Anticancer activity and embolization evaluation of open porous PBVHx embolic microspheres loaded with triptolide.

In this study, we developed triptolide (TP)-loaded open-pore PBVHx microspheres (TP@OPMs) and compared their anticancer and embolization capabilities with those of TP-loaded sealed PBVHx microspheres (TP@SMs). TP@OPMs, characterized by an open surface and interconnected internal pores, showed higher water absorption (2860%) and porosity (79.69%), but slightly lower TP encapsulation efficiency (79 ± 0.32%) compared to TP@SMs, and achieved sustained TP release of 75.29% over 21 days. All PBVHx microspheres without TP demonstrated good biocompatibility, but TP@OPMs exhibited superior inhibitory e

On-demand H₂S delivery from NIR-activated dithiocarbamate-functionalized AuAg microdroplets for cellular anti-inflammation.

The development of microscale biocompatible systems for precise regulation of biological processes presents considerable challenges, particularly in achieving spatiotemporal control over gaseous signaling molecules. In this work, we engineered coacervate microspheres through a self-assembly approach to co-encapsulate a hydrogen sulfide (H₂S) donor, histidine dithiocarbamate (His-DTC), together with near-infrared (NIR)-responsive gold-silver alloy nanoparticles (Au@Au/Ag NPs). These microspheres, with an average diameter of 13.6 ± 3.5 μm, exhibited strong photothermal conversion under 780 nm NI

PLXDC2 siRNA-Mediated Intervention Attenuates Microglial Senescence Through cGAS-STING Signaling.

Parkinson's disease (PD) is driven by neurodegeneration, iron accumulation, and microglial senescence, yet effective therapy is hindered by the blood-brain barrier (BBB). By constructing the largest-to-date single-cell atlas of the human substantia nigra, we identified a marked upregulation of PLXDC2 in PD microglia. Leveraging this finding, we developed a multifunctional biomimetic nanoplatform (HFn-GM@siPLXDC2/DFO/CeO2-NP). This system co-encapsulates the iron chelator deferoxamine and antioxidant CeO2 nanoparticles, carries PLXDC2-targeting siRNA, and features a ferritin-modified microglial

Hyaluronic acid engineered melanin-MOF nanoreactor synergistically remodeling redox and immune homeostasis for targeted acute lung injury therapy.

Reactive oxygen species (ROS)-triggered oxidative stress, inflammatory responses, and overactivation of immune cells are critical factors exacerbating acute lung injury (ALI). Conventional clinical therapies are often limited by non-specificity, single-target limitations, and significant side effects to precisely modulate the complex pathological microenvironment. Here, we designed a hyaluronic acid-engineered nanoreactor (HA@MZ) through in situ encapsulation of melanin (MA) within ZIF-67, allowing multi-target synergistic therapy of ALI. The HA coating facilitates mucus penetration and target

Biomimetic Nanoparticles Based on Tumor Cell Membrane Co-loaded with Vitamin E and Doxorubicin for Targeted Synergistic Treatment of Bladder Cancer.

The development of targeted nanotherapeutics that enhance tumor specificity while minimizing systemic toxicity remains a central challenge in bladder cancer treatment. Herein, we engineered a biomimetic nanoplatform (mPPE@DOX) by cloaking a poly(ethylene glycol)-block-poly(ε-caprolactone) (PEG-b-PCL) core, co-loaded with doxorubicin (DOX) and vitamin E, with a membrane derived from MB49 bladder cancer cells. This design leverages homologous targeting for improved tumor accumulation and combines the chemotherapeutic action of DOX with the chemosensitizing function of vitamin E. The resulting na

Nanoengineered extrusion-aligned tract bioprinting enables functional repair of spinal cord injuries.

Spinal cord repair demands biomaterials that replicate the aligned axonal architecture and mechanical softness of native tissue. However, most current scaffolds fail to support three-dimensional alignment and neuronal differentiation of human neural stem cells (hNSCs) in hydrated, low-stiffness environments. Here, we present NEAT (nanoengineered extrusion-aligned tract), a shear-stress-driven 3D bioprinting strategy that utilizes norbornene-functionalized collagen (NorCol) to generate highly aligned, mechanically stable hydrogels without post-processing. NEAT preserves the native triple-helica

Polymersomes preventing brain infiltration of CD177+ neutrophils to mitigate hemorrhagic transformation post-tPA thrombolysis.

As an intravenous thrombolytic agent, tissue plasminogen activator (tPA) is limited by hemorrhagic transformation (HT) and a narrow therapeutic time window. Here we develop ROS-triggered polymersomes conjugated with fibrin-targeting CREKA peptide (CP) for tPA encapsulation. CP@tPA achieves efficient thrombosis targeting and enhanced thrombolytic efficacy, however, it has not been able to avert the occurrence of hemorrhage subsequent to thrombolysis. Building from our clinical discovery that stroke patients suffer from tPA-induced HT featured strong expression of CD177, recombinant CD177 protei

Ion-Responsive Microneedles Delivering Subtype-Specific Mitochondrial Extracellular Vesicles from HEY1⁺ Cardiomyocytes for Cardiac Repair in Bama Minipigs with Myocardial Ischemia-Reperfusion Injury.

The pathogenesis of myocardial ischemia-reperfusion (MI/R) injury is intricately linked to mitochondrial dysfunction occurring during both the ischemic and reperfusion phases. Through single-cell transcriptome analysis, we identified a subpopulation of HEY1-high expressing cardiomyocytes (HEY1+ CMs) characterized by superior mitochondrial homeostasis. To leverage this, we isolated P5CS-type or ATP5B-type functional mitochondria from a ΔΨm-high subpopulation, which was obtained via membrane potential sorting following dual overexpression in HEY1+ CMs, and subsequently encapsulated them within H

TREM2 and microglial immunity in Alzheimer's disease: mechanisms, genetics, and therapeutic opportunities.

Alzheimer's disease (AD) is increasingly recognized as a disorder of innate immune dysregulation within the central nervous system. The triggering receptor expressed on myeloid cells 2 (TREM2), a microglial immunoreceptor, has emerged as a pivotal genetic risk factor for late-onset AD, underscoring the critical role of neuroimmune interactions in disease pathogenesis. This review synthesizes recent advances concerning TREM2's modulation of core microglial functions, including phagocytosis, inflammatory signaling, cellular metabolism, and survival, processes that are essential for responding to

Biomimetic Multichannel 3D MXene Nanofiber Sponges for Peripheral Nerve Regeneration.

Nerve guidance conduits play a critical role in bridging nerve gaps and facilitating neural repair. Recent advances highlight that structural optimization and functional enhancement can further enhance the regenerative outcomes. Herein, inspired by the structural features of celery stalks and native nerves, we propose a biomimetic multichannel three-dimensional (3D) MXene nanofiber sponge scaffold for reconstructing large-gap peripheral nerve defects. The scaffold integrates MXene-loaded sacrificial alginate microfibers, a gas-foamed 3D porous nanofiber sponge, and a gelatin methacryloyl hydro

When the test lies: Fatal cryptococcosis and the prozone effect in advanced HIV infection.

Cryptococcal meningitis is a life-threatening opportunistic infection in advanced HIV disease. Cryptococcal antigen (CrAg) detection by lateral flow assay (LFA) is the diagnostic cornerstone, offering high sensitivity and specificity. However, false-negative results may arise from the prozone effect - antigen excess disrupting antibody-antigen lattice formation. We report a 37-year-old man with newly diagnosed advanced HIV infection (CD4+ count 26 cells/µL) presenting with altered mental status. Initial CrAg LFA on serum and cerebrospinal fluid (CSF) was negative. India ink staining revealed e

Mesenteric gastrointestinal stromal tumor mimicking a pelvic mass - the diagnostic value of vascular pedicle tracing and multiplanar reconstructions: A case report.

Primary mesenteric gastrointestinal stromal tumors (GISTs), also referred to as extra-gastrointestinal stromal tumors (EGISTs), are rare entities and represent a diagnostic challenge due to their extraintestinal location and predominantly exophytic growth pattern. On imaging, they may mimic pelvic, gynecologic, or retroperitoneal masses, leading to diagnostic delay or inappropriate management. We report the case of a 53-year-old woman presenting with chronic pelvic discomfort, in whom contrast-enhanced computed tomography (CT) revealed a well-circumscribed abdominopelvic mass initially suspect

Broad Ligament Fibroid Mimicking an Ovarian Tumor: A Case Report.

Broad ligament leiomyomas are rare extrauterine smooth muscle tumors that can mimic adnexal neoplasms because of their atypical location and degenerative changes. We report the case of a 47-year-old multiparous woman with lower abdominal pain for five to six months, worsening over the preceding eight to 10 days. Ultrasonography demonstrated a large predominantly solid-cystic abdominopelvic mass with minimal vascularity, with neither ovary separately visualized. Magnetic resonance imaging (MRI) revealed a well-defined mass centered in the right broad ligament region, showing a T2-hypointense so

Development and Evaluation of Multi-Module Retinal Devices for Artificial Vision Applications.

Artificial retinal devices require a high-density electrode array and mechanical flexibility to effectively stimulate retinal cells. However, designing such devices presents significant challenges, including the need to conform to the curvature of the eyeball and cover a large area using a single platform. To address these issues, we developed a parylene-based multi-module retinal device (MMRD) integrating a complementary metal-oxide semiconductor (CMOS) system. The proposed device is designed for suprachoroidal transretinal stimulation, with each module comprising a parylene-C thin-film subst

Regeneration of Volumetric Muscle Loss Using MSCs Encapsulated in PRP-Derived Fibrin Microbeads.

Volumetric muscle loss (VML) is one of the major types of soft tissue injury frequently encountered worldwide. In case of VML, the endogenous regenerative capacity of the skeletal muscle tissue is usually not sufficient for complete healing of the damaged area resulting in permanent functional musculoskeletal impairment. Therefore, the development of new tissue engineering approaches that will enable functional skeletal muscle regeneration by overcoming the limitations of current clinical treatments for VML injuries has become a critical goal. Platelet-rich plasma (PRP) is an inexpensive and r

Vitamin B12-PEG4000 functionalized cellulose nanofibers for Amphotericin B delivery: In vitro evaluation for leishmaniasis treatment.

Leishmaniasis is a globally neglected infectious disease caused by protozoan parasites, and its treatment is often limited by several side effects. Amphotericin B (AmB) is highly effective anti-leishmanial drugs; however, its oral administration is restricted due to low bioavailability and toxicity. To overcome these limitations, a novel formulation (VitB12-PEG4000-AmB-CNFs) was developed for delivery of AmB using Vitamin B12-PEG4000 conjugate and functionalized cellulose nanofibers. The optimized formulation exhibits a particle size of 490 nm with a positive surface charge (+28.53 mv), and a

Recent advances in nano-based drug delivery systems for treatment of liver cancer.

Liver cancer is one of the aggressive primary tumors as evident by high rate of incidence and mortality. Conventional treatments (e.g. chemotherapy) suffer from various drawbacks including wide drug distribution, low localized drug concentration, and severe off-site toxicity. Therefore, they cannot satisfy the mounting need for safe and efficient cancer therapeutics, and alternative novel strategies are needed. Nano-based drug delivery systems (NDDSs) are among these novel approaches that can improve the overall therapeutic outcomes. NDDSs are designed to encapsulate drug molecules and target
