Pentagastrin infusions in patients with panic disorder. II. Neuroendocrinology.

Cholecystokinin (CCK) has well-documented anxiogenic effects in animals and normal people, and panicogenic effects in patients with panic disorder, but little is known about its neuroendocrine profile. We examined neuroendocrine responses to intravenous infusions of pentagastrin, a selective CCK-B receptor agonist, in 10 patients with panic disorder and 10 normal control subjects. Pentagastrin potently activated the hypothalamic-pituitary-adrenal (HPA) axis, but did not release growth hormone or any of several vasoactive peptides (neurokinin A, substance P, vasoactive intestinal peptide). The

An evaluation of basal hypothalamic-pituitary-thyroid axis function in depression: results of a large-scaled and controlled study.

In order to evaluate the function of the hypothalamic-pituitary-thyroid (HPT)-axis in unipolar depression, the authors measured basal 0800h plasma levels of free thyroxine (FT4), free triiodothyronine (FT3), and thyroid stimulating hormone (TSH) by means of the new, ultrasensitive assays (TSH-IRMA) in 69 healthy controls, 62 minor, 101 simple major, and 57 melancholic depressed subjects. Basal HPT-axis hormone levels of almost all (96.8%) unipolar depressed patients fell within the normal, euthyroid range. None of the major depressed subjects showed subclinical hypothyroidism. It was found tha

Beta-endorphin, ACTH, and catecholamine responses in chronic autonomic failure.

Plasma epinephrine (EPI), norepinephrine (NE), beta-endorphin, and corticotropin (ACTH) responses were measured during insulin-induced hypoglycemia in normal subjects and in patients with either multiple system atrophy (MSA) or idiopathic orthostatic hypotension (IOH). In normal subjects, there was a striking rise in EPI, NE, beta-endorphin, and ACTH following the nadir of hypoglycemia. Both beta-endorphin and ACTH responses were significantly lower than normal in patients with MSA, in contrast to normal levels in IOH patients. No correlation was observed between the degree of adrenergic insuf

Drug abuse and reproduction.

It is clear that a number of CNS agents, including drugs of abuse, can inhibit reproductive function. Figure 1 shows the chemical diversity of some of the drug groups that affect reproductive hormones. Their structural dissimilarity to the steroid hormones is also readily apparent in the figure. These chemically diverse drugs share an important pharmacologic property: they are highly potent neuroactive drugs, and they can disrupt hypothalamic-pituitary function. Although it is frequently difficult to distinguish between direct drug actions on the hypothalamic-pituitary axis and subsequent effe

The CRFs and their control: chemistry, physiology and clinical implications.

The 41-amino acid CRF fulfils all the criteria for a corticotrophin releasing factor, although considerable evidence suggests that other factors, particularly VP, also play a physiologically significant role in controlling ACTH release. Although human CRF has now been identified as a 41-residue peptide, most studies to date have used oCRF-41 in their exploration of the physiology and pathology of the hypothalamic--pituitary--adrenal axis. Low doses of oCRF-41 appear to be safe, and for specific tests of the readily-releasable pool of ACTH and related peptides 100 micrograms is a practical dose

Endocrine dysfunction in anorexia nervosa and bulimia: comparison with abnormalities in other psychiatric disorders and disturbances due to metabolic factors.

The eating disorders of anorexia nervosa and bulimia are associated with marked disturbances in endocrine function. Studies of the hypothalamic-pituitary-adrenal,-thyroid, and -ovarian axes are reviewed, in order to ascertain whether and to what extent alteration in endocrine response is the consequence of abnormal eating, or whether the endocrine abnormalities are primary. Many, if not all, of the disturbances which have been documented can be accounted for the metabolic consequences of disturbed eating behavior, including especially the effects of weight loss. However, it is possible that pr

Hypothalamic sarcoidosis and hypopituitarism.

4 patients with presumed pituitary hypothalamic sarcoidosis are described. 3 had histological diagnoses compatible with sarcoidosis and in the other this diagnosis was strongly suspected from chest X-rays. 2 patients presented with diabetes insipidus. ACTH reserve was diminished in 3 out of 4 and growth hormone reserve was diminished in the 3 who were tested. All 4 patients developed secondary amenorrhea. 3 patients had hypothalamic hypothyroidism. Prolactin dynamics were intact. Tomograms of the sella turcica in all 4 and computerized tomography of the hypothalamic area in 2 patients failed t

Ultrastructural changes of the hypothalamo-hypophysial neurosecretory system in the magnesium deficient rats.

A magnesium deficient diet caused transient but marked degenerative changes in the rat hypothalamo-hypophysial neurosecretory system which strongly resembled in many ultrastructural respects those induced by a prolonged administration of aldosterone as previously reported by us. The possible mechanism for this selective alteration in the neurosecretory neurons has been briefly discussed with regard to aldosterone secretion.

Mediating role of maternal cortisol in the association between maternal serum creatinine and fetal cortisol exposure.

BACKGROUND: Cortisol is the principal glucocorticoid regulated by the hypothalamic-pituitary-adrenal (HPA) axis and plays a crucial role in maintaining pregnancy and promoting fetal growth and development. Renal function is a key determinant of cortisol exposure. Given that serum creatinine serves as a key biomarker of renal function, it is plausible that maternal creatinine levels contribute to the regulation of fetal cortisol exposure, although the underlying mechanisms remain to be fully elucidated. OBJECTIVE: This study was designed to (1) examine the association between maternal serum cr

Intermittent Fasting Exerts Antidepressant-Like Effects by Restoring HPA Axis Homeostasis Through Coordinated GR-MR Signaling.

Fasting, as a non-pharmacological intervention, elicits antidepressant-like effects. However, its neural and molecular mechanisms remain unclear. This study investigated whether intermittent fasting (IF) alleviates corticosterone (CORT)-induced depressive-like behaviors in mice and explored the underlying mechanisms. This study demonstrated that IF reversed the CORT-induced reduction in hypothalamic glucocorticoid and mineralocorticoid receptor (GR/MR) expression in mice. In addition, IF reversed a CORT-induced decrease in arcuate nucleus (ARC) AgRP/NPY neuronal activity and NPY output from th

[Therapeutic mechanism of combination of Epimedii Folium and Ligustri Lucidi Fructus for glucocorticoid synthesis-restricted asthma in rats].

This study aims to elucidate the therapeutic effects and mechanisms of the combination of Epimedii Folium and Ligustri Lucidi Fructus on glucocorticoid(GC) synthesis-restricted asthma in rats. The classic asthma model induced by ovalbumin(OVA) sensitization and challenge, a complex asthma animal model with specific suppression of hypothalamic-pituitary-adrenal(HPA) axis at the synthesis level of endogenous GC by intraperitoneal administration of metyrapone(Met), was established. This model was designed to simulate the pathological condition of GC resistance or insufficient synthesis encountere

Associations of adverse childhood experiences, inflammation, and cognition in older Black adults.

BACKGROUND: Childhood adversity has profound implications for cognitive health, with particularly pronounced impacts in minoritized populations. We apply an intersectionality framework to explore how distinct overlapping patterns of adverse childhood experiences (ACEs) influence cognition and cognitive status in older Black adults, considering the potential mediating role of inflammatory pathways. METHODS: Participants included 536 individuals without dementia from the Minority Aging Research Study. Cognition was assessed using episodic memory, semantic memory, and perceptual speed tasks. 145

Hypothalamic RASA1/Ras/AKT/GnRH axis reprogramming mediates GLP-1RA's central rescue of PCOS HPG dysfunction.

AIM: Polycystic ovary syndrome (PCOS), characterised by hyperandrogenaemia and hypothalamic-pituitary-gonadal (HPG) axis dysregulation driven by gonadotropin-releasing hormone (GnRH) neuronal hyperactivity, remains mechanistically enigmatic, with limited targeted therapies. Although glucagon-like peptide-1 receptor agonists (GLP-1RAs) show potential in alleviating PCOS reproductive dysfunction, their presumed mechanisms have been confined to peripheral metabolic modulation. MATERIALS AND METHODS: Using liraglutide as a representative GLP-1RA, this study elucidated the central control mechanis

Sex-dependent effects of FGF21 on HPA axis regulation and adrenal regeneration after Cushing syndrome in mice.

BACKGROUND: Cushing's syndrome (CS) results from prolonged exposure to excessive glucocorticoids (GCs), leading to metabolic disturbances and adrenal insufficiency (AI). Fibroblast growth factor 21 (FGF21) has shown promise as a potential therapeutic target for metabolic disorders. This study explores the effects of FGF21 on adrenal gland function in a mouse model of AI following chronic hypercortisolism and investigates sex-dependent differences in the hypothalamic-pituitary-adrenal (HPA) axis response. METHODS: We employed a mouse model of AI after chronic corticosterone (CORT) treatment. T

Depression Exacerbates Hepatic Steatosis in C57BL/6J Mice by Activating the Hypothalamic-Pituitary-Adrenal Axis.

BACKGROUND/AIM: Depression is associated with metabolic disorders, including non-alcoholic fatty liver disease (NAFLD). However, the mechanisms underlying the interaction between them are still poorly known. MATERIALS AND METHODS: In this study, mice on a choline deficiency, L-amino acid-defined, high-fat diet (CDAHFD) developing steatosis were challenged with chronic restraint stress (CRS), a protocol widely used to induce depression. The development of depression and steatosis was evaluated using histopathology analysis, ELISA, q-PCR and Western Blot. RESULTS: The contribution of the activ

Echoes of conflict and displacement in maternal health: Life-course violence, timing, and maternal stress after childbirth at the northern Thailand-Myanmar border.

Armed conflict, displacement, and related violence is escalating globally, concentrated among civilians and migrants in border areas, and poses grave harms to women and children. The current study investigates how women's life-course experiences of conflict and displacement are linked to maternal stress and health outcomes after childbirth at the Thailand-Myanmar border, specifically stress, mental health, and cardiometabolic outcomes. Analyses are based on a cross-sectional population-based maternal and child health survey of 701 mothers, collected in 2017-18 in northern Thailand along the My
