A genomics-based framework for identifying biomarkers of human neurodevelopmental toxicity.

Human embryonic stem cell (hESC) neural differentiation models have tremendous potential for evaluating environmental compounds in terms of their ability to induce neurodevelopmental toxicity. Genomic based-approaches are being applied to identify changes underlying normal human development (in vitro and in vivo) and the effects of environmental exposures. Here, we investigated whether mechanisms that are shared between hESC neural differentiation model systems and human embryos are candidate biomarkers of developmental toxicities for neurogenesis. We conducted a meta-analysis of transcriptomi

The taming of the neural crest: a developmental perspective on the origins of morphological covariation in domesticated mammals.

Studies on domestication are blooming, but the developmental bases for the generation of domestication traits and breed diversity remain largely unexplored. Some phenotypic patterns of human neurocristopathies are suggestive of those reported for domesticated mammals and disrupting neural crest developmental programmes have been argued to be the source of traits deemed the 'domestication syndrome'. These character changes span multiple organ systems and morphological structures. But an in-depth examination within the phylogenetic framework of mammals including domesticated forms reveals that t

Neural control of voluntary eye closure: a case study and an fMRI investigation of blinking and winking.

The current paper describes a rare case of a patient who suffered from unilateral apraxia of eye closure as a result of a bilateral stroke. Interestingly, the patient's ability to voluntarily close both eyelids (i.e. blinking) was not affected, indicating that different neural mechanisms control each type of eye closure. The stroke caused damage to a large part of the right frontal cortex, including the motor cortex, pre-motor cortex and the frontal eye field (FEF). The lesion in the left hemisphere was restricted to the FEF. In order to further study the neural mechanisms of eye closure, we c

Evaluation of a neural network-based control strategy for a cost-effective externally-powered prosthesis.

This paper presents a control strategy that compensates for the nonlinearity in the inexpensive sensors and hardware of a cost effective prosthetic hand. The control strategy uses neural network-based force control and sensory feedback to detect disturbance induced by slippage. The neural network approach is chosen over other nonlinear models because it is easy to implement and it offered the additional advantage of having its parameters easily adjusted over the life span of the device. The proposed strategy was evaluated on a functional multi-digit underactuated prosthetic hand. The initial a

Monaural deprivation disrupts development of binaural selectivity in auditory midbrain and cortex.

Degraded sensory experience during critical periods of development can have adverse effects on brain function. In the auditory system, conductive hearing loss associated with childhood ear infections can produce long-lasting deficits in auditory perceptual acuity, much like amblyopia in the visual system. Here we explore the neural mechanisms that may underlie "amblyaudio" by inducing reversible monaural deprivation (MD) in infant, juvenile, and adult rats. MD distorted tonotopic maps, weakened the deprived ear's representation, strengthened the open ear's representation, and disrupted binaura

Neural tube defects and associated anomalies in South Carolina.

BACKGROUND: Neural tube defects (NTDs) occur as isolated malformations and in the company of other birth defects. This study was conducted to determine the frequency of coexisting anomalies and the relationship between them. METHODS: Since 1992, NTDs have been identified through prenatal and postnatal surveillance activities in South Carolina. The type of NTD and presence of associated anomalies were determined by prenatal ultrasound, postnatal and/or postmortem examination. RESULTS: During the ten-year period from 1992 to 2002, 564 NTDs were identified by the surveillance system. Seventeen

Diagnostic accuracy of ultrasound in carpal tunnel syndrome: analysis of established and novel parameters.

OBJECTIVES: This study evaluated the diagnostic accuracy of 3 ultrasound-based parameters-median nerve cross-sectional area (CSA), elasticity (E value), and blood flow pixel ratio (BFPR)-in grading carpal tunnel syndrome (CTS) severity. These parameters were assessed using high-frequency ultrasound, shear wave elastography (SWE), and superb microvascular imaging (SMI), with nerve conduction studies (NCS) as the reference standard. METHODS: A prospective study conducted between April 2024 and January 2025 included 128 patients with suspected CTS and 25 healthy controls, totalling 223 median ne

Therapeutic comparison of MitoTEMPO and rosmarinic acid as mitochondria targeted and non targeted antioxidants evaluated in rat sciatic nerve crush injury model.

Mitochondria are key organelles involved in various pathophysiological processes. As the primary site of reactive oxygen species (ROS) production, they represent a major target for many therapeutic agents. In this study, the therapeutic effects of a mitochondria-targeted antioxidant (MitoTEMPO, MT) and a conventional yet potent antioxidant compound (rosmarinic acid, RA) were compared in a rat model of sciatic nerve crush injury, focusing on nerve conduction parameters and histological structures. A total of 28 adult Wistar Albino rats were used in the study, with seven animals assigned to each

High serum levels of CCL11/Eotaxin-1 are associated with diabetic sensorimotor polyneuropathy and peripheral nerve function but not with cardiac autonomic neuropathy in people with type 2 diabetes.

AIMS: To investigate whether higher serum CCL11/Eotaxin-1, a biomarker for aging and neurodegenerative and neuroinflammatory disorders, is associated with diabetic sensorimotor polyneuropathy (DSPN), peripheral nerve dysfunction, and cardiac autonomic neuropathy in people with type 2 diabetes. METHODS: This cross-sectional study included 106 patients with type 2 diabetes and 40 healthy controls, matched for the age and sex distribution of the diabetes group as a whole. The CC chemokines CCL11/Eotaxin-1 and CCL22/MDC were measured in fasting serum samples. DSPN and peripheral nerve function we

Pirfenidone mitigates demyelination and electrophysiological alterations in multiple sclerosis: Targeting NF-κB, sirt1, and neurotrophic genes.

Multiple sclerosis (MS) is a demyelinating disease affecting the central nervous system associated with progressive neurodegeneration. Pirfenidone (Pir) is a well-known antifibrotic agent; however, Pir's function in MS is little explored. We evaluated the neuroprotective effects of Pir in MS and its possible underlying mechanisms. Forty male Swiss mice were divided equally into control, cuprizone (CPZ), Pir, and CPZ + Pir groups. Assessment of motor function was conducted using neurobehavioral tests, EMG, and nerve conduction velocity (NCV). Mice's brains were extracted to measure oxidative st

Peripheral nerve adaptations to 10 days of horizontal bed rest in healthy young adult males.

Space analogs, such as bed rest, are used to reproduce microgravity-induced morphological and physiological changes and can be used as clinical models of prolonged inactivity. Nevertheless, nonuniform decreases in muscle mass and function have been frequently reported, and peripheral nerve adaptations have been poorly studied, although some of these mechanisms may be explained. Ten young healthy males (18-33 yr) underwent 10 days of horizontal bed rest. Peripheral neurophysiological assessments were performed bilaterally for the dominant (DL) and nondominant upper and lower limbs (N-DL) on the

Guillain-Barré syndrome during SARS-CoV-2 pandemic: A case report and review of recent literature.

Acute demyelinating inflammatory polyneuropathy (AIDP) is the most common type of Guillain-Barré syndrome (GBS) in Europe, following several viral and bacterial infections. Data on AIDP-patients associated with SARS-CoV-2 (coronavirus-2) infection are scarce. We describe the case of a 54-years-old Caucasian female patient with typical clinical and electrophysiological manifestations of AIDP, who was reported positive with PCR for SARS-CoV-2, 3 weeks prior to onset of the neurological symptoms. She did not experience a preceding fever or respiratory symptoms, but a transient loss of smell and t

Motor chronic inflammatory demyelinating polyneuropathy (CIDP) in 17 patients: Clinical characteristics, electrophysiological study, and response to treatment.

Motor chronic inflammatory demyelinating polyneuropathy (CIDP) is a rare and poorly described subtype of CIDP. We aimed to study their clinical and electrophysiological characteristics and response to treatment. From a prospective database of CIDP patients, we included patients with definite or probable CIDP with motor signs and without sensory signs/symptoms at diagnosis. Patients were considered to have pure motor CIDP (PM-CIDP) if sensory conductions were normal or to have motor predominant CIDP (MPred-CIDP) if ≥2 sensory nerve action potential amplitudes were abnormal. Among the 700 patien

Development of In-Office Laryngeal Nerve Conduction Studies: Computed Tomography and Cadaveric Study.

OBJECTIVES/HYPOTHESIS: In-office recurrent laryngeal nerve conduction studies (NCSs) are a technique that can potentially provide information about laryngeal innervation. NCS is essential in the management of other neuropathies including carpal tunnel syndrome and spinal cord injury. We hypothesize that laryngeal NCS may have similar utility in managing patients with vocal fold paralysis, atrophy, and neurodegenerative disease. NCSs are technically challenging because they require transcervical stimulation of the recurrent laryngeal nerve (RLN). This study combines radiographic data with cadav

Corticocortical Systems Underlying High-Order Motor Control.

Cortical networks are characterized by the origin, destination, and reciprocity of their connections, as well as by the diameter, conduction velocity, and synaptic efficacy of their axons. The network formed by parietal and frontal areas lies at the core of cognitive-motor control because the outflow of parietofrontal signaling is conveyed to the subcortical centers and spinal cord through different parallel pathways, whose orchestration determines, not only when and how movements will be generated, but also the nature of forthcoming actions. Despite intensive studies over the last 50 years, t

Compound Muscle Action Potential Amplitude Predicts the Severity of Cubital Tunnel Syndrome.

BACKGROUND: Cubital tunnel syndrome has a spectrum of presentations ranging from mild paresthesias to debilitating numbness and intrinsic atrophy. Commonly, the classification of severity relies on clinical symptoms and slowing of conduction velocity across the elbow. However, changes in compound muscle action potential (CMAP) amplitude more accurately reflect axonal loss. We hypothesized that CMAP amplitude would better predict functional impairment than conduction velocity alone. METHODS: A retrospective cohort of patients who underwent a surgical procedure for cubital tunnel syndrome over

Cortico-anorectal, Spino-anorectal, and Cortico-spinal Nerve Conduction and Locus of Neuronal Injury in Patients With Fecal Incontinence.

BACKGROUND & AIMS: The neuropathophysiology of fecal incontinence (FI) is incompletely understood. We examined the efferent brain-anorectal and spino-anorectal motor-evoked potentials (MEP) to characterize the locus of neuronal injury in patients with FI. METHODS: We performed bilateral transcranial, translumbar, and transsacral magnetic stimulations in 27 patients with FI (19 female) and 31 healthy individuals (controls, 20 female) from 2015 through 2017. MEPs were recorded simultaneously from the rectum and anus using 4 ring electrodes. The difference in MEP latencies between the transcrani

A cross-sectional study investigating frequency and features of definitely diagnosed diabetic painful polyneuropathy.

This cross-sectional multicentre study aimed at investigating frequency and features of painful diabetic polyneuropathy. We consecutively enrolled 816 patients attending hospital diabetic outpatient clinics. We first definitely diagnosed diabetic polyneuropathy and pure small-fibre polyneuropathy using clinical examination, nerve conduction study, and skin biopsy or quantitative sensory testing. Adhering to widely agreed criteria, we then identified neuropathic pain and diagnosed painful polyneuropathy using a combined approach of clinical examination and diagnostic tests. Of the 816 patients,
