Supportive neural mechanisms underlying successful emotion regulation for reappraisal success.

Although the prefrontal cortex (PFC) is well-known for its role in emotion regulation, the interactions between cortical and subcortical regions that facilitate this process remain incompletely understood. The present study aims to enhance our understanding of the brain pathways and connectivity involved in emotion regulation. Thirty healthy, right-handed participants were presented with a total of 48 aversive images characterized by low normative valence and high arousal, along with 24 neutral photos. The preprocessing pipeline included slice timing correction, motion correction, structural a

The Rostral Ventrolateral Medulla Input to Central Amygdala Regulates Anxiety-Like Behaviors in Mice for Chronic Light Exposure.

BACKGROUND: Emerging evidence increasingly links environmental light exposure to the development of anxiety disorder. The rostral ventrolateral medulla (RVLM), a key brainstem center that regulates sympathetic outflow and blood pressure (BP), has been implicated in the autonomic dysregulation frequently observed in anxiety. High blood pressure is a recognized exacerbating factor for anxiety-related pathology. Although aberrant projections from the central amygdala (CeA) to brainstem regions have been reported in multiple mood disorders, it remains unclear whether the CeA receives input from th

A GABAergic neural circuit from the suprachiasmatic nucleus to DG drives cognitive impairment in migraine mice.

BACKGROUND: Cognitive impairment and circadian rhythm disturbances are prevalent comorbidities of migraine, yet their underlying neural circuit mechanisms remain poorly understood. The suprachiasmatic nucleus (SCN), as the master circadian pacemaker, and the hippocampus, essential for memory, are key regions involved in these symptoms. The dentate gyrus (DG) in the hippocampus acts as the main entry point for information and is essential for pattern separation, a key memory process. We hypothesized that a direct neural pathway from the SCN to the DG might link circadian dysfunction to cognitiv

Parental mental illness and child brain structure: A diffusion MRI study of emotion regulation related pathways.

BACKGROUND: Children of parents with mental illness (at-risk) face an increased risk for developing mental health issues themselves than children of healthy parents (controls), yet the neurobiological underpinnings remain unclear. This study explored differences in white matter (WM) between at-risk children and controls, focusing on brain pathways related to emotion regulation and the associations between parenting behaviours, stress, and WM microstructural properties. METHODS: WM properties were assessed via diffusion-weighted imaging in 64 children (28 at-risk, 36 control) across five emoti

Abnormal dynamic functional connectivity in different gray matter volume subtypes of patients with major psychiatric disorders.

BACKGROUND: Major psychiatric disorders (MPDs), including schizophrenia (SZ), bipolar disorder (BD), major depressive disorder (MDD), and obsessive-compulsive disorder (OCD), have been classified as distinct diagnostic categories based on clinical symptoms. However, neuroimaging studies suggest that these MPDs share similar brain structure and functional characteristics. The present study aim to explore potential subtypes of MPDs based on gray matter volume (GMV) in a trans-diagnostic sample. METHODS: We involved 84 patients with MPDs (22 SZ, 22 BD, 18 MDD, 22 OCD) and 39 healthy controls (HC

Localization of ictal fear and related brain network.

BACKGROUND: Ictal fear (IF) is a common psychiatric manifestation in focal epilepsy, yet its electrophysiological mechanisms and associated brain networks remain poorly understood. Existing models are largely extrapolated from canonical fear-processing frameworks rather than direct intracranial evidence. METHODS: We retrospectively analyzed stereo-electroencephalography (SEEG) recordings from patients with drug-resistant epilepsy who exhibited clinically and electrophysiologically confirmed IF. Epileptogenicity Index (EI) analysis was used to classify brain regions into epileptogenic, propaga

Neurovascular coupling dysfunction of the frontal‒thalamic circuit in postconcussion syndrome.

Postconcussion syndrome (PCS) is characterized by alterations in perfusion and neural activity within specific regions, such as the frontal‒thalamic circuit. Despite extensive research on the neurovascular coupling (NVC) function of the PCS, how the involved brain regions interact as a dynamic network in the processing of cognitive and behavioral functions is still elusive. We recruited 38 participant with PCS and 38 healthy controls for magnetic resonance imaging examinations. We subsequently investigated four types of NVC, namely, amplitude of low-frequency fluctuation (ALFF), fractional ALF

A neuromodulatory circuit-to-molecular pathway for reformatting aversive memories during recall.

Memories can be altered when they are recalled through the process of reconsolidation, requiring gene expression in brain cells that store these memories. How brain circuits reformat memories during recall by directing molecular signaling in specific neuronal populations is not known. Here, we show that brainstem noradrenaline projections to the amygdala, a brain region that stores aversive emotional memories, control memory reconsolidation in rats. During reconsolidation, this circuit regulates the nuclear translocation of CREB-regulated transcriptional coactivator-1 (CRTC1), a molecule impor

Resolving mesoscale brainstem-prefrontal-striatal pathways underlying decisions upon salient events using submillimeter-resolution fMRI.

Value-based decision-making engages brain-wide motivational, cognitive, and motor processes. Yet, information integration and gating that culminate in immediate decisions upon salient events likely occur within small neural nuclei and cortical layers at the mesoscale not resolved with conventional human neuroimaging. Using submillimeter-resolution 7 T functional MRI with acquisition-matched anatomical references and a lottery choice task incorporating salient superhigh stakes, we dissociated mesoscale operations spanning a brainstem-prefrontal-striatal pathway during choice and outcome process

Nonlinear Functional Connectivity and ICA Reveal Default Mode Network Hyperconnectivity in Parkinson's Disease: A Resting-State fMRI Study.

Parkinson's disease (PD) disrupts intrinsic brain networks that support motor and cognitive functions. Using resting-state fMRI from 138 PD patients and 54 controls, we combined independent component analysis (ICA) with nonlinear functional connectivity (FC) based on distance correlation. Compared with conventional Pearson-based measures, nonlinear FC revealed stronger and spatially distinct connectivity patterns, especially in parietal and sensorimotor regions. Group comparisons showed robust differences (p < 1×10⁻⁷), with four ICA networks (Components 4, 10, 19, and 20) displaying consistent

Brunner's Glands in the Duodenal Bulb Are Indirectly Innervated by Vagal Preganglionics That Connect With Cholinergic Enteric Neurons in the Myenteric Plexus.

Canonical parasympathetic innervation of tissues/organs is accomplished by two in-series neurons, often called pre- and postganglionic neurons. Such two-neuron in-series organization has recently been challenged by claiming direct vagal preganglionic innervation of Brunner's glands (BGs) in the duodenal bulb, as well as the spleen, and the liver. To resolve this controversial issue, we labeled vagal preganglionic neurons in the dorsal motor nucleus of the vagus (DMV) by injecting a Cre-dependent AAV construct to express tdTomato in ChAT-IRES-Cre transgenic mice and analyzed the distribution of

Brain Connectivity and Machine Learning Approaches to assess the underlying neurobiology and prediction accuracy of anorexia nervosa: A replication study.

Resting-state fMRI has been used to study aberrant functional connectivity properties in patients with anorexia nervosa (AN) at several stages of the illness. One popular way to extract these metrics is to use graph theory to showcase aberrant brain connectivity between patients with AN versus controls. However, most studies use classic analyses to investigate these differences, which could limit the number and choices of features used in one model. Instead, machine learning models have proven to be a promising tool in studying the functional connectivity of various disorders. In this study, w

Insular cortex-lateral habenula circuit mediates chronic itch-associated affective comorbidities.

Pruritus triggers scratching-induced skin damage and anxiety/depression, forming a vicious cycle that exacerbates inflammatory skin diseases. The insular cortex (IC) integrates itch and emotional processing, while the lateral habenula (LHb) regulates depression.Using acute (3-day) and chronic (14-day) histamine-induced murine itch models combined with chemogenetic manipulation, we demonstrate that the insular cortex (IC) bidirectionally regulates acute itch and anxiety-like behaviors, whereas chronic itch engages the IC-lateral habenula (LHb) circuit to drive comorbid depression. These results

Asthma's impact on brain function: an investigation of changes in functional connectivity and network topology.

Asthma is a reversible condition characterized by airflow limitation and chronic airway inflammation. Previous neuroimaging studies have confirmed brain structural and functional abnormalities associated with asthma, primarily occurring in regions such as the anterior cingulate cortex, superior frontal gyrus, occipital lobe, postcentral gyrus, and corpus callosum. These changes may be attributable to asthma-related systemic inflammation. This study investigates modifications in the topological organization of functional brain networks in individuals with asthma using functional magnetic resona

Brain network dynamics in the wake-sleep transition reorganize according to task engagement.

Alertness fluctuations and cognitive processing comprise overlapping brain circuits that include frontoparietal, thalamocortical, and sensory pathways. It is unclear how these brain circuits can simultaneously support such disparate but foundational human processes. To develop an integrated framework of consciousness and cognition, it is important to understand how fluctuations in alertness and cognitive processing interact in their shared circuits. In this electroencephalography (EEG)-functional magnetic resonance imaging (fMRI) study, we assessed individuals who fluctuated between alert and

Altered Effective Connectivity of Precuneus Subregions and Associated Gene Profiles in Chronic Insomnia Disorder Patients Before and After Treatment.

BACKGROUND: Previous studies have highlighted the crucial role of the precuneus in the hyperarousal state of insomnia. However, the causal effects between its subregions and other regions remain unclear. Both traditional Chinese medicines (TCMs) and benzodiazepines are effective in insomnia treatment, but limited studies have compared the therapeutic mechanisms of these two treatments. METHODS: This study included 82 chronic insomnia disorder (CID) patients and 80 healthy controls (HCs). CID patients were assigned to receive either modified suanzaoren decoction, a TCM for treating insomnia, o
