[Treatment of deep chondral defects of the knee using autologous chondrocytes cultured on a support--results after one year].

PURPOSE OF THE STUDY: The treatment of deep chondral defects is one of the key issues in current orthopedics and traumatology. We developed a new technique that greatly improved the outcome of treatment in young active patients. The method is based on transplantation of in vitro cultured autologous chondrocytes carried on the fibrin glue Tissucol. MATERIAL: In the years 2000 to 2002, 19 patients with deep chondral defects were treated by chondrocyte transplantation with the use of Tissucol. At 1 year, 14 defects (average size, 4.31 cm2; range, 2.0 to 10.0 cm2) in 12 patients were evaluated. T

Embedded cell-only bioprinting to engineer structurally aligned meniscal fibrocartilage.

The engineering of functional meniscal grafts remains elusive, largely due to an inability to recapitulate the highly organized collagen architecture of the native tissue which is integral to its biomechanical function. In this study, we investigated whether external geometric confinement can direct collagen alignment in fibrocartilaginous tissues generated by mesenchymal stem/stromal cells (MSCs). First, MSCs were cast within non-adhesive agarose channels, which supported the development of fibrocartilaginous tissues with collagen fibres aligned parallel to the long axis of the confining agar

3D-Printable, Honeycomb-Inspired Tissue-Like Bioelectrodes for Patient-Specific Neural Interface.

The unique gyral patterns of the human brain demand patient-specific neural interfaces to achieve precise neuromodulation, mitigate adverse tissue responses, and optimize therapeutic efficacy and safety. One-size-fits-all, conventional rigid electrocorticography (ECoG) electrodes, standardized for mass production through lithographic techniques, exhibit limited conformability to the brain's heterogeneous cortical topography. This mechanical mismatch results in poor electrode-tissue contact, signal loss, and foreign body responses. To address these limitations, we present an integrated novel pl

Engineering functional vasculatures to reconstruct sympathetic-parasympathetic circuits for bladder function after spinal cord injury.

Spinal cord injury (SCI) leads to severe neurogenic bladder dysfunction, and its recovery highly depends on the restoration of bladder innervation of sympathetic-parasympathetic nerves. Stable blood perfusion is a prerequisite for sustaining neural regeneration, yet functional microvasculature rarely forms within injured spinal tissue. In this study, we engineered an integrating hydrogel-based spinal cord substitute to support vascularization through temporally staged angiogenic signaling combined with immune modulation. Early delivery of vascular endothelial growth factor (VEGF) promoted endo

Mathematical modeling of heat transfer in tissues with skin tumor during thermotherapy.

The study of thermal therapy to tumors and the response of living cells to this therapy used to treat tumor is very important due to the complexity of heat transfer in biological tissues. In the past few years, there has been a growing interest among clinicians, mathematicians, and engineers regarding the use of computational and mathematical methods to simulate biological systems. Numerous medical proceedings also employ mathematical modeling and engineering techniques as a means to guarantee their safety and evaluate the associated risks effectively. This manuscript provides an analytical so

Biomimetic in situ tracheal microvascularization for segmental tracheal reconstruction in one-step.

Formation of functional and perfusable vascular network is critical to ensure the long-term survival and functionality of the engineered tissue tracheae after transplantation. However, the greatest challenge in tracheal-replacement therapy is the promotion of tissue regeneration by rapid graft vascularization. Traditional prevascularization methods for tracheal grafts typically utilize omentum or muscle flap wrapping, which requires a second operation; vascularized segment tracheal orthotopic transplantation in one step remains difficult. This study proposes a method to construct a tissue-engi

Temporal characterization of hyaluronidases after peripheral nerve injury.

Hyaluronic acid (HA) is ubiquitously found in biological tissues and mediates wound healing mechanisms after injury by promoting cell migration and proliferation. With the development of tissue-engineered neural therapeutics, including off-the-shelf grafts for peripheral nerve repair, HA is an attractive material for clinical use because of its various biological roles. HA-based biomaterials have been carefully engineered to elicit specific in vivo host responses, however an important design feature that should be considered in these scaffolds is endogenous degradation. Hyaluronidases (HYALs)

Eye Socket Regeneration and Reconstruction.

Purpose: When an eye has become irreversibly blind or painful it is removed by enucleation or evisceration. The resulting anophthalmic socket usually receives a volume replacing implant and is subsequently fitted with a prosthetic shell for adequate cosmesis. Trauma, tumour or immunological pathomechanisms can induce loss of bone, orbital soft tissue volume, and conjunctival contraction or implant exposure, which result in difficult or impossible prosthesis wear. In this situation as well as in numerous diseases limited to the conjunctiva (e.g. Pterygium, or cicatrizing conjunctivitis) strateg

The Impact of γ-Irradiation and EtO Degassing on Tissue Remodeling of Collagen-based Hybrid Tubular Templates.

Clinical implementation of novel products for tissue engineering and regenerative medicine requires a validated sterilization method. In this study, we investigated the effect of γ-irradiation and EtO degassing on material characteristics in vitro and the effect on template remodeling of hybrid tubular constructs in a large animal model. Hybrid tubular templates were prepared from type I collagen and Vicryl polymers and sterilized by 25 kGray of γ-irradiation or EtO degassing. The in vitro characteristics were extensively studied, including tensile strength analysis and degradation studies. Fo…

Carboxymethyl chitosan microspheres structurally reinforce silk fibroin hydrogels for integrated aesthetic and functional soft tissue repair.

Achieving concurrent aesthetic volume contouring and physiological healing remains a major hurdle in soft tissue reconstruction. While conventional hydrogels hold potential for soft tissue reconstruction, their clinical application is limited by insufficient mechanical durability for aesthetic volume contouring and a lack of bioactive signals for physiological healing. We engineered a structurally reinforced hydrogel (SFCC) where zinc crosslinked carboxymethyl chitosan microspheres bind to a thermal induced silk fibroin network, driving the formation of a coarsened fibrillar architecture. Macroporous Poly-N-Isopropylacrylamide and Cationic Co-Polymer Hydrogels: Long-Term Subcutaneous Implant Biocompatibility.

Biomaterials have been developed with a great potential in tissue engineering. Hydrogels, especially those based on poly-N-isopropylacrylamide (PNIPAM), have innate mechanical and physicochemical similarities with the native extracellular matrix. We have previously reported the in Vitro compatibility of PNIPAM and its copolymers with a varied number of cell lines, displaying good proliferation rates and bioadhesive capacity. However, the in Vivo biocompatibility assessment is necessary to evaluate. In this work, biocompatibility and immunological acceptance of macroporous hydrogels based on PN…

Prepatterned Tissue Stiffness Gradient Controls Organ shape and size.

Organ morphogenesis is orchestrated by precise coupling of mechanical and biochemical signals, yet how these two signals are integrated within tissue niche to regulate organ shape and size remains poorly understood. Here, using sweat gland as a tractable model of exocrine mini-organs, we demonstrate that dermal fibroblasts play a key role to translate prepatterned tissue stiffness gradients into spatial Wnt5a gradients via Piezo1-mediated mechanosensing. Following epidermal placode formation, progenitor cells collectively invaginate into the dermis, where spatiotemporally graded Wnt5a directs

A Recombinant Elastic Peptide Rescues Elasticity From a Self-Assembled Dermal Sheet Model Treated With Ascorbic Acid.

The extracellular matrix (ECM) plays a pivotal role in determining the structure and function of the skin. Collagen and elastin, in particular, are responsible for providing tensile strength and elasticity, respectively. However, imbalances in the components of the extracellular matrix (ECM) during tissue engineering often result in the deterioration of the mechanical properties and physiological relevance of dermal substitutes, in part due to the detrimental effect of ascorbic acid (AA) on elastic fibre biosynthesis. The objective of this study is to investigate the potential of a synthetic e…

NIR-II photothermal-driven pulsatile PTH release for osteoporotic bone repair.

Integrating light-controlled intelligent materials into three-dimensional (3D) printing technology holds great promise for bone tissue engineering, enabling spatiotemporal control over in situ drug dosing. However, this integration faces a critical molecular design challenge, as incorporated photoresponsive molecules must simultaneously fulfill the dual demands of high ultraviolet (UV)-stability during UV-mediated photopolymerization, and strong long-wavelength absorption for deep-tissue activation. Here, we report a wavelength-selective quinolinium dye (QC7CN) as photoresponsive 'dye ink' for…

Semi-synthetic fibrous fibrin composites promote 3D microvascular assembly, survival, and host integration of endothelial cells without mesenchymal cell support.

Vasculogenic assembly of 3D capillary networks remains a promising approach to vascularizing tissue-engineered grafts, a significant outstanding challenge in tissue engineering and regenerative medicine. Current approaches for vasculogenic assembly rely on the inclusion of supporting mesenchymal cells alongside endothelial cells, co-encapsulated within vasculo-conducive materials such as low-density fibrin hydrogels. Here, we established a material-based approach to circumvent the need for supporting mesenchymal cells and report that the inclusion of synthetic matrix fibers in dense (>3 mg mL-…
