Trauma Exposure, PTSD, and Methylation of the Blood Brain Barrier Claudin-5 Gene.

Posttraumatic stress disorder (PTSD) is associated with early onset of neurological conditions, but the mechanism by which PTSD relates to diseases of the central nervous system is unclear. One possibility is that PTSD perpetuates breakdown of the blood brain barrier (BBB), allowing for bidirectional passage of molecules across the periphery and central nervous system that promote neuropathology. Preclinical studies have implicated claudin-5 (CLDN5), a protein integral to the integrity of the BBB tight junctions, in the pathogenesis of depression. Based on this, we evaluated if trauma exposure

Serum levels of Wnt5a in Egyptian women with obesity and their association with toll like receptor 2 Arg753Gln gene polymorphism in a pilot case control study of obesity as a state of metaflammation.

Female obesity is a worldwide health issue linked to chronic metabolic low-grade inflammation (metaflammation) causing multiple obesity-related co-morbid conditions. We aimed to assess the serum levels of wingless integration site family member 5 A (Wnt5a), leptin, and tumor necrosis factor-alpha (TNF-α) as markers of obesity-associated metaflammation and investigate the association with toll-like receptors2 (TLR2) gene (Arg753Gln) single nucleotide polymorphism (SNP) among Egyptian females. The study included 60 females with obesity and 30 matched controls. Serum levels of Wnt5a, leptin, and

The regulatory roles of pparα on hepatic drug-metabolizing enzymes in primary sclerosing cholangitis Syrian hamsters.

Primary sclerosing cholangitis (PSC) is a chronic liver disease characterized by bile duct inflammation and fibrosis, ultimately leading to liver dysfunction. The role of peroxisome proliferator-activated receptor α (pparα) in regulating hepatic drug-metabolizing enzymes, including cytochrome P450s (P450s) and UDP-glucuronosyltransferases (UGTs), in PSC remains insufficiently understood. In this study, wild-type and pparα -/- Syrian hamsters were used to establish a PSC model, aiming to investigate the effects of pparα on the expression and activity of P450 and UGT enzymes. Sequential windowed

Assessment of health effects of potato crop phytopharmaceuticals and storage products in a murine model.

Potatoes are among the most widely consumed staple foods worldwide, but their cultivation and storage frequently involve multiple phytopharmaceutical products (PPPs), raising concerns about the health risks of dietary pesticide residues. The health effects of multi-residue PPPs used in potato cultivation were assessed in an in vivo murine model, involving 36 mice, and evaluated through omics analyses. Two field cultivation methods (conventional and organic) and two post-harvest storage conditions (using 1,4-dimethylnaphthalene and mint essential oil as sprout inhibitor treatments) were conside

Airway overlap syndromes with asthma.

The airways, defined as the segment of the respiratory system extending from the pharynx to the alveoli, represent a crucial site for various inflammatory airway diseases. Among the most common are asthma, chronic obstructive pulmonary disease (COPD), bronchiectasis, and obstructive sleep apnea (OSA). Owing to their high individual prevalence, it is not unusual for a single patient to have 2 or more of these conditions simultaneously. Beyond this epidemiologic coincidence, several pathophysiologic links, despite notable differences among the diseases, have been identified. These connections in

A bioluminescence-based chemical screen identifies a bactericidal naphthalene scaffold targeting MmpL3 in Mycobacterium abscessus.

Mycobacterium abscessus pulmonary disease (Mabs-PD) presents a significant and growing global health threat, particularly in individuals with underlying lung conditions like cystic fibrosis and chronic obstructive pulmonary disease. A key challenge in treating Mabs-PD is the lack of bactericidal antibiotics effective at therapeutically relevant concentrations, underscoring an urgent need for drug discovery. Targeting cell-wall synthesis is a promising approach, as evidenced by the success of broad-spectrum β-lactam antibiotics and the frontline antituberculosis drug isoniazid. However, these a

Mechanisms, efficacy, and future perspectives of cellular-based therapies for liver fibrosis/cirrhosis: focusing on mesenchymal stromal cells.

Liver fibrosis/cirrhosis, characterized by excessive deposition of extracellular matrix (ECM) and formation of fibrous scars, arises from chronic liver injury and poses a significant global health burden. Although liver transplantation remains the sole curative option, its application is limited by donor scarcity, immune rejection risks, and high costs. Cellular therapies, particularly those based on mesenchymal stromal cells (MSCs), have emerged as a promising alternative. This review comprehensively examines the therapeutic mechanisms and clinical efficacy of diverse cell therapies for liver

Finite element analysis of a neural implant for cytostatic hypothermia and a novel heat management system.

The treatment of glioblastoma (GBM) presents significant challenges, with median survival rates remaining low despite standard-of-care therapies. A novel approach, cytostatic hypothermia (CH), is under development against GBM; it is a window of temperature (typically 20 °C-25 °C) which halts tumor growthin vivo.Objective.This feasibility study expands upon the findings through the computational evaluation of a fully implantable system. Our simulations evaluate a thermoelectric cooler with a microwire array (NeuraTEC) and a novel ambient recirculating core (ARC) to achieve uniform cooling of a

microRNA-22 Inhibition Stimulates Mitochondrial Homeostasis and Intracellular Degradation Pathways to Prevent Muscle Wasting.

MicroRNA-22 (miR-22) is a negative regulator of mitochondrial biogenesis, as well as lipid and glucose metabolism, in metabolically active tissues. Silencing miR-22 holds promise as a potential treatment of obesity and metabolic syndrome, as it restores metabolic capacity-enhancing oxidative metabolism-and reduces ectopic fat accumulation in chronic obesity, a driver of impaired metabolic flexibility and muscle mass loss. Intramuscular adipose accumulation and defective mitochondrial function are features associated with obese-mediated muscle atrophy and hallmarks of neuromuscular disorders su

A rare case of a simultaneous post-dissection saccular aneurysm of the ascending aorta and large pulmonary artery aneurysm with secondary embolism: a case report.

BACKGROUND: Aneurysms of the pulmonary arteries and the ascending aorta are rare, and both bear a high mortality risk if left untreated. In general, these entities are primarily caused by etiologies such as hypertension, pulmonary arterial hypertension, infection or congenital disorders. Treatment requires a rapid diagnostic work-up or even immediate surgical intervention in acute cases. Nevertheless, surgery entails serious perioperative risks, in particular in patients with multiple comorbidities. CASE PRESENTATION: We discuss a 70-year-old woman presented with decompensated heart failure b

Uncovering Novel Extracellular Matrix Transcriptome Alterations in Lesions of Multiple Sclerosis.

The extracellular matrix (ECM) of the central nervous system (CNS) is an interconnected network of proteins and sugars with critical roles in both homeostasis and disease. In neurological diseases, excessive ECM deposition and remodeling impact both injury and repair. CNS lesions of multiple sclerosis (MS), a chronic inflammatory and degenerative disease, cause prominent alterations of the ECM. However, there are a lack of data investigating how the multitude of ECM members change in relation to each other and how this affects the MS disease course. Here, we evaluated ECM changes in MS lesions

Using Passive and Active Data to Predict Post-Traumatic Stress Disorder Symptoms and Cannabis Use in Recently Discharged UK Veterans: A Protocol for the MAVERICK Feasibility Study.

INTRODUCTION: Post-traumatic stress disorder (PTSD) and comorbid cannabis use disorder (CUD) is a growing concern amongst UK veterans. Co-occurrence of problematic cannabis use, and PTSD is associated with greater PTSD symptom severity, decreased likelihood of cannabis use cessation, worse clinical outcomes, and increased societal burden. Despite increased screening efforts among veterans, there are no effective strategies for predicting risk for PTSD and problematic drug use escalation before these conditions develop, worsen, or become chronic. METHODS: This feasibility study employs a 4-wee

Targeting ferroptosis: a new therapeutic opportunity for kidney diseases.

Ferroptosis is a form of non-apoptotic regulated cell death (RCD) that depends on iron and is characterized by the accumulation of lipid peroxides to lethal levels. Ferroptosis involves multiple pathways including redox balance, iron regulation, mitochondrial function, and amino acid, lipid, and glycometabolism. Furthermore, various disease-related signaling pathways also play a role in regulating the process of iron oxidation. In recent years, with the emergence of the concept of ferroptosis and the in-depth study of its mechanisms, ferroptosis is closely associated with various biological co
