Hybrid fiber alterations in exercising seniors suggest contribution to fast-to-slow muscle fiber shift.

BACKGROUND: Human skeletal muscle is composed of a functional and metabolic continuum of slow (Type I) and fast fibers (IIa and IIx). Hybrid fibers co-expressing different myosin heavy chains are also present and seem to be more prominent in aging muscle. Their role is debated; hybrid fibers were reported either in a transitional state, between slow and fast fibers, or as fixed individual entities. This study examined the fate of hybrid fibers with an endurance exercise intervention in an elderly sedentary population. METHODS: Twenty-two sedentary healthy elderly men and women underwent a 16-

Structure, dynamics and thermodynamics of the human centrin 2/hSfi1 complex.

Centrin, an EF-hand calcium-binding protein, has been shown to be involved in the duplication of centrosomes, and Sfi1 (Suppressor of fermentation-induced loss of stress resistance protein 1) is one of its centrosomal targets. There are three isoforms of human centrin, but here we only considered centrin 2 (HsCen2). This protein has the ability to bind to any of the approximately 25 repeats of human Sfi1 (hSfi1) with more or less affinity. In this study, we mainly focused on the 17th repeat (R17-hSfi1-20), which presents the highest level of similarity with a well-studied 17-residue peptide (P

Modeling the structure of agitoxin in complex with the Shaker K+ channel: a computational approach based on experimental distance restraints extracted from thermodynamic mutant cycles.

Computational methods are used to determine the three-dimensional structure of the Agitoxin (AgTx2)-Shaker complex. In a first stage, a large number of models of the complex are generated using high temperature molecular dynamics, accounting for side chain flexibility with distance restraints deduced from thermodynamic analysis of double mutant cycles. Four plausible binding mode candidates are found using this procedure. In a second stage, the quality and validity of the resulting complexes is assessed by examining the stability of the binding modes during molecular dynamics simulations with

Dynamic monitoring of immunoglobulin G as a prognostic indicator after curative resection in high-risk stage II-III colorectal cancer: a retrospective cohort study.

BACKGROUND: Reliable markers for predicting postoperative recurrence in high-risk stage II-III colorectal cancer remain limited. Dynamic changes in immunoglobulin G (IgG) may provide prognostic information beyond carcinoembryonic antigen (CEA). METHODS: This single-centre retrospective cohort study included 192 patients with high-risk stage II or III colorectal cancer who underwent curative resection between January 2021 and June 2023. The study evaluated whether dynamic postoperative monitoring of serum IgG predicts 2-year disease-free survival (DFS) compared with CEA. Serial IgG and CEA mea

A Novel Eigen-Volume-based Co-Activation Pattern Framework for Dynamic Functional Biomarkers of Multiple Sclerosis.

Imaging biomarkers are essential for monitoring multiple sclerosis (MS), wand resting-state functional MRI (rs-fMRI) offers functional insights that complement structural imaging. This study investigates whether a novel co-activation pattern (CAP) approach for dynamic rs-fMRI can function as a dual-purpose biomarker in MS, aiding diagnosis and tracking disease severity. RS-fMRI scans from 25 relapsing-remitting MS patients and 41 healthy controls (HCs) were analyzed using a novel CAP-based approach. CAPs derived from individual time frames to capture dynamic brain activity patterns incorporate

The prognostic impact of perioperative dynamic changes in cachexia index in patients with hepatocellular carcinoma.

BACKGROUND: The cachexia index (CXI), which consists of skeletal muscle, inflammation, and nutritional status, has been associated with prognosis in patients with hepatocellular carcinoma (HCC). We hypothesized that dynamic changes in CXI might be associated with long-term outcomes in HCC. METHODS: This study comprised 131 patients who had undergone primary hepatic resection for HCC between 2008 and 2019. Preoperative CXI (pre-CXI) and postoperative CXI (post-CXI) were calculated by the following formula: skeletal muscle index x serum albumin level / neutrophil-to-lymphocyte ratio. Pre- and p

Cross-frequency phase-amplitude coupling in repetitive movements in patients with Parkinson's disease.

Bradykinesia is a cardinal motor symptom in Parkinson's disease (PD), the pathophysiology of which is not fully understood. We analyzed the role of cross-frequency coupling of oscillatory cortical activity in motor impairment in patients with PD and healthy controls. High-density EEG signals were recorded during various motor activities and at rest. Patients performed a repetitive finger-pressing task normally, but were slower than controls during tapping. Phase-amplitude coupling (PAC) between β (13-30 Hz) and broadband γ (50-150 Hz) was computed from individual EEG source signals in the prem

Cerebrovascular reactivity and dynamic autoregulation in tetraplegia.

Humans with spinal cord injury have impaired cardiovascular function proportional to the level and completeness of the lesion. The effect on cerebrovascular function is unclear, especially for high-level lesions. The purpose of this study was to evaluate the integrity of dynamic cerebral autoregulation (CA) and the cerebrovascular reactivity in chronic tetraplegia (Tetra). After baseline, steady-state hypercapnia (5% CO(2)) and hypocapnia (controlled hyperventilation) were used to assess cerebrovascular reactivity in 6 men with Tetra (C5-C7 lesion) and 14 men without [able-bodied (AB)]. Molecular Structure Design of Dynamically Bonded Recyclable Commercial Polyurethane Foams for Enhanced Reprocessing Performance and Upcycling.

Although commercial polyurethane foam (PUF) possessed significant recycling value, its permanently cross-linked structure made reprocessing and recycling highly challenging. This study utilized biosourced vanillin (VAN) and 4,4'-dithiodianiline (4-DTDA) to design a dual dynamic bond cross-linker (APDV) containing both S-S bonds and C═N bonds for the recycling and reprocessing of commercial PUF. Under hot-pressing conditions, the hydroxyl groups in APDV underwent exchange reactions with the urethane bonds in the polyurethane chains, enabling the one-step conversion of commercial PUF into high-p

Influence of the Hardness of Insoles on the Abductor Hallucis Muscle in Baxter's Entrapment: A Cross-Over Randomized Study.

Background: The etiology and diagnosis of heel pain are complex and multifactorial, and it has been reported that Baxter's entrapment is responsible for up to 20% of foot disorders. The most conservative treatment of Baxter's entrapment has been the use of custom insoles. Electromyography was considered an effective test to assess muscle activity. The aim of this study was to test the use of insoles of different hardness on muscle activity of the abductor hallucis muscle in subjects with entrapment of the lateral branch of the external plantar nerve. Methods: 18 subjects (7 women and 11 men) d

Dynamic Scheduling for Data-Parallel Path Tracing of Large-Scale Instanced Scenes.

Data-parallel ray tracing is a crucial technique for rendering large-scale scenes that exceed the memory capacity of a single compute node. It partitions scene data across multiple nodes and accesses remote data through inter-node communication. However, the resulting communication overhead remains a significant bottleneck for practical performance. Existing approaches mitigate this bottleneck by enhancing data locality through dynamic scheduling during rendering, typically employing spatial partitioning to enable access prediction. Although effective in some scenarios, these methods incur sig

Mimetic violence and wartime organ donation between Israeli Jews and Palestinians.

This paper develops a theoretical framework for understanding how wartime organ donation can both interrupt and reinforce cycles of mimetic violence. Drawing on René Girard's mimetic theory, we analyze ethnographic and interview-based material collected between August 2021 and October 2022 from three cases of cross-community organ donation between Israeli Jews and Palestinians during armed conflict, examining how donor families and recipients frame bodily giving as an ethical, relational, and symbolic practice. The findings reveal a dual dynamic. Organ donation functions simultaneously as a bo

Molecular pathogenesis, global epidemiological trends, and treatment strategies for pteropine orthoreoviruses: a narrative review.

Pteropine orthoreoviruses are emerging bat-borne zoonotic viruses of the genus Orthoreovirus (family Reoviridae), increasingly recognized as causes of acute respiratory disease in humans. Originally grouped with the largely non-pathogenic mammalian orthoreoviruses, they have challenged that view through their association with severe influenza-like illness, evidence of human-to-human transmission, and a broad geographic range across the Old World. Maintained primarily in fruit bats of the family Pteropodidae, they are now linked to neurological as well as respiratory disease. This narrative rev

Systematic analysis of the potential prebiotic effects of Blackberry Seed Polysaccharides by utilizing the in vitro spatio-temporal dynamic digestion models.

Blackberry seed polysaccharides (BSPs) are commonly considered to be a beneficial living substance. The digestive mechanism and functional properties of BSPs in the human gastrointestinal tract were investigated by simulating chronological digestion and dynamic fermentation in vitro. SEM and FT-IR analyses demonstrated that the dynamic digestion of BSPs resulted in solely morphological changes, with no effect on structural characteristics. The results of the alpha diversity and beta diversity analyses suggested that BSPs had the capacity to enhance the diversity of intestinal microorganisms to

Investigating the Design of ssPalmO-Derived Lipid Nanoparticles for mRNA Delivery Applications Using Molecular Dynamics Simulations.

The rational design of lipid nanoparticles (LNPs) is essential for the effective transport of drugs and genetic material, as their structural and dynamic properties are heavily influenced by lipid composition and functional group modifications. In this study, we employed molecular dynamics simulations with density functional theory (DFT) derived force fields to investigate the bilayer properties of ssPalmO lipids, their phenyl ester (ssPalmO-phe) and benzyl ester (ssPalmO-ben) derivatives, as well as their cis and trans isomers. While all systems formed stable bilayers, cis-ssPalmO deviated by
