Co-morbidity in Gaucher's disease results of a nationwide enquiry in Spain.

SHORT INTRODUCTION: Gaucher's disease (GD) is an autosomal recessive disease produced by mutations of the Glucocerebrosidase gene. Carriers are considered to be healthy subjects because there is no manifestation of the disease, but they show signs of macrophage disfunction. The aim of the study was to determine if GD patients and non affected carriers risk suffering other diseases when compared to healthy non-carrier relatives. DESIGN: Epidemiologic study of historic cohorts. The fact that they have one or two mutated alleles has been considered to be the risk factor leading to other conditio

Oncogenicity testing of 2-ethylhexanol in Fischer 344 rats and B6C3F1 mice.

2-Ethylhexanol (2EH) is a weak nongenotoxic hepatic peroxisome proliferator in the rat. It is a high-volume chemical intermediate in the preparation of the plasticizers bis-(2-ethylhexyl) adipate (DEHA), bis-(2-ethylhexyl) phthalate (DEHP), and tris-(2-ethylhexyl) phosphate (TEHP), which are weak hepatocellular tumorigens in female mice. In consequence, the oncogenic potential of 2EH was evaluated in male (M) and female (F) rats and mice (50 animals/sex/group). Oral gavage doses of 2EH in 0.005% aqueous Cremophor EL (polyoxyl-35 castor oil) were given five times a week to rats: 0 (water), 0 (v

CT-guided indocyanine green fluorescence localization demonstrates superior reliability over hook-wire for pulmonary ground-glass nodules: a retrospective cohort study.

BACKGROUND: Lung cancer is the leading cause of cancer-related mortality worldwide. With the widespread use of computed tomography (CT) screening, pulmonary ground-glass nodules are increasingly detected. Video-assisted thoracoscopic surgery (VATS) enables minimally invasive resection; however, the precise intraoperative localization of these non-palpable nodules remains challenging. The current commonly used preoperative CT-guided hook-wire localization, while effective, is associated with documented risks such as dislodgement (reported in up to ~10% of cases), pneumothorax requiring interven

Bone marrow transplantation from genetically HLA-nonidentical donors in children with fatal inherited disorders excluding severe combined immunodeficiencies: use of two monoclonal antibodies to prevent graft rejection.

OBJECTIVE: For children with life-threatening inborn errors of metabolism without a matched related bone marrow donor, transplantation from an HLA genetically nonidentical donor is the only therapeutic option. To reduce the high risk of graft rejection in this setting without increasing the conditioning regimen, a protocol based on the infusion of an antiadhesion antibody directed against the CD11a (leukocyte function-associated antigen 1 [LFA-1]) molecule was performed by the European Bone Marrow Transplantation-European Society for Immunodeficiency group with promising results. To optimize e

Pre-employment examinations for preventing injury, disease and sick leave in workers.

BACKGROUND: Many employers and other stakeholders believe that health examinations of job applicants prevent occupational diseases and sickness absence. This is an update of the original Cochrane review (Mahmud 2010). OBJECTIVES: To evaluate the effectiveness of pre-employment examinations of job applicants in preventing occupational injury, disease and sick leave compared to no intervention or alternative interventions. SEARCH METHODS: We searched CENTRAL (the Cochrane Library), MEDLINE, EMBASE, CINAHL, PsycINFO and PEDro (up to 31 March 2015). We did not impose any restrictions on date, la

Digital Order System Redesign in Emergency-to-Intensive Care Unit Admissions: Prospective Observational Study.

BACKGROUND: Emergency-to-intensive care unit (ICU) admissions are high-stakes transitions in care, where delays or documentation errors can compromise patient safety and disrupt operational efficiency. Although digital order systems can support these workflows, traditional platforms often lack real-time traceability and structured input logic, leaving them vulnerable to duplicate submissions, lost orders, and misaligned ICU bed reservations. OBJECTIVE: The study aimed to examine whether an updated digital order system was associated with differences in emergency-to-ICU admission workflows, in

Bacopa monnieri confers neuroprotection against middle cerebral artery occlusion (MCAO)-induced neurobehavioral and biochemical dysregulation in the frontal cortex and hippocampus of male Wistar rats.

ETHNOPHARMACOLOGICAL RELEVANCE: Bacopa monnieri (Brahmi) is a well-known medicinal herb widely used in traditional Ayurvedic and Unani systems of medicine for enhancing memory, cognition, and mental health. The medication serves as a treatment for patients who experience stress-related conditions and anxiety disorders and epilepsy symptoms and age-related cognitive deterioration. The research builds upon conventional ethnopharmacological methods by using scientific data which demonstrates Bacopa monnieri safeguards brain cells from ischemic damage thus validating its historical application for

The IDEA3 pilot trial: feasibility, acceptability, and preliminary efficacy of an online adaptation of an evidence-based intervention for campus sexual assault prevention.

BACKGROUND: Enhanced Assess, Acknowledge, Act (EAAA) is the only intervention shown in a randomized controlled trial to reduce sexual violence victimization against undergraduate women by 50%. EAAA is an in-person, small-group intervention led by two trained peer facilitators over four three-hour sessions. Implementation cost and train-the-trainer complexities have led to limited uptake with relatively few institutions implementing the intervention, despite its efficacy. Adapting EAAA for online delivery would enable use of a centralized facilitator pool, dramatically cutting cost and complexi

Reinventing amlodipine.

Attention-deficit/hyperactivity disorder (ADHD) is a prevalent neurodevelopmental disorder, and pharmacological treatments have limited mechanistic specificity. Most nonstimulants target noradrenergic tone but show modest efficacy. l-Type calcium channels (LTCCs) modulate neuronal excitability, catecholaminergic transmission, cortical plasticity, and neuroinflammation, processes central to ADHD pathophysiology. This review evaluates the evidence for repurposing the LTCC blocker amlodipine as a novel ADHD therapeutic. We propose a mechanistic framework where amlodipine acts within attention/aro

Distinct metabolic perturbations link liver steatosis and incident CVD in lean but not obese PWH.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a key risk factor for cardiovascular disease (CVD), potentially driven by shared metabolic mechanisms. Metabolic perturbations associated with MASLD and CVD remain underexplored in people with HIV (PWH). METHODS: We used data from the longitudinal multicenter 2000HIV study comprising 1895 virally suppressed PWH, out of which 970 had available liver and carotid artery measurements. Transient elastography with controlled attenuation parameter (CAP) was performed for the assessment of liver steatosis (CAP > 263 dB/m)

Ruxolitinib plus standard of care in severe hospitalized adults with severe fever with thrombocytopenia syndrome (SFTS): an exploratory, single-arm trial.

BACKGROUND: Severe fever with thrombocytopenia syndrome (SFTS) is an emerging tick-borne infectious disease, and its morbidity and mortality are increasing. At present, there is no specific therapy available. An exacerbated IFN-I response and cytokine storm are related to the mortality of patients with SFTS. Ruxolitinib is a Janus kinase (JAK) 1/2 inhibitor that can block proinflammatory cytokines and inhibit the type I IFN pathway. We aimed to explore the use of ruxolitinib plus standard of care for severe SFTS. METHODS: We conducted a prospective, single-arm study of severe SFTS. We recruit

Minimal and Maximal Models to Quantitate Glucose Metabolism: Tools to Measure, to Simulate and to Run in Silico Clinical Trials.

Several models have been proposed to describe the glucose system at whole-body, organ/tissue and cellular level, designed to measure non-accessible parameters (minimal models), to simulate system behavior and run in silico clinical trials (maximal models). Here, we will review the authors' work, by putting it into a concise historical background. We will discuss first the parametric portrait provided by the oral minimal models-building on the classical intravenous glucose tolerance test minimal models-to measure otherwise non-accessible key parameters like insulin sensitivity and beta-cell res

Impact of the skull contour definition on Leksell Gamma Knife® Icon™ radiosurgery treatment planning.

INTRODUCTION: The Gamma Knife® planning software (TMR 10, Elekta Instruments, AB, Sweden) affords two ways of defining the skull volume, the "historical" one using manual measurements (still perform in some centers) and the new one using image-based skull contours. Our objective was to assess the potential variation of the dose delivery calculation using consecutively in the same patients the two above-mentioned techniques. MATERIALS AND METHODS: We included in this self-case-control study, 50 patients, treated with GKRS between July 2016 and January 2017 in Lausanne University Hospital, Swit

Genetic structure in the southernmost populations of black-and-gold howler monkeys (Alouatta caraya) and its conservation implications.

Black-and-gold howler monkeys Alouatta caraya, are arboreal primates, inhabitants of Neotropical forests, highly susceptible to the yellow fever virus, considered early 'sentinels' of outbreaks, and thus, of major epidemiological importance. Currently, anthropogenic habitat loss and modifications threatens their survival. Habitat modification can prevent, reduce or change dispersal behavior, which, in turn, may influence patterns of gene flow. We explored past and contemporary levels of genetic diversity, elucidated genetic structure and identified its possible drivers, in ten populations (n =
