Leukoencephalopathy with calcifications, developmental brain abnormalities and skeletal dysplasia due to homozygosity for a hypomorphic CSF1R variant: A report of three siblings.

We report three siblings homozygous for CSF1R variant c.1969 + 115_1969 + 116del to expand the phenotype of "brain abnormalities, neurodegeneration, and dysosteosclerosis" (BANDDOS) and discuss its link with "adult leukoencephalopathy with axonal spheroids and pigmented glia" (ALSP), caused by heterozygous CSF1R variants. We evaluated medical, radiological, and laboratory findings and reviewed the literature. Patients presented with developmental delay, therapy-resistant epilepsy, dysmorphic features, and skeletal abnormalities. Secondary neurological decline occurred from 23 years in sibling

Upregulation of APAF1 and CSF1R in Peripheral Blood Mononuclear Cells of Parkinson's Disease.

Increased oxidative stress and neuroinflammation play a crucial role in the pathogenesis of Parkinson's disease (PD). In this study, the expression levels of 52 genes related to oxidative stress and inflammation were measured in peripheral blood mononuclear cells of the discovery cohort including 48 PD patients and 25 healthy controls. Four genes, including ALDH1A, APAF1, CR1, and CSF1R, were found to be upregulated in PD patients. The expression patterns of these genes were validated in a second cohort of 101 PD patients and 61 healthy controls. The results confirmed the upregulation of APAF1

Effect of prior chemotherapy on hematopoietic stem cell mobilization.

A number of studies have suggested that prior chemotherapy correlates negatively with the efficiency of hematopoietic stem cell mobilization. However, little data exist with regard to the relative effects of the specific chemotherapeutic drug classes. We retrospectively reviewed the records of 201 consecutive patients with nonmyeloid malignancies undergoing CD34+ cell mobilization with chemotherapy+granulocyte colony-stimulating factor (G-CSF). The number of prior chemotherapy courses correlated negatively with the peripheral CD34+ cell concentration (pCD34) on the first day of collection (P<0

Activated liver macrophages in human liver diseases.

Immunohistochemical analysis using monoclonal antibodies specific for cells of monocyte/macrophage lineage reveals that resident liver macrophages have a phenotype distinct from that of monocytes or activated liver macrophages. Liver macrophages consist of heterogeneous cell populations in maturation (matured 25F9-positive and immature 25F9-negative) but the ratio of two populations is constant in normal and diseased livers. The expression of CD14 is down-regulated in resident liver macrophages as compared to that in monocytes, while the expression of 25F9 is up-regulated. On the other hand, t

The role of interleukin-8 and other cytokines in the pathogenesis of Felty's syndrome.

OBJECTIVE: Felty's syndrome (FS) is defined as rheumatoid arthritis (RA) with neutropenia and, in some cases, splenomegaly; the outcome is primarily determined by the risk of infection, which is related to the degree of neutropenia. We analysed whether the clinical manifestations of FS, especially neutropenia, could be explained by abnormalities in cytokine production. METHODS: We examined the production in FS of five cytokines involved in the maturation and activation of polymorphonuclear cells (PMNs): IL-1 beta, TNF alpha, IL-8, G-CSF and GM-CSF. Because of the role of systemic IL-8 in neut

Regulation of parasite-induced eosinophilia: selectively increased interleukin 5 production in helminth-infected patients.

Production of the eosinophilogenic cytokines interleukin 3 (IL-3), granulocyte/macrophage colony-stimulating factor (GM-CSF), and IL-5 by mitogen-stimulated peripheral blood mononuclear cells was compared between 11 noneosinophilic individuals and seven patients with helminth-induced eosinophilia. Both the kinetics and quantities of IL-3 and GM-CSF were similar in the two groups. In contrast, IL-5 production at both the protein and the mRNA level was markedly greater in the eosinophilic patients, an observation suggesting that IL-5 may be particularly important in mediating the selective eosin

Tongue microstructure physically constrains division of labor in bumblebee foraging.

Bumblebee (Bombus terrestris) is a primitively eusocial species that sustains its colonies by dividing foraging duties between queens and workers. Previous research emphasizes hormonal, metabolic, and behavioral contrasts between the castes but seldom considers why queens relinquish foraging once workers emerge. Here, we link foraging efficiency to tongue microstructure, providing physical evidence for the shift in bumblebee foraging behavior.

Erysipelothrix rhusiopathiae Suppurative Arthritis in a 12-year-old Boy After an Unusual Fresh Water Exposure.

We report the case of a 12-year-old boy who experienced recurrent suppurative arthritis of the distal interphalangeal joint after near simultaneous exposure to a frog, a dog bite and lake water. Anaerobic cultures of synovial fluid obtained during operative debridement grew small round gray colonies ultimately identified as Erysipelothrix rhusiopathiae by routine laboratory tests and mass spectrometry.

Polycystic nephropathy in slender lorises (Loris lydekkerianus).

In a colony of slender lorises, 20 deaths that occurred over a period of 11 years were investigated postmortem. Juvenile/adult polycystic nephropathy was observed in one newborn and 13 adult slender lorises. Although polycystic kidney disease (PKD) in humans and other animals is known to be inherited, it is not clear whether kidney alterations in slender lorises are genetically transmitted, stress related, or induced by microbiological influences.

CD30+ anaplastic large cell lymphoma complicated by pyoderma gangrenosum with increased levels of serum cytokines.

We here present the case of a 5-yr-old girl with pyoderma gangrenosum (PG) in association with underlying CD30+ anaplastic large cell lymphoma with increased serum cytokine levels (interleukin-8, granulocyte colony-stimulating factor). An association between PG and increased cytokine levels was suggested. Even in children, dermatosis of PG should receive prompt careful evaluation for underlying hematological malignancy.

Pyogranulomatous peritonitis associated with Nocardia sp.-like organisms in a raccoon (Procyon lotor).

During 1992 raccoons (Procyon lotor) were live-trapped in northeastern Pennsylvania (USA). In one of these animals a localized pyogranulomatous peritonitis was seen. Grossly a large mass with a central area of liquifactive necrosis was present in the anterior abdomen. Microscopically the lesion contained multiple colonies of filamentous organisms with histomorphologic and histochemical charcteristics resembling Nocardia sp. This appears to be the first report of Nocardia infection in the raccoon.

Evidence for genetic involvement in feline dilated cardiomyopathy.

Quantitative genetic evaluation of clinical dilated cardiomyopathy (DCM) was conducted in a large cattery with known history. Data showed that clinically affected cats were significantly more interrelated than randomly chosen case-control populations from the same colony. The results of this study suggest that quantitative inheritance, either interactive with or independent of nutrition or presently unknown factors, is involved in feline dilated cardiomyopathy.

Intestinal lymphomas in golden hamsters (Mesocricetus auratus).

Pathological examination of 226 adult hamsters culled in a commercial breeding colony because of health deterioration revealed malignant lymphomas arising from the intestines of 24 animals. Histologically the tumours were classified as centrocytic-centroblastic lymphomas. They were transplantable with cellular suspension, but not with serum or cell-free ascitic fluid of the tumour bearers. The tumor did not metastasize into the spleen. Other types of lymphomas were not diagnosed.

Early events in the suppression of myeloid leukemic cells by biological regulators.

Differentiation of mouse and human myeloid leukemic cells in vitro can be induced by some members of the granulocytemacrophage family of colony-stimulating factors. In the mouse, the most active molecule (G-CSF) is able to suppress leukemic stem cell self-generation in an irreversible, asymmetric process, suggesting that the factor permanently modifies newly synthesized one or more daughter chromatids in dividing leukemic stem cells.

Acute rheumatic fever or rheumatoid arthritis? The role of hydroxychloroquine: a case report.

BACKGROUND: Rheumatic fever is endemic in our part of the world. Arthritis is the most common clinical presentation and is characteristically migratory in nature. Rheumatoid-like arthritis in acute rheumatic fever is not described in literature. Although arthritis is typically treated with nonsteroidal anti-inflammatory drugs, the role of hydroxychloroquine remains less well studied. CASE PRESENTATION: A 24-year-old Pakistani female individual presented with symmetrical polyarthritis involving the small joints of the hands along with morning stiffness. She was diagnosed with rheumatic heart d

Establishment of a rapid and quantitative method for detecting the range of infection exposure in preclinical dental education.

BACKGROUNDS: Safe dental treatments that prevent nosocomial and cross-infections are essential for patients and dental workers. However, dental students sometimes pay inadequate attention to infection control, especially in preclinical practice, because of too much focus on technical training, such as the use of equipment, etc. The spread of infections such as SARS-CoV-2, antibiotic-resistant bacteria, and oral bacteria are sometimes lethal for medically compromised patients. Thus, the rapid and inexpensive detection system to detect and measure dental practice-related infection spread during

Microbial contamination risks and clinical safety in platelet-rich plasma therapy and evaluation of rapid microbial detection methods.

INTRODUCTION: Platelet-rich plasma (PRP) is widely used for pain relief, wound healing, and so on. However, recent reports have documented cases of infections occurring after PRP therapy. In this study, we aimed to examine potential microbial contamination risks associated with PRP therapy and evaluate rapid microbial detection methods suitable for clinical use. METHODS: We assessed the risk of microbial contamination during blood collection, PRP preparation, and sterility testing. To evaluate suitable detection methods, we compared the microbial detection sensitivities of flow cytometry (FCM

SPHENISCID ALPHAHERPESVIRUS 1 IN AFRICAN PENGUINS (SPHENISCUS DEMERSUS) WITHIN THE SPECIES SURVIVAL PLAN© MANAGED POPULATION IN THE US: INCIDENCE AND POPULATION IMPACT.

Spheniscid alphaherpesvirus-1 (SpAHV-1) is associated with respiratory disease in juvenile African penguins (Spheniscus demersus). In 2020, this virus was detected in adult birds with clinical signs of respiratory disease in a previously asymptomatic colony of 21 birds following a recommended introduction of three new birds, including two with a history of herpesvirus as juveniles. Mild to moderate respiratory signs were noted in 33% (8/24) of the colony. SpAHV-1 DNA was detected in respiratory secreta by herpesvirus consensus PCR in three birds, although as the definitive source of the presen

Stress adaptation under in vitro evolution influences survival and metabolic phenotypes of clinical and environmental strains of Vibrio cholerae El-Tor.

Bacterial adaptation to stress can lead to phenotypic variants with diverse levels of niche competitiveness, pathogenicity, and antimicrobial resistance. In this work, we employed experimental evolution to investigate whether exposure to various stress conditions results in new phenotypic and metabolic properties in clinical and environmental strains of Vibrio cholerae. Our findings revealed the emergence of variants with metabolic and genetic variations and enhanced survival under stress compared to the parental isolates. Phenotypic changes in the evolved variants included colony morphology,
