Chronic amitriptyline treatment attenuates nigrostriatal degeneration and significantly alters trophic support in a rat model of parkinsonism.

In addition to alleviating depression, long-term adaptive changes induced by antidepressants may regulate neural plasticity in the diseased brain, providing symptomatic and disease-modifying effects in Parkinson's disease. The present study investigated whether chronic treatment with a frequently prescribed tricyclic antidepressant was neuroprotective in a 6-hydroxydopamine (6-OHDA) rat model of parkinsonism. In lesioned animals, chronic amitriptyline (AMI; 5 mg/kg) treatment resulted in a significant sparing of tyrosine hydroxylase-immunoreactive (THir) neurons in the substantia nigra pars co

Craniosynostosis, psychomotor retardation, and facial dysmorphic features in a Spanish patient with a 4q27q28.3 deletion.

CASE REPORT: We describe an unusual clinical case with an 11-Mb deletion at 4q27 (chr4: 123094652-134164491), craniosynostosis (CS), mild psychomotor retardation, and facial dysmorphic features. This deletion involves 18 genes; FGF2, NUDT6, and SPRY1 are primarily or secondarily implicated in human cranial bone and sagittal suture development and could play an important role in CS. CONCLUSIONS: Clinicians should always contemplate genetic studies in patients with syndromic CS. Mutational targeted genetic testing is appropriate for patients with classical or specific CS syndrome. Nevertheless,

The medial frontal-prefrontal network for altered awareness and control of action in corticobasal syndrome.

The volitional impairments of alien limb and apraxia are a defining feature of the corticobasal syndrome, but a limited understanding of their neurocognitive aetiology has hampered progress towards effective treatments. Here we combined several key methods to investigate the mechanism of impairments in voluntary action in corticobasal syndrome. We used a quantitative measure of awareness of action that is based on well-defined processes of motor control; structural and functional anatomical information; and evaluation against the clinical volitional disorders of corticobasal syndrome. In patie

Primary visual response (M100) delays in adolescents with FASD as measured with MEG.

Fetal alcohol spectrum disorders (FASD) are debilitating, with effects of prenatal alcohol exposure persisting into adolescence and adulthood. Complete characterization of FASD is crucial for the development of diagnostic tools and intervention techniques to decrease the high cost to individual families and society of this disorder. In this experiment, we investigated visual system deficits in adolescents (12-21 years) diagnosed with an FASD by measuring the latency of patients' primary visual M100 responses using MEG. We hypothesized that patients with FASD would demonstrate delayed primary v

Abuse liability and reinforcing efficacy of oral tramadol in humans.

BACKGROUND: Tramadol, a monoaminergic reuptake inhibitor, is hepatically metabolized to an opioid agonist (M1). This atypical analgesic is generally considered to have limited abuse liability. Recent reports of its abuse have increased in the U.S., leading to more stringent regulation in some states, but not nationally. The purpose of this study was to examine the relative abuse liability and reinforcing efficacy of tramadol in comparison to a high (oxycodone) and low efficacy (codeine) opioid agonist. METHODS: Nine healthy, non-dependent prescription opioid abusers (6 male and 3 female) part

The impact of attentional, auditory, and combined cues on walking during single and cognitive dual tasks in Parkinson disease.

Auditory and attentional cues improve gait in Parkinson disease (PD), but it is unclear if combining the two cueing strategies offers additional benefit. Further, the effect of a secondary cognitive task on cue efficacy is unknown. Therefore, this study aimed to assess the effects of cue type and task complexity on gait in PD. 11 participants with PD, 11 age-matched controls, and 11 young controls performed 3 walking trials on a GAITRite walkway under the following cueing conditions: no cue (baseline), rhythmic auditory cue at 10% below (AUD-10) and 10% above (AUD+10) self selected cadence, at

A functional promoter polymorphism of neuronal nitric oxide synthase moderates prefrontal functioning in schizophrenia.

Cognitive deficits in tasks involving the prefrontal cortex such as working memory or verbal fluency are a key component of schizophrenia. This led to the hypofrontality hypothesis of schizophrenia, which is widely accepted even though molecular underpinnings are elusive. While disturbances of glutamatergic neurotransmission might play a role, other components have rarely been investigated. Recently, the promoter region of nitric oxide (NO) synthase-I (NOS-I, encoded by the gene NOS1), impacting on prefrontal glutamate transmission, has repeatedly been associated with schizophrenia. We thus te

Evidence for functional specialization of hippocampal subfields detected by MR subfield volumetry on high resolution images at 4 T.

Animal studies suggest an involvement of CA3 and dentate gyrus (CA3&DG) in memory encoding and early retrieval and an involvement of CA1 in late retrieval, consolidation and recognition. The aim of this study was to test if similar associations could be found between hippocampal subfield volumes measured in vivo using a manual parcellation scheme and selected scores of the California Verbal Learning Test II (CVLTII): total immediate free recall discriminability (IFRD), short free recall discriminability (SFRD), and delayed recall discriminability (DRD). 50 elderly subjects (25 controls and 25

Failure to engage spatial working memory contributes to age-related declines in visuomotor learning.

It is well documented that both cognitive and motor learning abilities decline with normative aging. Given that cognitive processes such as working memory are engaged during the early stages of motor learning [Anguera, J., Reuter-Lorenz, P., Willingham, D., & Seidler, R. Contributions of spatial working memory to visuomotor learning. Journal of Cognitive Neuroscience, 22(9), 1917-1930, 2010], age-related declines in motor learning may be due in part to reductions in cognitive ability. The present study examined whether age-related declines in spatial working memory (SWM) contribute to deficits

Altered motor network activation and functional connectivity in adult Tourette's syndrome.

Tourette's syndrome (TS) is a developmental neuropsychiatric disorder characterized by motor and vocal tics as well as psychiatric comorbidities. Disturbances of the fronto-striatal-thalamic pathways responsible for motor control and impulse inhibition have been previously described in other studies. Although differences in motor performance are well recognized, imaging data elucidating the neuronal correlates are scarce. Here, we examined 19 adult TS patients (13 men, aged 22-52 years, mean = 34.3 years) and 18 age- and sex-matched controls (13 men, aged 24-57 years, mean = 37.6 years) in a f

Basal functional connectivity within the anterior temporal network is associated with performance on declarative memory tasks.

Spontaneous fluctuations in the blood oxygenation level-dependent (BOLD) signal, as measured by functional magnetic resonance imaging (fMRI) at rest, exhibit a temporally coherent activity thought to reflect functionally relevant networks. Antero-mesial temporal structures are the site of early pathological changes in Alzheimer's disease and have been shown to be critical for declarative memory. Our study aimed at exploring the functional impact of basal connectivity of an anterior temporal network (ATN) on declarative memory. A heterogeneous group of subjects with varying performance on tasks

Visuotactile interactions in the congenitally acallosal brain: evidence for early cerebral plasticity.

Studies in patients with an isolated, congenital agenesis of the corpus callosum have documented potentials and limits of brain plasticity. Literature suggests that early reorganization mechanisms can compensate for the absence of the corpus callosum in unisensory tasks that involve interhemispheric transfer. It is unknown, however, how the congenitally acallosal brain processes multisensory information, which presumably requires interhemispheric transfer of modality-specific input. Therefore, we tested five patients with total and one patient with partial agenesis of the corpus callosum in a

Association of antenatal corticosteroids with mortality and neurodevelopmental outcomes among infants born at 22 to 25 weeks' gestation.

CONTEXT: Current guidelines, initially published in 1995, recommend antenatal corticosteroids for mothers with preterm labor from 24 to 34 weeks' gestational age, but not before 24 weeks due to lack of data. However, many infants born before 24 weeks' gestation are provided intensive care. OBJECTIVE: To determine if use of antenatal corticosteroids is associated with improvement in major outcomes for infants born at 22 and 23 weeks' gestation. DESIGN, SETTING, AND PARTICIPANTS: Cohort study of data collected prospectively on inborn infants with a birth weight between 401 g and 1000 g (N = 10

Decision making under risk condition in patients with Parkinson's disease: a behavioural and fMRI study.

We aimed to study whether previously described impairment in decision making under risky conditions in patients with Parkinson's disease (PD) is affected by deficits in using information about potential incentives or by processing feedback (in terms of fictitious gains and losses following each decision). Additionally, we studied whether the neural correlates of using explicit information in decision making under risk differ between PD patients and healthy subjects. We investigated ten cognitively intact PD patients and twelve healthy subjects with the Game of Dice Task (GDT) to assess risky d

Symmetrical hemispheric priming in spatial neglect: a hyperactive left-hemisphere phenomenon?

Hemispheric rivalry models of spatial neglect suggest that the left hemisphere becomes hyperactive following right-hemisphere lesions since the two hemispheres normally exert an inhibitory influence on each other via callosal connections. Using a masked hemifield priming paradigm, we investigated whether the putative change in hemispheric balance involves other, higher-order abstract representational systems in spatial neglect. Participants consisted of 12 neglect patients with right-hemisphere damage and three groups of control participants, i.e., 12 young healthy controls, 10 age-matched hea

Evaluation of a conceptual framework for retraining high-level mobility following traumatic brain injury: two case reports.

Traumatic brain injury (TBI) is the primary cause of death and disability for 18- to 45-year-olds. High-level mobility is important for many of the social, leisure, sporting, and employment roles of young adults. The aim of these case reports was to evaluate a conceptual framework for retraining high-level mobility after TBI. The progression of 2 patients who had sustained a severe TBI but had contrasting clinical presentations was monitored over 6 months. Patient 1 presented with left hemiplegia following a TBI 10 years earlier, whereas patient 2 presented with ataxia 2 months following a TBI

Anxiety, cognitive self-evaluation and performance: ERP correlates.

The relation between anxiety, cognitive self-evaluation, performance, and electrical brain activity (event-related potentials, ERPs) in a sustained attention task (Go/NoGo; SART) was investigated in 18 participants. No significant correlation was found between reaction times and anxiety (assessed by State and Trait Anxiety Inventory or STAI), and cognitive self-evaluation (assessed by Cognitive Failures Questionnaire or CFQ). N2 (ERP time-window 250-350ms) and P3 (350-650ms) amplitudes were found to be related to anxiety and cognitive self-evaluation. N2 amplitude increased in trait and state

White matter differences predict cognitive vulnerability to sleep deprivation.

OBJECTIVES: In other disciplines, white matter (WM) differences have been linked to cognitive impairments. This study sets out to clarify whether similar microstructural differences in WM tracts predict a person's cognitive vulnerability to the effects of total sleep deprivation (TSD). DESIGN: Participants completed a simple visual-motor task both before and after 24 h of TSD. Using a median split on the percent change in accuracy from pre-TSD to post-TSD, participants were separated into susceptibility groups. A diffusion tensor MR imaging (DTI) scan was acquired from each participant, and f
