Investigating the etiology of Haff disease: Optimization and validation of a sensitive LC-MS/MS method for palytoxins analysis in directly associated freshwater and marine food samples from Brazil.

Haff disease typically develops after eating contaminated marine or freshwater species, especially fish. Despite still having an unknown etiology, recent reports have suggested its possible correlation with palytoxins. Therefore, the present work aimed to optimize and perform a validation of a sensitive method using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) for the analysis of palytoxin and some of its analogs, with the main purpose of investigating their presence in marine and freshwater food samples associated with Haff disease in Brazil. The method optimization

Detection of Coxiella burnetii DNA in sheep and goat milk and dairy products by droplet digital PCR in south Italy.

Coxiella burnetii is a Gram-negative obligate intracellular bacterium that is responsible for Q fever, a common zoonosis which is present virtually worldwide. This microorganism infects a wide range of wild and domestic mammals, but the main reservoirs are cattle, goats and sheep, which also represent sources of human infection. A potential route of transmission of this pathogen to humans is the consumption of C. burnetii-contaminated raw milk or dairy products derived from contaminated raw milk, although the role of these foods as possible infection sources is controversial. The aims of this

Toxoplasma gondii Oocyst Infectivity Assessed Using a Sporocyst-Based Cell Culture Assay Combined with Quantitative PCR for Environmental Applications.

Toxoplasma gondii is a ubiquitous foodborne protozoan that can infect humans at low dose and displays different prevalences among countries in the world. Ingestion of food or water contaminated with small amounts of T. gondii oocysts may result in human infection. However, there are no regulations for monitoring oocysts in food, mainly because of a lack of standardized methods to detect them. The objectives of this study were (i) to develop a reliable method, applicable in biomonitoring, for the rapid detection of infectious oocysts by cell culture of their sporocysts combined with quantitativ

Household Socioeconomic and Demographic Correlates of Cryptosporidium Seropositivity in the United States.

BACKGROUND: Cryptosporidium are parasitic protozoa that infect humans, domestic animals, and wildlife globally. In the United States, cryptosporidiosis occurs in an estimated 750,000 persons annually, and is primarily caused by either of the Cryptosporidium parvum genotypes 1 and 2, exposure to which occurs through ingestion of food or water contaminated with oocytes shed from infected hosts. Although most cryptosporidiosis cases are caused by genotype 1 and are of human origin, the zoonotic sources of genotype 2, such as livestock, are increasingly recognized as important for understanding hu

Worldwide Surveillance of Foodborne Disease: the Need to Improve †.

A foodborne-disease surveillance program is an essential part of a food safety program. Foodborne surveillance should be able to issue early alerts on contaminated food to which a large population is exposed; collate notifications of enteric diseases and laboratory isolations; report foodborne disease incidents on a regular basis; and use sentinel and specific epidemiological studies as required. Although most countries have some kind of reporting of notifiable diseases, few have foodborne-disease surveillance programs, and little is known of foodborne disease in general on a worldwide basis.

Foodborne and Waterborne Disease in Canada - 1982 Annual Summary.

Data on foodborne disease in Canada in 1982 are compared with those for 1981. A total of 988 incidents comprising 791 outbreaks and 197 single cases, caused illnesses in 7,384 persons in 1982. These figures are greater than for 1981, and, in fact, the numbers of incidents and cases are the highest on record. Salmonella , Staphylococcus aureus , Clostridium perfringens and Bacillus cereus caused most of the illnesses. The main Salmonella serovars were S. typhimurium , S. muenster and S. infantis . Escherichia coli 0157:H7 hemorrhagic colitis was reported for the first time. Parasitic diseases w

Genomic Epidemiology of Salmonella enterica Circulating in Surface Waters Used in Agriculture and Aquaculture in Central Mexico.

Salmonella enterica can survive in surface waters (SuWa), and the role of nonhost environments in its transmission has acquired increasing relevance. In this study, we conducted comparative genomic analyses of 172 S. enterica isolates collected from SuWa across 3 months in six states of central Mexico during 2019. S. enterica transmission dynamics were assessed using 87 experimental and 112 public isolates from Mexico collected during 2002 through 2019. We also studied genetic relatedness between SuWa isolates and human clinical strains collected in North America during 2005 through 2020. Amon

Engineered Reporter Phages for Rapid Bioluminescence-Based Detection and Differentiation of Viable Listeria Cells.

The pathogen Listeria monocytogenes causes listeriosis, a severe foodborne disease associated with high mortality. Rapid and sensitive methods are required for specific detection of this pathogen during food production. Bioluminescence-based reporter bacteriophages are genetically engineered viruses that infect their host cells with high specificity and transduce a heterologous luciferase gene whose activity can be detected with high sensitivity to indicate the presence of viable target cells. Here, we use synthetic biology for de novo genome assembly and activation as well as CRISPR-Cas-assis

Visual detection platform based on RPA-CRISPR/Cas12a for Klebsiella pneumoniae and Carbapenem-resistant Klebsiella pneumoniae in clinical and food safety settings.

INTRODUCTION: The rise of Klebsiella pneumoniae (KP) and carbapenem-resistant KP (CRKP) poses grave threats to public health and food safety, creating an urgent demand for rapid point-of-care testing (POCT). Traditional detection methods are limited by laboratory barriers, making them unsuitable for POCT implementation. METHODS: Herein, a one-tube assay integrating recombinant polymerase amplification (RPA) with CRISPR/Cas12a technology was developed for the rapid, sensitive and specific detection of KP and blaOXA-48-carrying CRKP. Specific primers targeting the KP-specific rpoB gene and carb

Occurrence, behaviour, and human exposure pathways and health risks of toxic geogenic contaminants in serpentinitic ultramafic geological environments (SUGEs): A medical geology perspective.

Serpentinitic ultramafic geological environments (SUGEs) contain toxic geogenic contaminants (TGCs). Yet comprehensive reviews on the medical geology of SUGEs are still lacking. The current paper posits that TGCs occur widely in SUGEs, and pose human health risks. The objectives of the review are to: (1) highlight the nature, occurrence and behaviour of TGCs associated with SUGEs; (2) discuss the human intake pathways and health risks of TGCs; (4) identify the key risk factors predisposing human health to TGCs particularly in Africa; and (5) highlight key knowledge gaps and future research dir

Antimicrobial activity of pomegranate (Punica granatum L.) fruit peels.

Food-borne diseases such as listeriosis and diseases caused by the emergence of multi-drug resistant pathogens (e.g. Staphylococcus aureus) are globally recognized as environmental hazards to the food supply and human health. Natural inhibitors for pathogenic microorganisms have been explored in many plants. The antimicrobial activity against some food-borne pathogens by various extracts from pomegranate fruit peels was evaluated using both in vitro (agar diffusion) and in situ (food) methods. The 80% methanolic extract of peels (WME) was a potent inhibitor for Listeria monocytogenes, S. aureu

Outbreak of typhoid fever in vaccinated members of the French Armed Forces in the Ivory Coast.

In 2001, an outbreak of typhoid fever occurred among the members of the French Armed Forces. All had received a typhoid vaccination as per the immunization schedule practiced in the Armed Forces (every 5 years). A retrospective cohort study was conducted in 94 personnel. The objectives were to confirm the diagnosis, determine the source of contamination and identify the factors associated with defective vaccinal efficacy. Twenty-four cases were clinically identified. A cucumber salad was identified as the contaminating dish (Risk Ratio = 3.6; 95%CI 1.5-8.9). Only one factor was related to defe

Development of a novel real-time PCR assay to detect intact astA-harboring Escherichia coli in food based on complete DNA sequence of astA gene variants.

Enteroaggregative Escherichia coli (EAEC) may produce an enterotoxin termed EAEC heat-stable enterotoxin 1 (EAST1), which is encoded by the astA gene. E. coli strains harboring astA but without virulence markers specific for other diarrheagenic E. coli pathotypes (EAST1EC), have been isolated from patients with diarrhea in foodborne outbreaks in Japan. Various nucleotide variants of astA have been reported, some of which are incomplete and non-functional. However, the astA variants carried by strains isolated from foodborne outbreaks have not yet been fully characterized. Therefore, we identif

Prevalence, antimicrobial resistance, and genetic characterization of Listeria monocytogenes in the Korean pork production chain.

Listeria monocytogenes, a major foodborne pathogen, causes invasive listeriosis with a high mortality rate. Its distribution and genomic diversity in livestock products in Korea remain poorly understood. This study investigated the prevalence and genomic characteristics of L. monocytogenes across Korean pork production and distribution chains. Samples from pigs, carcasses, meat, and the environment were collected from farms, slaughterhouses, and supermarkets nationwide. L. monocytogenes was recovered from 60 retail meat samples (19.0 %) but not from farms or slaughterhouses, suggesting that co

Portable detection of Salmonella in food of animal origin via Cas12a-RAA combined with an LFS/PGM dual-signaling readout biosensor.

A highly specific and sensitive rapid two-signal assay was developed for the detection of Salmonella typhimurium in foods of animal origin. The invA gene of Salmonella was used as the biorecognition element and recombinase-assisted amplification (RAA) technology for signal amplification. By utilizing the specific recognition and efficient trans-cleavage activity of CRISPR/Cas12a, point-of-care testing (POCT) for S. typhimurium was achieved via lateral flow strips (LFS) and personal glucometer (PGM) biosensors as dual signal readout systems, with sensitivities of 33 CFU/mL and 20 CFU/mL, respec
