Unfolded protein response in bronchopulmonary dysplasia: mechanisms, pathways, and therapeutic implications.

Bronchopulmonary dysplasia (BPD) is a prevalent chronic lung disease in preterm infants, characterized by impaired alveolarization and abnormal vascular development. Growing evidence indicates that endoplasmic reticulum stress (ERS) and the unfolded protein response (UPR) are closely involved in the pathogenesis of BPD. Under hyperoxic conditions, excessive reactive oxygen species promote protein misfolding and accumulation within the endoplasmic reticulum, leading to activation of the three major UPR branches-IRE1, PERK, and ATF6. Dysregulation of these pathways contributes to lung injury and

[Research progress on mechanism of diabetes peripheral neuropathy based on macrophage polarization and prevention and treatment of traditional Chinese medicine].

Diabetic peripheral neuropathy(DPN) is one of the most common chronic complications of diabetes, characterized by a complex pathogenesis and limited clinical treatment options. In recent years, increasing attention has been drawn to the role of macrophage polarization in DPN development and progress. This paper systematically reviewed the mechanisms of M1/M2 macrophage polarization in DPN, as well as research advances in traditional Chinese medicine(TCM) for DPN prevention and treatment by macrophage polarization regulation. Under normal conditions, M1/M2 macrophages are in a dynamic balance,

Characterization of polypharmacy and potential drug-drug interactions in Mexican older adults with cognitive decline.

BACKGROUND: Mild cognitive impairment (MCI) and dementia are common in older adults with multiple chronic conditions. Polypharmacy is more frequent in these populations and may increase the risk of potential drug-drug interactions (pDDIs). Most evidence on polypharmacy-related adverse outcomes originates from high-income countries, whereas data from Mexico remain limited. We aimed to determine the frequency of polypharmacy and pDDIs in older adults with and without cognitive impairment. METHODS: A case-control study was conducted in 141 participants from Mexico, 47 cases, and 94 controls. Eac…

Evaluation of handgrip strength and prostate cancer risk using propensity score matching in 64,371 European men.

BACKGROUND: The predictive value of low handgrip strength (HGS) for prostate cancer remains unclear. This study assessed whether low HGS is associated with increased prostate cancer risk among older European men. METHODS: Data were drawn from a longitudinal cohort of 64,371 men aged 50 and above at baseline, recruited in wave 1 (2004) of the Survey of Health, Ageing, and Retirement in Europe (SHARE), and followed prospectively across multiple waves from 2004 to 2022. Prostate cancer was identified via self-reported physician diagnoses. HGS was measured using a Smedley dynamometer. We applied

Timing of decompressive craniectomy and short-term outcomes in pediatric severe traumatic brain injury: a nationwide observational study in Germany.

Decompressive craniectomy (DC) is a critical intervention for managing severe traumatic brain injury (sTBI) in children when medical therapy fails, but the optimal timing remains unclear. This study evaluated the association between different DC timing and short-term outcomes in pediatric sTBI in Germany. A retrospective cohort study of the German national hospital discharge database was conducted for cases < 18 years undergoing DC following sTBI were extracted from 2016 to 2022. Time from admission to DC were calculated as complete hours and data were compared between early (time to DC ≤ 2 h)

Successful treatment of carbapenem-resistant Acinetobacter baumannii and Enterobacterales coinfection with sulbactam-durlobactam combined with aztreonam: a case report.

BACKGROUND: Carbapenem-resistant Gram-negative bacilli (CRGNB) often cause nosocomial infections difficult to treat. Their widespread dissemination, associated high mortality, and increasing disease burden constitute a significant global public health challenge. This situation is further exacerbated by mixed CRGNB infections, which exhibit elevated mortality rates and increased therapeutic complexity. Thus, it is imperative to establish effective antibiotic strategies with minimal adverse effects to address these polymicrobial infections. We present this case to evaluate the efficacy of sulbac

A Model of Piroxicam-Accelerated Enterocolitis in Interleukin-10 Knockout Mice for Studying Inflammation-Induced Metabolic Alterations.

Inflammatory bowel disease (IBD) is a chronic inflammatory disorder of the digestive tract affecting over 10 million individuals globally. While substantial research has focused on the immunologic mechanisms and consequences underlying IBD, less is understood about how mucosal inflammation contributes to metabolic dysregulation, including weight loss and reduced appetite. Here, the authors describe a mouse model of piroxicam-accelerated enterocolitis in interleukin-10-knockout (IL-10-KO) mice to study inflammation-induced metabolic dysregulation. IL-10-KO mice are known to develop spontaneous

TRPV4-Mast Cell Interactions in Neurogenic Inflammation and Chronic Diseases: A Narrative Review.

Transient receptor potential vanilloid 4 (TRPV4) is a polymodal cation channel that is widely expressed in sensory neurons, immune cells, and structural tissues, where it integrates mechanical, osmotic, and chemical stimuli to regulate both physiological responses and disease-associated signaling. Mast cells (MCs), key immune effector cells capable of rapid mediator release through degranulation, also express TRPV4. Increasing evidence supports TRPV4-MC signaling as an important neuroimmune interface, linking mechanical and inflammatory stimuli to tissue hypersensitivity and pain. In this revi

Impact of pediatric gastrointestinal disorders on learning and cognitive development in schoolchildren.

Schoolchildren nowadays encounter multiple challenges within the teaching process. Learning disabilities (LD) are characterized as complex conditions that affect academic achievement and cognitive performance. LD are typically linked to neurodevelopmental and genetic factors. However, emerging evidence suggests they may be associated with gastrointestinal (GI) pathologies, including celiac disease, inflammatory bowel disease, irritable bowel syndrome, gastroesophageal reflux disease and Helicobacter pylori infection. The aforementioned GI disorders have been (indirectly) linked with a spectrum

An intricate relationship between fibrosis and autoimmune diseases: a specific focus on unraveling its molecular, immunological, and epigenetic drivers.

Fibrosis is a pathological process of wound healing, characterized by excessive deposition of extracellular matrix (ECM) and chronic activation of fibroblasts, leading to organ scarring and a decline in function. Fibrogenesis is predominantly initiated by tissue injury and sustained inflammation, further driven by the complex interplay of growth factors, cytokines, metabolic alterations, and epigenetic reprogramming. Activated myofibroblasts and immune cells function as primary profibrotic mediators. The central molecular pathways implicated include TGF-β/SMAD signaling, non-canonical cascades

CA1 parvalbumin interneurons display alterations of myelin patterning driven by microgliosis in temporal lobe epilepsy.

Temporal lobe epilepsy (TLE) is one of the most common forms of epilepsy, characterized by significant reorganization of hippocampal neuronal circuits. While changes in GABAergic inhibitory circuits are a major feature of this reorganization, other prominent alterations include hippocampal myelination disruption, neuronal loss, and pronounced glial and inflammatory responses. Recent studies have established that parvalbumin (PV +) interneurons in the hippocampus are partially myelinated under normal conditions, representing a notable structural feature of this interneuron subtype. However, whe
