Identification of Association Between Mitochondrial Dysfunction and Sarcopenia Using Summary-Data-Based Mendelian Randomization and Colocalization analyses.

BACKGROUND: Mitochondrial dysfunction has been demonstrated to be an important hallmark of sarcopenia, yet its specific mechanism remains obscure. In this study, mitochondrial-related genes were used as instrumental variables to proxy for mitochondrial dysfunction, and summary data for sarcopenia-related traits were used as outcomes to examine their genetic association. METHODS: A total of 1 136 mitochondrial-related genes from the human MitoCarta3.0 database were extracted. Genetic instruments for them were obtained from gene expression quantitative trait locus (eQTLs) study (n = 31 684). Ag

The rationale and design of the PERindopril GENEtic association study (PERGENE): a pharmacogenetic analysis of angiotensin-converting enzyme inhibitor therapy in patients with stable coronary artery disease.

BACKGROUND: Angiotensin-converting enzyme (ACE) inhibitors reduce clinical symptoms and improve outcome in patients with hypertension, heart failure, and stable coronary artery disease (CAD) and are among the most frequently used drugs in these patient groups. For hypertension, treatment is guided by the level of blood pressure. In the secondary prevention setting, there are no means of guiding therapy. Prior attempts to target ACE-inhibitors to those patients that are most likely to benefit have not been successful, mainly due to the consistency in the treatment effect in clinical subgroups.

Preimplantation Genetic Testing for FMR1 CGG Repeat Expansions: Clinical and Molecular Aspects in Assisted Reproduction.

Fragile X syndrome is a monogenic disorder caused by the expansion of CGG trinucleotide repeats in the fragile X mental retardation 1 (FMR1) gene and is a major cause of inherited intellectual disability and reproductive impair-ment. Preimplantation genetic testing for monogenic disor-ders (PGT-M) has become an important strategy in assisted reproduction to prevent the transmission of FMR1-related disorders. This narrative review aimed to summarize cur-rent evidence on the molecular basis, technical approaches, clinical outcomes, and challenges associated with PGT-M for FMR1 CGG repeat expansi

Possible relationship between genetic polymorphisms and the risk of exercise addiction among Turkish elite athletes: an exploratory pilot study.

Exercise addiction (EA) is a complex behavioral phenotype characterized by a loss of control over exercise despite adverse physical, psychological, and social consequences. Although its psychological and performance-related aspects have been widely studied, its underlying etiology remains largely unclear, leading to increased interest in potential genetic vulnerability. Therefore, an exploratory pilot study investigated genetic variants associated with EA among elite athletes using a genome-wide association study (GWAS). The study comprised 168 Turkish elite athletes. Allele frequencies of cri

Potentially Functional Variants of DCTD and ENTPD2 in the Metabolism of Nucleotide Pathway Genes Predict Survival of HBV-Related Hepatocellular Carcinoma Patients.

Purpose: Nucleotide metabolism plays a critical role in cancer development, but the prognostic significance of genetic variants in nucleotide metabolism genes for hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC) patients remains unclear. Methods: We performed Cox proportional hazards regression analyses to evaluate the association between genetic variants in 94 nucleotide metabolism-related genes and overall survival (OS) in 866 HBV-HCC patients. To assess the potential biological relevance of the identified variants, the Bayesian false discovery probability and false-positive re

The outcomes of surrogate pregnancy using donor egg compared to surrogate's egg: a systematic review.

PURPOSE: Surrogacy is either gestational (GS) using donor egg or traditional (TS) using surrogate's own egg. GS is increasing, partly from assumptions over less difficulty in transferring the baby to intended parents. This systematic review investigates obstetric outcomes in GS vs TS and vs surrogate's own spontaneous conceptions (SC), comparing genetically unrelated embryos versus genetically related embryos. METHODS: Included were studies with historical or concurrent GS to TS or to SC comparisons, reporting any obstetric outcomes. MEDLINE, Embase, Global Health, and PsycINFO databases were

[Genetic analysis of a fetus with renal dysplasia/hypoplasia due to a variant of GREB1L gene and literature review].

OBJECTIVE: To carry out clinical and genetic analysis for a fetus with absent kidneys and bladder. METHODS: Clinical data of the fetus were collected. Whole exome sequencing (WES) and Sanger sequencing were carried out on fetal tissue and peripheral blood samples from its family members. A RDDC online tool was used to predict the impact of the variant on the GREB1L gene splicing, which was further validated by in vitro minigene assays. Long-range PCR was employed to verify the exonic deletional variant. An I-TASSER server was used to predict the three-dimensional structure of the mutant prote

Genetic evidence for causality between thyroid function and endometriosis: A bidirectional 2-sample Mendelian randomization study.

Accumulating observational evidence suggests an association between thyroid function and endometriosis, though the causal relationship remains unclear. To investigate potential bidirectional causal relationships, including for endometriosis subtypes, we conducted a bidirectional 2-sample Mendelian randomization (MR) analysis utilizing summary genetic data. Data sources included the ThyroidOmics Consortium (free thyroxine [FT4], thyroid-stimulating hormone [TSH], subclinical hypothyroidism, subclinical hyperthyroidism: N = 72,167, thyroid peroxidase antibody [TPOAb]: N = 18,297), IEU database (

Shared genetic regulators and diagnostic biomarkers in gallbladder stone and cholangiocarcinoma: A comprehensive comparative transcriptomic analysis.

Gallbladder stone (GS) and cholangiocarcinoma (CHOL) are common biliary tract disorders that share certain clinical and pathological characteristics but exhibit distinct molecular hallmarks. While GS-related inflammation, cholestasis, and epithelial injury-repair responses may provide a biologically plausible link, the shared genetic mechanisms underlying the pathogenesis of both diseases remain largely unexplored. The aim of this study was to elucidate this shared genetic landscape using a comparative transcriptomic approach. We identified 671 and 5197 differentially expressed genes (DEGs) in

Expanding the phenotypic spectrum of THAP1 dystonia: a scoping review with an illustrative case of late-onset focal limb dystonia.

THAP1-associated dystonia (DYT-THAP1) is classically described as an early-onset disorder with craniocervical involvement and potential for generalization. However, increasing reports suggest a broader phenotypic spectrum, including adult-onset presentations. We aimed to characterize the clinical phenotype of genetically confirmed DYT-THAP1 with age at onset ≥ 40 years, and to highlight an illustrative case. A scoping review was conducted following Joanna Briggs Institute methodology. Comprehensive searches of PubMed, MDSGene, Google Scholar, and regional databases identified studies reporting

Association of the COMT Gene Polymorphism rs4680 with Cognitive Impairment in Schizophrenia: A Narrative Review.

BACKGROUND: Cognitive impairment in schizophrenia patients is characterized by decreased functioning, reduced quality of life, and is a predictor of a more severe course of the disease. The rs4680 variant of the COMT gene (Val158Met), which encodes catechol-O-methyltransferase, affects dopamine metabolism in the prefrontal cortex and is a key genetic modifier of cognitive endophenotypes. However, the associations of the rs4680 alleles with the severity of cognitive impairment remain unclear. This review summarizes and critically re-evaluates the evidence on the role of rs4680 in the developmen

Carbapenem-resistant Morganella morganii carrying blaKPC-2 or blaNDM-1 in the clinic: one-decade genomic epidemiology analysis.

UNLABELLED: Carbapenem-resistant Morganella morganii (CRMM) isolates, particularly those producing Klebsiella pneumoniae carbapenemase-2 (KPC-2) or New Delhi metallo-β-lactamase-1 (NDM-1), are increasingly being recognized as causative agents of nosocomial infections. However, systematic phylogeography and genetic characterization of these isolates worldwide are still lacking. Here, through seven years of surveillance of CRMM in a tertiary hospital, we analyzed the genomic characteristics of blaKPC-2- or blaNDM-1-positive CRMM isolates. Furthermore, we conducted a global genomic epidemiologica

High familial risks in some rare cancers may pinpoint to hidden germline genetics: focus on esophageal, stomach, small intestinal, testis, thyroid and bone cancers.

BACKGROUND: Germline genetic susceptibilities of rare cancers of the esophagus, stomach, small intestine, testis, (nonmedullary) thyroid gland and bone with high familial risks are not well known. Here, we use familial risk data from the Swedish Family-Cancer Database which contains records of cancers in Swedish families obtained over a century. We compare familial risks for offspring diagnosed with any of these cancers when their parent had or had not that cancer. We review the global literature of the reported constitutional variants that may explain part of the familial risk. MAIN BODY: Fa
