Motor-to-Limbic Design of Direct Synaptic Communication between Dopamine Neurons in the Midbrain.

Midbrain dopamine (DA) neurons are diverse with distinct subpopulations being essential for key functions of the brain: nigrostriatal DA neurons for voluntary movement and mesolimbic DA neurons for learning from reward prediction errors. In addition to being primarily associated with distinct senso-motor or limbic cortical-striatal circuits, DA subpopulations also directly communicate with each other via local DA release in the midbrain. While the inhibitory synaptic nature of this dopamine-to-dopamine signaling has been well established, the pre- and postsynaptic identity and logic of connect

Bidirectional Mendelian randomization study of causal associations between white-matter structural connectivity and multiple sclerosis.

BACKGROUND: Multiple sclerosis (MS) is associated with widespread network disruption, but whether specific white-matter structural connectivity (WMSC) phenotypes contribute causally to MS susceptibility remains unclear. METHODS: We performed bidirectional two-sample Mendelian randomization (MR) using genome-wide association study (GWAS) summary statistics for 206 tractography-derived WMSC phenotypes (UK Biobank; N = 26,333) and MS susceptibility (IMSGC; N = 115,803 of European ancestry). Primary inference used inverse-variance weighted (IVW) MR under a multiplicative random-effects model, com

Mapping Cerebellar Connectivity to Cognition in Psychosis: Convergent Evidence From Functional Magnetic Resonance Imaging and Transcranial Magnetic Stimulation.

BACKGROUND: Cerebellar dysfunction is strongly implicated in both cognitive impairments and psychosis, and the cognitive dysmetria hypothesis posits cognition as the intermediary in the pathway from cerebellar dysfunction to psychopathology. However, the nuanced cerebellum-behavior relationships in psychosis remain unclear. Establishing such a link is crucial to understand cerebellar mechanisms and to identify treatment targets for cognitive deficits in patients. METHODS: We studied 100 patients with early psychosis from the Human Connectome Project for Early Psychosis study. Connectome-based

Alterations in beta oscillatory activity occur with the emergence of parkinsonian motor signs across the basal ganglia-thalamocortical network.

Mounting evidence suggests that elevated beta oscillatory activity in the basal ganglia thalamocortical (BGTC) network is associated with the cardinal motor signs in people with Parkinson's disease (PD). The evolution of abnormal beta oscillatory activity across the BGTC network as motor signs emerge, however, is not well understood. The goal of this study was to investigate whether beta oscillatory activity in the BGTC network changes prior to and how it evolves during the emergence of mild parkinsonian motor signs. To address this question, we performed simultaneous neural recordings in the

Compensative inter-module functional connectivity enhancement at the latter half of verbal fluency task in patients with temporal lobe epilepsy.

BACKGROUND: Previous studies proposed that disrupted dynamic interaction between language subsystems diminishes temporal lobe epilepsy (TLE) patients' adaptability to changing cognitive loads. However, how the interaction is dynamically performed remained unclear. Due to the graphic syllabic logographic nature of Chinese characters, the Chinese Character verbal fluency (VFC) task provides changing demands throughout the task, being an ideal tool to analyze the dynamic interaction of language subsystems. METHODS: Twenty-nine neurotypical controls and 58 patients with TLE from an ongoing cohort

Energy state guides reward seeking via an extended amygdala to lateral hypothalamus pathway.

Impaired regulation of food intake underlies numerous health problems, including obesity and type 2 diabetes, yet how brain systems controlling reward seeking become dysregulated to promote overeating is unknown. Glutamatergic neurons of the lateral hypothalamic area (LHA) are thought to act as a brake on feeding, which is dysregulated during diet-induced obesity. These neurons receive input from the extended amygdala, including the bed nucleus of the stria terminalis (BNST). However, the circuit mechanisms underlying the ability of this pathway to control feeding behavior and how they contrib

The Bed Nucleus of the Stria Terminalis-Paraventricular Nucleus of the Hypothalamus Neural Circuit Regulates Neuropathic Pain Through the Brain-Spleen Axis.

Neuropathic pain is a chronic condition caused by damage or dysfunction in the nervous system. While the spleen may influence neuropathic pain, its role has been poorly understood. This study demonstrates that the spleen plays a crucial role in regulating neuropathic pain through the bed nucleus of the stria terminalis (BNST) - paraventricular nucleus of the hypothalamus (PVN) neural circuit in a chronic constriction injury (CCI) mouse model. Splenectomy, splenic denervation, or splenic sympathectomy significantly increased the mechanical withdrawal threshold (MWT) and reduced macrophage infil

Shared phonological networks in frontal and temporal cortex for language production and comprehension.

In this functional magnetic resonance imaging study, we investigated whether language production and understanding recruit similar phoneme-specific networks. We did so by comparing the brain's response to different phoneme categories in minimal pairs: Bilabial-initial words (eg "monkey") were contrasted to alveolar-initial words (eg "donkey") in 37 participants performing both language production and comprehension tasks. Individual-specific region-of-interest analyses showed that the same sensorimotor networks were activated across the language modalities. In motor regions, word production and

Inhibition of hippocampal or thalamic inputs to the nucleus accumbens reverses stress-induced alterations in dopamine system function.

BACKGROUND: Symptoms of psychosis are often observed in patients with post-traumatic stress disorder (PTSD) and are driven by aberrant regulation of the mesolimbic dopamine system. We have previously shown that targeting upstream brain regions that regulate dopamine neuron activity, the ventral hippocampus (vHipp), and paraventricular nucleus of the thalamus (PVT) maybe a novel approach to restore dopamine system function. The vHipp and PVT work in concert to regulate ventral tegmental area (VTA) dopamine neuron activity through a multisynaptic circuit that begins with inputs to the nucleus ac

Unveiling anomalies in visual processing regions in obsessive-compulsive disorder: evidence from gray matter volume and functional connectivity analyses.

Obsessive-compulsive disorder (OCD) is a prevalent neurological condition characterized by recurring obsessive thoughts and compulsive behaviors. The underlying mechanisms of the disorder remain poorly understood due to its considerable heterogeneity. Given the extensive brain impairments observed in OCD, this study aimed to investigate gray matter volume and functional connectivity in individuals with OCD using a multimodal neuroimaging approach, while also exploring the relationship between these neural changes and clinical symptom severity. Structural and resting-state functional magnetic r

Altered Basal Ganglia Network Topology Associated With Auditory-Motor Synchronization.

INTRODUCTION: Auditory-motor synchronization (AMS) embedded in Rhythmic Auditory Stimulation (RAS) is a validated method to improve gait, upper limb function, and motor speech in people with neurologic disorders like Parkinson's disease (PD). Predictable auditory cues optimize spatial movement patterns, and research has suggested that AMS reduces the brain's reliance on dopaminergic (DA) response in the ventral striatum. To gain a mechanistic understanding of the positive clinical outcomes related to AMS, this pilot study investigates the effects of AMS on the basal ganglia network (BGN) using

Effects of alcohol and cannabis co-use on salience network resting state functional connectivity in individuals who drink alcohol heavily.

INTRODUCTION: The salience network may be linked to addiction. Evidence suggests less salience network resting state functional connectivity (rsFC) from heavy alcohol use, but higher rsFC within and between brain networks from regular cannabis use. Given the rise in alcohol-cannabis co-use, the present study sought to elucidate rsFC between regions within the salience network and regions across the whole brain in individuals who use no drugs regularly, those who use alcohol only heavily, and those who co-use alcohol-cannabis. METHODS: This is a secondary analysis of three clinical laboratory

The correct connectivity of the DG-CA3 circuits involved in declarative memory processes depends on Vangl2-dependent planar cell polarity signaling.

In the hippocampus, dentate gyrus granule cells connect to CA3 pyramidal cells via their axons, the mossy fibers (Mf). The synaptic terminals of Mfs (Mf boutons, MfBs) form large and complex synapses with thorny excrescences (TE) on the proximal dendrites of CA3 pyramidal cells (PCs). MfB/TE synapses have distinctive "detonator" properties due to low initial release probability and large presynaptic facilitation. The molecular mechanisms shaping the morpho-functional properties of MfB/TE synapses are still poorly understood, though alterations in their morphology are associated with Down syndr

First Application of a Novel Brain Template: Motor Training Improves Cortico-cerebellar Connectivity in Cerebellar Ataxia.

People with cerebellar degeneration show characteristic ataxic motor impairments. Despite cerebellar dysfunction, they can improve motor performance through sensorimotor training. Yet, how such training changes functional brain networks affected by cerebellar degeneration is unknown. We here investigated neuroplastic changes in the cortico-cerebellar network after a 5-d forearm training in 40 patients with mild to severe cerebellar degeneration and 40 age- and sex-matched healthy controls. Human participants (41 female) were assigned to one of four motor training conditions, varying online vis

Causal relationships between white matter connectome and mental disorders: a large-scale genetic correlation study.

BACKGROUND: Abnormalities in white matter integrity in mental disorders have attracted widespread attention, yet the genetic correlations and causal effects between white matter structural connectome and various psychiatric conditions remain largely unexplored. METHODS: In this study, we employed linkage disequilibrium score (LDSC) and high-definition likelihood (HDL) methods to analyze genetic correlations between white matter connectome and mental disorders, followed by bidirectional two-sample Mendelian randomization (MR) analysis to investigate causal relationships. We utilized 206 white

Thalamocortical white matter connectivity and the entorhinal cortex as neural correlates of electroconvulsive therapy response in patients with schizophrenia spectrum disorder.

BACKGROUND: Electroconvulsive therapy (ECT) is an effective treatment for patients with treatment-resistant schizophrenia spectrum disorder (SSD); however, the underlying mechanisms of ECT remain unclear, and region-specific neuromodulation targets are not well defined. Abnormally reduced thalamo-prefrontal cortex (PFC) and increased thalamo-parietal cortex (PC) connectivity have been linked to schizophrenia pathophysiology. In this study, thalamocortical white matter (WM) connectivity was examined as a potential biomarker of ECT response, and the brain regions that mediate ECT-induced changes

The nucleus accumbens-prefrontal connectivity as a predictor of chronic low back pain.

The nucleus accumbens (NAc) and its prefrontal connections are implicated in the aetiology of chronic low back pain (CLBP). Animal and human studies suggest that the NAc and its connections play a critical role in the transition from acute to CLBP. However, whole-brain connectivity in individuals with longstanding CLBP has not been systematically investigated. Using a functional connectomics approach, we examined whether the 2 NAc subregions-shell and core-exhibit different whole-brain connectivity between CLBP patients and healthy controls (HCs; total N = 197). The identified connections were

Savings in visuomotor learning are associated with connectivity changes within a cerebello-thalamo-cortical network encoding movement errors.

Savings refer to faster relearning upon re-exposure to a previously experienced movement perturbation. One theory posits that the brain recognizes past errors, enabling more efficient learning from them. If this is the case, there should be a modification in the neural response to errors during re-exposure to the perturbation. To investigate this hypothesis, we used fMRI to measure brain activity as participants adapted to a visuomotor perturbation across two sessions spaced one day apart, focusing on neural responses to movement errors. The magnitude of the movement error was incorporated int

Abnormal Connectivity of the Head Neural Integrator in Cervical Dystonia.

BACKGROUND: Cervical dystonia is characterized by abnormal neck and head movements, possibly related to a dysfunction of the interstitial nucleus of Cajal (INC) and the head neural integrator, a system responsible for the control of head and eye movements. However, neuroanatomical evidence of alterations in the head neural integrator in cervical dystonia is sparse. OBJECTIVES: We investigated structural and functional integrity of the INC and its connections in cervical dystonia. METHODS: This cross-sectional, observational study compared 19 cervical dystonia patients and 21 healthy controls
