Applying Opal multiplex immunofluorescence (OMI) to characterize intestinal tissues of genetically engineered mouse models provides an excellent tool for studying complex processes. However, detecting appropriate signals from multiple target molecules is challenging. Here, we present a protocol to characterize mouse intestinal epithelial cell lineage using OMI. We describe steps for processing small intestine and colonic mouse tissues and designing and optimizing panels for OMI in mouse intestinal tissues. We then detail procedures for performing a quantitative evaluation of acquired images. F

In regenerative medicine, cells, tissues and organs are often replaced, engineered or regrown in order to restore their function after they have been damaged or lost. Local anesthetics, corticosteroids and contrast agents are commonly employed for both diagnostic and therapeutic objectives in interventional pain and musculoskeletal treatments for regenerative medicine. There is growing evidence that routine injectables promote catabolism and disease processes. Thus, understanding the effects of these compounds on regenerative medicine injectates and target tissues such as tenocytes, chondrocyt

Trends in regenerative medicine are transforming the field by moving beyond simple repair to actively reprogram cellular and tissue environments. This review explores new approaches that integrate nanotechnology, metabolic regulation, and immune engineering to improve the regeneration of tissues that are otherwise difficult to regenerate. Advances in nanocarrier designs, such as lipid nanoparticles, ROS-sensitive polymers, hydrogels, and extracellular vesicles, have enabled the rapid, responsive, and precise delivery of therapeutic agents to targeted areas, including fibrosis, chronic inflamma

Two cases of calcified bone and dental tissue integration with titanium implants are presented, along with a review of the literature on the … ocumented natural formation of periodontal tissues around dental implants provided a foundation for tissue engineering studies aimed at realizing implant-bone relationships similar to those of natural bone-tooth structures. The primary challenges remain the long-term preservation of periodontal-like tissue formed on implants and the imparting of functional proprioce …

Pediatric patients with a neurogenic urinary bladder, caused by developmental abnormalities including spina bifida, exhibit chronic urological problems. Surgical management in the form of enterocystoplasty is used to enlarge the bladder, but is associated with significant clinical complications. Thus, alternative methods to enterocystoplasty have been explored through the incorporation of stem cells with tissue engineering strategies. Within the context of this review, we will examine the use of bone marrow stem cells and induced pluripotent stem cells (iPSCs), as they relate to bladder regene…

Osteochondral defect is a complex tissue loss disease caused by arthritis, high-energy trauma, and many other reasons. Due to the unique structural characteristics of osteochondral tissue, the repair process is sophisticated and involves the regeneration of both hyaline cartilage and subchondral bone. However, the current clinical treatments often fall short of achieving the desired outcomes. Tissue engineering bioscaffolds, especially those created via three-dimensional (3D) printing, offer promising solutions for osteochondral defects due to their precisely controllable 3D structures. The mi…

The evolution of regenerative medicine has repositioned the extracellular matrix (ECM) from a passive structural scaffold to a dynamic signaling hub that dictates host immunity and tissue remodeling. A critical driver of this bioactivity is the matrix-bound nanovesicle (MBV), a distinct subclass of extracellular vesicles (EVs) physically embedded within collagen fibers. Unlike fluid-phase EVs, MBVs exhibit unique release kinetics triggered by matrix degradation and possess tissue-specific molecular signatures that dictate their therapeutic potential. This review evaluates the biogenesis, isola

Plant roots are extremely heterogeneous and non-photosynthetic tissue that is drastically different from leaves. Pioneering studies have revealed the existence of pattern-triggered immune responses in roots; however, whether roots mount tissue-specific effector-triggered immunity (ETI) is a fundamental question in plant immunity. By utilizing inducible effector-expressing lines, we systematically characterize non-conventional physiological and molecular responses during ETI activation in roots. Root ETI seems to be weaker than leaf ETI, evidenced by restricted cell death in the transition zone

Irregular endometrial damage often evolves into chronic conditions, such as endometrial thinning, functional tissue fibrosis, and intrauterine adhesions. These conditions pose significant challenges to female reproductive health. Addressing this, we developed a dynamic double-cross-linked hydrogel for the repair of fragile and dynamic endometrial tissue. This hydrogel integrates natural bioactive components (chitosan, protocatechuic aldehyde, and Fe(III)) and serves as a scaffold for Mn single-atom nanozymes (Mn SAN). Through dynamic covalent and hydrogen bonding, the hydrogel exhibits excepti

Periodontitis is a severe gum infection characterized by inflammation of the tissues surrounding the teeth. The disease is challenging to manage due to its exposure to a wet and dynamic oral environment, where conventional hydrogels often suffer from weak adhesion, short residence time, and vulnerability to bacterial invasion. In this study, an innovative hydrogel system based on in situ light curing is proposed. The hydrogel precursor, comprising sodium alginate and a calcium ion network, is designed and adhere to the irregular and smooth surfaces of periodontal tissue before curing. Upon lig

Myocardial infarction (MI) is a major cause of morbidity and mortality worldwide, especially in aging and metabolically unhealthy populations. A major target of regenerative tissue engineering is the restoration of viable cardiomyocytes to preserve cardiac function and circumvent the progression to heart failure post-MI. While bone tissue is known for its inherent regenerative abilities, various pathological conditions and trauma can disrupt its meticulously regulated processes of bone formation and resorption. Bone tissue engineering aims to replicate the extracellular matrix of bone tissue as well as the sophisticated biochemical mechanisms crucial for effective regeneration. Traditionally, the field has relied on external agents like growth factors and pharmaceuticals to modulate these processes. Although efficacious in certain scenarios, this strategy is compromised by limitations such as safety issues and…

Advances in the engineering of nanoparticles (NPs), which represent particles of less than 100 nm in one external dimension, led to an increasing utilization of nanomaterials for biomedical purposes. A prerequisite for their use in diagnostic and therapeutic applications, however, is the targeted delivery to the site of injury. Interactions between blood-borne NPs and the vascular endothelium represent a critical step for nanoparticle delivery into diseased tissue. Here, we show that the endothelial glycocalyx, which constitutes a glycoprotein-polysaccharide meshwork coating the luminal surfac
