The regulatory role of cerebrospinal fluid (CSF) in brain physiology is well established, while our understanding of its role in brain immunity is undefined. We demonstrate that normal rat CSF suppresses the in vitro development of mastocytoma-specific CTL activity in restimulated splenocytes from Balb/c mice, a strain unable to reject this tumor from the brain. Suppression is dependent on TGF-beta, revealed by reversal of suppression with specific neutralizing antibody. In contrast, mice which can reject this tumor from the brain, such as Balb/c mice with immunological memory to the tumor or

Plant cell suspension cultures are a valuable biotechnological platform for the controlled production of high-value phytochemicals. Yet, the molecular mechanisms by which culture medium composition impacts cellular transcriptional programs remain poorly characterised, especially in non-model plant species. Here, we investigated how two culture media, Murashige & Skoog (MS)- and Linsmaier & Skoog (LS)-based, influence growth, gene expression and secondary metabolite production in Cannabis sativa cell suspension cultures derived from a non-commercial genotype, Line 43/178. Cultures were analysed

BACKGROUND: Maternal diabetes and exposure to a high‑fat diet (HFD) disrupt pancreatic development and glucose homeostasis, potentially inducing transgenerational metabolic changes.
OBJECTIVE: Investigate how maternal diabetes and HFD exposure impact the endocrine pancreas across generations. Elucidate their effects on oxidative stress, cellular senescence, proliferation, cell death, and islet architecture in a transgenerational manner.
METHODS: Female Sprague‑Dawley rats (F1) were allocated into four groups (n=10/group): i) Offspring from non‑diabetic dams fed a standard diet (OC/SD); ii) O…

Glycosylation plays a significant part in cancer pathophysiology, shaping tumour progression, metastasis, immune evasion, and drug resistance. Aberrant glycosylation patterns in tumours serve as impending biomarkers and therapeutic targets for enriched cancer diagnostics and treatment strategies. Developments in glycosylation-mediated drug delivery systems (DDSs) have enabled more targeted targeting of tumour cells through receptor-specific interactions, boosting drug efficacy while mitigating systemic toxicity. Glycosylated nanocarriers, antibodies, and fusion proteins have proven favourable

INTRODUCTION: The "longevity gene" sirtuin 1 (SIRT1) is a critical regulator of oxidative stress and cellular senescence, making it a high-value target for anti-aging dermatological interventions. However, the stratum corneum poses a formidable barrier to the transdermal delivery of therapeutic genes. This study systematically evaluated the efficacy of non-viral SIRT1 plasmid delivery to a full-thickness 3D human skin equivalent (T-Skin™) using distinct vector formulations and physical enhancement methods.
MATERIALS AND METHODS: We compared the delivery efficiency of naked plasmid DNA and a c…

The association between higher levels of physical activity and lower cancer risk and mortality is well established. However, a causal link is yet to be proven. Recent studies showed a decrease in the proliferation rates of cultured human cancer cells when the human serum used to stimulate them was conditioned by acute exercise. Here, we tested the hypothesis that serum mediates some of the putative benefits of exercise on cancer through alterations to the growth pattern and susceptibility to chemotherapy agents of cancer cells. To this end, human non-small cell lung cancer cells were exposed t

Mitochondria are morphologically and functionally heterogeneous and dynamically adapt to the current metabolic status of their hosting cell. Moreover, they are prominent sources but also sensitive targets of redox modulation and oxidative stress. Such subcellular ROS/redox signals are considered pivotal aspects in health and disease. Yet, their deciphering requires advanced optical tools. Here we took advantage of transgenic redox-indicator mice expressing a mitochondria-targeted reduction/oxidation-sensitive green fluorescent protein (roGFPm) in excitatory projection neurons. The cGAS-STING pathway is an essential cytosolic DNA sensing mechanism that activates innate immune responses upon detection of microbial or aberrant self-DNA. This evolutionarily conserved signaling axis plays critical roles in autoimmune diseases, sterile inflammation, and cellular senescence. While its transient activation provides protective immunity, dysregulated cGAS-STING signaling contributes to the pathogenesis of various inflammatory and autoimmune conditions. Growing evidence indicates its functional convergence with multiple cell death pathways-particularly PANoptosis, a distinct i…

PURPOSE: Vision-threatening ocular diseases are impacted by aging-associated molecular changes, including mitochondrial dysfunction, cellular senescence, and chronic inflammation. Anti-VEGF therapies targeting VEGF-A/VEGFR2 signaling remain the frontline standard of care, but many patients exhibit suboptimal or nondurable responses, often due to compensatory and/or compromised antiangiogenic and anti-inflammatory pathways. We aimed to elucidate shared mechanisms underlying treatment failure and disease progression.
METHODS: We applied an integrative systems biology framework that combined mul…

BACKGROUND: Despite advances in antihypertensive therapies, uncontrolled hypertension remains a major global health challenge, particularly among older adults. Ferroptosis, an iron-dependent programmed cell death, has been implicated in age-related vascular dysfunction, but its regulatory mechanisms in hypertension are unclear. This study investigates Sestrin2, a highly conserved stress-induced protein linked to cellular senescence, in regulating ferroptosis in hypertension.

Plant-derived exosome-like nanovesicles (PDEVs) are emerging as breakthrough platforms for the treatment of age-related diseases (ARDs). These endogenous nanocarriers contain a variety of bioactive molecules, including microRNAs, proteins, lipids, and phytochemicals, which play crucial roles in therapy. PDEVs have strong potential to treat chronic inflammation, oxidative stress, cellular senescence, and mitochondrial dysfunction, all of which are related to aging. Their pleiotropic effects support wide therapeutic applications in neurodegenerative, cardiovascular, and metabolic diseases; sarco…

The muscle-tendon junction (MTJ) is a specialised interface between muscle and tendon and transmits muscle-generated force to the tendon. The MTJ is particularly vulnerable to injuries compared to muscle and tendon and becomes more injury prone with age. Despite its clinical importance, the mechanisms driving MTJ ageing and age-related functional deterioration remain poorly understood. In this study, young (3-month-old) and old (23-month-old) male mice were used to provide the first comprehensive three-dimensional characterisation of age-related structural and cellular changes at the mouse Ach

BACKGROUND: Antifibrotic therapy slows fibrotic interstitial lung disease (ILD) progression. However, fibrotic ILD remains a major life-threatening condition, with associated morbidity and mortality increasing annually. Growth differentiation factor 15 (GDF15), a stress-responsive cytokine within the transforming growth factor beta superfamily, has garnered increasing attention regarding its association with cachexia and immunomodulation. It is also involved in cellular senescence and ageing, all of which have been implicated in the progression of lung fibrosis, METHODS: We measured serum GDF1…

Alzheimer's disease (AD) is a multifactorial neurodegenerative disorder driven by complex interactions between protein aggregation, oxidative stress, neuroinflammation, and cellular dysfunction. Among plant-derived compounds, curcumin has emerged as one of the most extensively studied polyphenols due to its broad spectrum of biological activities. This review provides a critical synthesis of the mechanistic, preclinical, and clinical evidence on curcumin in AD. Experimental studies consistently demonstrate that curcumin modulates key pathogenic processes, including neuroinflammatory signaling,
