pluripotent stem cells, have emerged as versatile human-relevant systems for studying retinal development, modeling disease, and evaluating therapeutic strategies. This review summarizes the evolution of human RO technology, from early stem-cell-based differentiation protocols to advanced culture platforms that improve scalability, reproducibility, and functional maturation. We compare human ROs with native human retinas regarding developmental trajectory, cellular composition, and photoreceptor function, while also highlighting persistent gaps in va

Bst2-targeted senotherapy restores visual function by eliminating senescent retinal cells.

Senescent cells contribute to degenerative processes in multiple tissues, including the retina. In the retinal pigment epithelium (RPE), their accumulation is closely associated with retinal aging and disease progression. Eliminating senescent RPE cells has shown therapeutic potential, but conventional senolytics often lack the specificity required to spare non-senescent cells, raising safety concerns. To overcome this, we performed integrated transcriptomic analyses of male mouse-derived RPE cells under natural aging and chemically induced senescence conditions. These analyses identified Bst2

Exploring the Effectiveness, Tolerability, and Safety of the Adjunctive Use of Microneedling With Tranexamic Acid in the Treatment of Melasma.

BACKGROUND: Melasma is a chronic pigment disorder associated with significant psychosocial burden. The gold standard triple combination cream is effective but limited by relapse and adverse effects. Tranexamic acid (TXA) and microneedling have independently shown promise in the treatment of melasma, and their combined use may provide enhanced efficacy, durability, and tolerability. AIMS: This review aims to synthesize and compare clinical evidence assessing the effectiveness, safety, patient satisfaction, and durability of microneedling-based TXA for the treatment of melasma. METHODS: A PubM

Hydroxycarboxylic acid receptor 2 (GPR109A) and retinopathies: pathways and prospects.

GPR109A, also known as the hydroxycarboxylic acid receptor 2 (HCAR2), is a G protein-coupled receptor with emerging significance in ocular health. Although considerable attention has focused on its role in the diabetic retina, growing evidence suggests that GPR109A may also play an important role in other retinal pathologies, including hypertensive retinopathy (HR) and retinopathy of prematurity (ROP), where inflammation, oxidative stress, and vascular instability similarly drive disease progression. Expressed in key retinal cell types, including retinal pigment epithelial cells, endothelial c
