Anti-Periprosthetic Infection Strategies: From Implant Surface Topographical Engineering to Smart Drug-Releasing Coatings.

Despite advanced implant sterilization and aseptic surgical techniques, periprosthetic bacterial infection remains a major challenge for orthopedic and dental implants. Bacterial colonization/biofilm formation around implants and their invasion into the dense skeletal tissue matrices are difficult to treat and could lead to implant failure and osteomyelitis. These complications require major revision surgeries and extended antibiotic therapies that are associated with high treatment cost, morbidity, and even mortality. Effective preventative measures mitigating risks for implant-related infect

Oral Cavity as a Source of Mesenchymal Stem Cells Useful for Regenerative Medicine in Dentistry.

The use of mesenchymal stem cells (MSCs) for regenerative purposes has become common in a large variety of diseases. In the dental and maxil … here are different sources of MSCs that have been individually proposed as promising candidates for tissue engineering protocols. The therapeutic strategy based on MSCs can be direct, by using cells as components of the tissue to be regenerated, or indirect, aimed at delivering local growth factors, cytokines, and chemokines produced by the MSCs. Here, the authors o …

Altered Adipose-Derived Stem Cell Characteristics in Macrodactyly.

Macrodactyly is a congenital disease characterized by aggressive overgrowth of adipose tissue in digits or limbs frequently accompanied with hyperostosis and nerve enlargement; its pathological mechanism is poorly understood. Adipose-derived stem cells (ASCs) have been extensively studied in tissue engineering and regenerative medicine as an ideal alternative substitute for bone marrow-derived mesenchymal stem cells (BM-MSCs), but their pathological role is largely unknown. In this study, ASCs from macrodactyly adipose tissues (Mac-ASCs) were isolated and compared to ASCs derived from the norm…

Transcriptional comparisons between equine articular repair tissue, neonatal cartilage, cultured chondrocytes and mesenchymal stromal cells.

Human and equine cell transplant strategies for cartilage lesions usually result in scar tissue that is similar to what is produced naturally during the repair process. In this study, culture-expanded de-differentiated chondrocytes and primary bone marrow stromal cells at a pre-transplantation time-point were compared along with neonatal cartilage to repair tissue. Transcriptional profiling using a 9413-probeset equine-specific cDNA microarray and targeted real-time quantitative polymerase chain reaction validation were used to characterize relationships between these cell types and repair tis

Engineering Immunoregenerative Therapy via an Immunomodulatory Binary Pharmacology Hydrogel Depot for Prolonged Allograft Survival.

Immunosuppressive therapy following organ transplantation is essential for ensuring long-term graft survival but leaves patients vulnerable to complications such as infection, malignancy, and severe side effects. Immunosuppressants currently in use are typically hydrophobic with low oral bioavailability and must be taken indefinitely to maintain immune tolerance. Furthermore, these drugs lack tissue repair capacity, which limits their effectiveness in transplantation. To address this clinical gap, an immunomodulatory hydrogel (iGEL) was developed, integrating prodrug engineering with inflammat

[NELL-1: a novel highly efficient and specific growth factor].

Regeneration of bone tissue, as well as other tissues, requires involvement and interaction of cells, scaffolds and relevant growth factors, among which growth factors play a crucial role in maintaining the stability of microenvironment. Nel-like-type 1 molecule (NELL-1), a novel growth factor in tissue engineering, has been studied intensively in recent years. Researches mainly covered gene and protein structure and their expression profiling, biological function, molecular mechanisms and disease relevance. NELL-1 expressed in embryonic tissue is essential for growth and development of bone t…

Prevascularized grafts with spatially organized MSC spheroids to accelerate therapeutic angiogenesis in ischemic disease.

OBJECTIVE: Ischemic diseases, characterized by impaired blood flow and progressive tissue necrosis, remain a major challenge in regenerative medicine. Surgical revascularization remains the gold standard for restoring blood flow in major vessels, but still shows limited effects on microvascular regeneration. To address this unmet need, various strategies in therapeutic angiogenesis have been explored to induce microvascular formation, including delivery of bioactive molecules, stem cells, or pre-vascularized grafts. However, injection-based approaches often suffer from off-target effects, and

A novel recombinant attenuated Salmonella carrying RGD and melittin genes induces apoptosis of B16 cells and inhibits melanoma growth.

The tumor provides a favorable microenvironment for the growth of anaerobes and facultative anaerobes, such as Salmonella. Melittin (MEL) is a major component of bee venom that has a distinct inhibitory effect on tumor cell metastasis and invasion, but its clinical application is limited owing to non-specific cytotoxicity. Arg-Gly-Asp (RGD) is a potent competitive antagonist of extracellular matrix binding to αvβ3, and is often used as a tumor-targeting marker. Here, we engineered Salmonella typhimurium to selectively express and release RGD-MEL. The engineered bacteria were intratumourally in

Biomimetic mineralized collagen nanostructures orchestrate osteogenesis and osteoimmunomodulation to improve osseointegration of titanium alloy microporous interfaces.

Inadequate osseointegration remains a major clinical challenge in joint replacement surgery, frequently resulting in aseptic loosening and necessitating revision procedures. Herein, we present a novel approach that combines 3D printing, tissue engineering, and biomimetic technologies to address this problem. We developed a novel surface modification strategy to engineer two functional interfaces on microporous titanium implants (PTi): an intrafibrillar/extrafibrillar mineralized collagen interface (I-EMC/PTi) and an extrafibrillar mineralized collagen interface (EMC/PTi). The fabrication proce…

Engineered Protein-Iron Oxide Hybrid Biomaterial for MRI-traceable Drug Encapsulation.

Labeled protein-based biomaterials have become a popular for various biomedical applications such as tissue-engineered, therapeutic, or diagnostic scaffolds. Labeling of protein biomaterials, including with ultrasmall super-paramagnetic iron oxide (USPIO) nanoparticles, has enabled a wide variety of imaging techniques. These USPIO-based biomaterials are widely studied in magnetic resonance imaging (MRI), thermotherapy, and magnetically-driven drug delivery which provide a method for direct and non-invasive monitoring of implants or drug delivery agents. Where most developments have been made u

Drug discovery and therapeutic delivery for the treatment of B and T cell tumors.

Hematological malignancies manifest as lymphoma, leukemia, and myeloma, and remain a burden on society. From initial therapy to endless relapse-related treatment, societal burden is felt not only in the context of healthcare cost, but also in the compromised quality of life of patients. Long-term therapeutic strategies have become the standard in keeping hematological malignancies at bay as these cancers develop resistance to each round of therapy with time. As a result, there is a continual need for the development of new drugs to combat resistant disease in order to prolong patient life, if
