The effect of acetylcholine (Ach) on smooth-muscle strips isolated along the transversal axis of cat lower esophageal sphincter (LES) is studied. Ach in low concentrations (10(-11)--10(-9) g/ml) causes contraction of the muscle strips. Increase of the concentration to 10(-8) g/ml leads to biphasic effect: contraction with relaxation. Inhibitory response predominates at Ach 10(-6) and 10(-5) g/ml. Atropine (10(-6) M) eliminates the excitatory phase but it has no effect on the second relaxation phase. Propranolol (10(-6), 2 X 10(-6) M) as well as phentolamine turn the inhibitory response to Ach …

1. The effects of acetylcholine, noradrenaline and pentagastrin on the action potential of canine antral circular muscle were determined using the intracellular micro-electrode technique. 2. Acetylcholine increased the amplitude and duration of the plateau potential of the action potential. Since these effects were blocked by atropine but not by hexamethonium, the effects of acetylcholine were on muscarinic receptors, probably located on the smooth muscle cell. 3. Pentagastrin 2 x 10(-10) M increased the size of the plateau potential and the frequency of the action potential; pentagastrin 1 x …

A technique is described which has enabled us to measure change in 22Na+ efflux from smooth muscle plasma membrane vesicles. The resting 22Na+ efflux from these sealed vesicles showed a concentration-dependent increase in response to acetylcholine and other muscarinic agonists, in similar concentrations to those which increased 42K+ efflux in whole muscle. The kinetics of this efflux were complex and could not be described by less than three exponential processes. The response to agonists has, therefore, been characterized by measurement of the half-life of 22Na+ efflux (t1/2). The acetylcholi…

Cerebral regional acetylcholine (ACh) levels and utilization were studied in vivo in thiamin deficient (TD), pair-fed asymptomatic (PFC) and ad libitum fed control (ALC) rats. ACh levels in the cortex, corpus striatum, midbrain, diencephalon and brainstem of TD rats were comparable to those observed in the control groups. However, ACh utilization was slightly to moderately (10-41%) decreased in cortex, midbrain, diencephalon and brainstem. The decrease was significantly different in the midbrain of TD rats as compared to PFC and ALC rats.

The direct effects of acetylcholine (ACh) on Na+- or Ca2+-dependent action potentials of curarized sympathetic ganglion cells in bullfrogs were investigated under a condition where membrane depolarization caused by the muscarinic action of ACh was nullified by means of a hyperpolarizing current. ACh decreased the after-hyperpolarization of Na+-action potentials in Ringer's solution, and increased the after-depolarization of Ca2+-action potentials in the isotonic Ca2+ solution. In both solutions, the maximum rates of rise of the spikes were decreased and the slope membrane resistance at the ori…

The in vitro frog spinal cord has been used to study acetylcholine (ACh) release and spinal root potentials. The preparation bathed in an eserine-containing medium spontaneously released ACh into the bathing fluid. This release was enhanced by atropine in a dose-related and long-lasting manner and transiently by oxotremorine. The release rate of ACh was increased by low frequency ventral root stimulation; this increase was found to be proportionally the same after application of atropine. Oxotremorine did not modify the elctrically-evoked ACh release but blocked or reduced the effect of atropi…

Nitric oxide (NO), stimulated by the muscarinic receptor agonist acetylcholine (ACh) released from the endothelium via mitochondrial acetyl-CoA production, appears to contribute to physiological flow-mediated vasodilation. Diphenhydramine (DPH), a muscarinic receptor antagonist, may interfere with ACh-induced NO-mediated vasodilation. However, whether DPH affects ACh-induced, NO-mediated vasodilation remains unknown. This study aimed to examine the effect of DPH on ACh-induced, NO-mediated vasodilation and its underlying mechanisms, including muscarinic receptor subtypes. The effects of diphen…

The α9α10 nicotinic acetylcholine receptor (nAChRs) is a potential therapeutic target for several diseases, including inner ear disorders, chronic pain, cancer, pemphigus vulgaris, and inflammatory disorders. However, the influence of key sites that modulate the receptor's pharmacological properties remains unclear. The crystal and cryo-electron microscopy structures of various nAChRs have helped elucidate functionally important residues. The X-ray crystal structure of the homomeric α9 subunit extracellular domain strongly implicates Thr147, unique to α9 and α10 subunits, and the invariant Arg…

BACKGROUND: The prevalence of cancer poses significant challenges to treatment, largely because of drug resistance along with other side effects. Current studies have been investigating the growth factors more than other biologic tumor features, such as neurobiologic features. Here in this review, we highlight the role of nicotinic acetylcholine receptors (nAChRs) in cancer development with their context-dependent activation and downstream effectors.
RECENT FINDINGS: Some nAChR subtypes stimulate tumorigenic pathways, EGFR/ERK1/2, PI3K/AKT, and MAPK, with varying responses based on the recept…

BACKGROUND: The prognostic significance of the Coronary Vasomotor Disorders International Study Group (COVADIS) criteria during acetylcholine (ACh) provocation testing is uncertain.
AIMS: The aim of this study was to assess the prognostic impact of COVADIS criteria in patients with myocardial ischaemia (INOCA) or myocardial infarction (MINOCA) and non-obstructive coronary arteries undergoing ACh provocation testing.
METHODS: We enrolled consecutive INOCA and MINOCA patients undergoing ACh provocation testing. The occurrence of each COVADIS criterion was recorded. The primary outcome was the …
