Mitochondrial AIF loss causes metabolic reprogramming, caspase-independent cell death blockade, embryonic lethality, and perinatal hydrocephalus.

OBJECTIVES: Apoptosis-Inducing Factor (AIF) is a protein involved in mitochondrial electron transport chain assembly/stability and programmed cell death. The relevant role of this protein is underlined because mutations altering mitochondrial AIF properties result in acute pediatric mitochondriopathies and tumor metastasis. By generating an original AIF-deficient mouse strain, this study attempted to analyze, in a single paradigm, the cellular and developmental metabolic consequences of AIF loss and the subsequent oxidative phosphorylation (OXPHOS) dysfunction. METHODS: We developed a novel A

Chronic low-grade inflammation drives skeletal aging and neurocognitive decline: inflammaging as a central hub coupling bone-brain aging.

BACKGROUND: Global population aging has driven a marked rise in the co-prevalence of osteoporosis and cognitive impairment, and a bidirectional epidemiological association between the two conditions is now supported by multiple meta-analyses. The shared biological mechanisms underlying this comorbidity, however, remain incompletely defined, and the two disorders continue to be managed within largely separate clinical disciplines. AIM AND SCOPE: This review consolidates existing evidence into an integrative immunopathological framework in which inflammaging-the chronic, low-grade, sterile syst

Neuro faces of beneficial T cells: essential in brain, impaired in aging and neurological diseases, and activated functionally by neurotransmitters and neuropeptides.

T cells are essential for a healthy life, performing continuously: immune surveillance, recognition, protection, activation, suppression, assistance, eradication, secretion, adhesion, migration, homing, communications, and additional tasks. This paper describes five aspects of normal beneficial T cells in the healthy or diseased brain. First, normal beneficial T cells are essential for normal healthy brain functions: cognition, spatial learning, memory, adult neurogenesis, and neuroprotection. T cells decrease secondary neuronal degeneration, increase neuronal survival after central nervous sy

High sulfate exacerbates heavy metal toxicity in Chinese cabbage (Brassica chinensis L.) by impairing plant ROS scavenging in heavy metals contaminated soil.

Accumulation of heavy metals (HMs) and sulfates in farmland near mines is one of the most serious environmental problems posing a threat to crops and human health. Moderate sulfate alleviates the toxicity of HMs in crops, but the impact of high sulfate on crops in HM-contaminated soil remains unclear. Hence, we integrated physiological, transcriptomic and metabolomic profiling to investigate the effect of high sulfate on Chinese cabbage (Brassica chinensis L.) under HMs through controlled pot experiments. According to our results, compared with the HMs only group, the growth indicators of Chin

The role of NLRP3 inflammasome in age-related macular degeneration: mechanisms and therapeutic prospects.

Age-related macular degeneration (AMD) is a fundus oculi disease that progressively impairs the central vision of patients. To date, its pathogenesis has not been fully elucidated, and therapeutic options for dry AMD remain limited. Recently, chronic low-grade inflammation has been recognized as an important pathogenic factor in various neurodegenerative diseases, including AMD. The NLRP3 inflammasome, a key component of the innate immune system, has emerged as a critical integrator of retinal stress signals. This review first delineates the molecular architecture and activation modalities of

Hematological complications in solid organ transplant recipients with telomere biology disorders: a narrative review.

Telomeres are repetitive nucleotide sequences at the ends of chromosomes that preserve genomic integrity. Defects in telomere maintenance mechanisms lead to premature telomere shortening, resulting in cellular senescence, apoptosis, and organ dysfunction, collectively termed telomere biology disorders (TBDs). Short telomere length is associated with an increased risk of end-stage fibrotic disease of the lung and/or liver, which may necessitate lung or liver transplantation. Beyond pulmonary and hepatic involvement, TBDs can also affect cardiac and renal function. Importantly, the bone marrow f…

Mitochondria-targeted esculetin mitigates age-associated cardiac remodeling through coordinated activation of AMPK-SIRT1 and inhibition of p38 MAPK signaling.

Age-associated cardiac remodeling, characterized by hypertrophy and fibrosis, is a major contributor to cardiovascular morbidity in the elde … axis. Depletion of either AMPK or SIRT1 markedly reduced the ability of these compounds to suppress cellular senescence, hypertrophic responses, and fibrotic signaling, indicating a critical role for this pathway. AMPK functioned upstream of SIRT1, as SIRT1 activation was attenuated following AMPK depletion. In parallel, both compounds significantly suppressed p38 MA …

Everolimus suppresses glucose transporter 3 membrane trafficking to improve therapeutic efficacy of umbilical cord blood-derived mesenchymal stem cell transplantation in diabetic retinopathy.

Diabetic retinopathy (DR) is a microvascular and retinal neurologic disorder that occurs in patients with long-term diabetes. Umbilical cord blood-derived mesenchymal stem cell (UCB-MSC) therapy has emerged as a promising treatment because of its regenerative potential; however, its effectiveness is limited under hyperglycemic conditions, which results in the overproduction of mitochondrial reactive oxygen species (mtROS), leading to cellular senescence. In this study, we examined the potential of everolimus, a mammalian target of rapamycin (mTOR) inhibitor, to enhance the efficacy of MSCs in …

Hyperglycemia Leads to BMSC Impaired Osteogenesis, Enhanced Adipogenesis, and Altered Metabolism.

Diabetes is a major risk factor for osteoporosis, which negatively impacts bone health, but the mechanisms underlying the effects of hyperglycemia on bone marrow mesenchymal/stromal cells (BMSC) are not fully understood. This study investigated how high glucose levels influence BMSC differentiation, proliferation, viability, and metabolism. The results demonstrated that high glucose inhibits osteogenesis in human BMSC, as evidenced by reduced alkaline phosphatase activity, impaired calcium deposition, and downregulation of key osteogenic genes (RUNX2, ALP). Conversely, high glucose conditions
