Vitamin A2-based photopigments within the pineal gland of a fully terrestrial vertebrate.

Fully terrestrial vertebrates were previously thought to exclusively employ vitamin A1 to generate visual pigments. However, recent studies on the visual system of the lizard Anolis carolinensis have shown that its visual pigments are vitamin A2-based. This unexpected result prompted an investigation of the pineal photopigments in this species [13]. HPLC analysis has shown that this extraretinal photoreceptor also exclusively utilizes a vitamin A2-derived chromophore. The adaptive significance of this chromophore within the pineal is unclear. The extended long wavelength sensitivity characteri

Flatpea intoxication in sheep and indications of ruminal adaptation.

This paper describes the signs of toxicity when seed-bearing flatpea (Lathyrus sylvestris L) hay is fed to sheep. Signs of intoxication (including seizure, muscular trembling and spasmotic torticollis) are similar to those observed for ammonia toxicity in ruminants. Accumulation of ammonia may be a direct consequence of flatpea ingestion, given that 2,4-diaminobutyric acid (DABA, a toxic constituent of flatpea) is known to inhibit hepatic urea synthesis. However, other modes of toxicity for DABA as well as other flatpea toxins may also contribute to this process of intoxication. Our evidence s

The adaptation of Plasmodium falciparum to oxidative stress in G6PD deficient human erythrocytes.

It has recently been found that growth of P. falciparum in human G6PD deficient red cells is impaired in vitro; however, the inhibition is overcome after two or three growth cycles. There is evidence to suggest that the parasite can produce its own G6PD enzyme which may compensate for the lack of host enzyme and could account for the resumption of normal growth in G6PD deficient host cells. It is unclear whether the parasite enzyme can enable the host cell to resist oxidative stress as normal cells do. To answer this question, P. falciparum was grown in vitro in: (a) normal red cells, (b) G6PD

Metabolic adaptation of rat faecal microflora to cyclamate in vitro.

Previous studies have demonstrated that the rat faecal microflora maintained in vitro under conditions of continuous flow possesses bacteriological and metabolic characteristics similar to those of the native bacterial population of the caecum. Addition of sodium cyclamate (75 mM) to the culture concurrent with the progressive dilution of the growth medium promoted metabolism of cyclamate to cyclohexylamine (sulphamatase activity) within 4 wk. The maximum formation of cyclohexylamine was attained in about 8 wk and was equivalent to a 2-3% molar conversion of cyclamate to cyclohexylamine. The r

Duodenal and ileal adaptation to dietary calcium restriction: in vivo studies in the rat.

Intestinal adaptation by the growing rat to a low-calcium diet was studied by in situ perfusion of duodenum and ileum in vivo. Rats were fed diets containing either 1.2 or 0.02% Ca for 17-24 days. To study plasma-to-lumen flux and net calcium absorption, rats were loaded parenterally with 45Ca and perfused intraluminally with 3.4 mM calcium. Calcium restriction caused net absorption in ileum to increase fourfold, in duodenum almost twofold. With calcium restriction, plasma-to-lumen flux decreased in duodenum but not in ileum and was small relative to absorption. However, net calcium secretion,

Application of a Biologically Contained Reporter System To Study Gain-of-Function H5N1 Influenza A Viruses with Pandemic Potential.

Natural adaptation of an antigenically novel avian influenza A virus (IAV) to be transmitted efficiently in humans has the potential to trigger a devastating pandemic. Understanding viral genetic determinants underlying adaptation is therefore critical for pandemic preparedness, as the knowledge gained enhances surveillance and eradication efforts, prepandemic vaccine design, and efficacy assessment of antivirals. However, this work has risks, as making gain-of-function substitutions in fully infectious IAVs may create a pathogen with pandemic potential. Thus, such experiments must be tightly

Behavioral changes in host foraging: Experiments with Clinostomum (Trematoda, Digenea) parasitizing Loricariichthys platymetopon (Loricariidae).

Complex life cycle parasites can manipulate the behavior of intermediate hosts in order to reduce their fitness and increase the chance of completing life cycle. In order to understand the effects of the trematode parasites of the genus Clinostomum on host fish Loricariichthys platymetopon, a filmed experiment was carried out to quantify the foraging activity of hosts with different intensities of infection. The results suggest that hosts with higher parasite intensities reduced foraging activity early in the morning when compared to hosts with low intensities. This period may be critical for

Evolutionary pathways to antibiotic resistance are dependent upon environmental structure and bacterial lifestyle.

Bacterial populations vary in their stress tolerance and population structure depending upon whether growth occurs in well-mixed or structured environments. We hypothesized that evolution in biofilms would generate greater genetic diversity than well-mixed environments and lead to different pathways of antibiotic resistance. We used experimental evolution and whole genome sequencing to test how the biofilm lifestyle influenced the rate, genetic mechanisms, and pleiotropic effects of resistance to ciprofloxacin in Acinetobacter baumannii populations. Both evolutionary dynamics and the identitie

Ecogeographic variation across morphofunctional units of the human nose.

OBJECTIVES: Although the internal nose is overwhelmingly responsible for heat and moisture exchange during respiration, external nasal morphology is more commonly cited as evincing climatic adaptation in humans. Here, we assess variation across all four morphofunctional units of the complete nasorespiratory tract (external pyramid, nasal aperture, internal nasal fossa, and nasopharynx) to determine which units provide the strongest evidence of climatic adaptation. MATERIALS AND METHODS: We employ 20 linear measurements collected on 837 modern human crania from major geographic (Arctic Circle,

Placental adaptations in growth restriction.

The placenta is the primary interface between the fetus and mother and plays an important role in maintaining fetal development and growth by facilitating the transfer of substrates and participating in modulating the maternal immune response to prevent immunological rejection of the conceptus. The major substrates required for fetal growth include oxygen, glucose, amino acids and fatty acids, and their transport processes depend on morphological characteristics of the placenta, such as placental size, morphology, blood flow and vascularity. Other factors including insulin-like growth factors,

Combined support for wholesale taxic atavism in gavialine crocodylians.

Morphological and molecular data sets favor robustly supported, contradictory interpretations of crocodylian phylogeny. A longstanding perception in the field of systematics is that such significantly conflicting data sets should be analyzed separately. Here we utilize a combined approach, simultaneous analyses of all relevant character data, to summarize common support and to reconcile discrepancies among data sets. By conjoining rather than separating incongruent classes of data, secondary phylogenetic signals emerge from both molecular and morphological character sets and provide solid evid

Tests of the adaptive modulation hypothesis for dietary control of intestinal nutrient transport.

According to the adaptive modulation hypothesis, an intestinal transporter should be repressed when its biosynthetic and other costs (of maintenance) exceed the benefits it provides. This leads to two contrasting predictions: transport of a sugar or amino acid worth calories should tend to be increased by its substrate, and transport of a vitamin should be modulated downwards by its substrate and upmodulated in deficiency. In a test of the first prediction, omnivorous desert iguanas eating alfalfa pellets (a high-carbohydrate diet) were compared with desert iguanas eating mealworms (a low-carb

State-matched redox nanomedicine for cell fate control and multicompartment therapeutic windows.

Tumors persist under sustained redox stress by coupling increased reactive-species production to antioxidant buffering, metabolic plasticity and multicellular support. This creates therapeutic vulnerabilities whose tractability depends on the biological state of the exposed compartment. Here, we frame redox nanomedicine as state-matched manipulation of oxidant flux, antioxidant buffering, oxygen and substrate availability, localization, duration and spatial coverage. We propose that therapeutic outcome emerges from chemistry, exposure and the presence of an executable redox dependency together

A harmonized framework for studying exceptional cognitive aging in Latin America.

INTRODUCTION: Biological, genetic, and sociocultural factors underlying cognitive resilience and resistance to pathology remain unexplored in Latin America. The Life-Course Epidemiology of Aging, Resilience, and Neurodegeneration in Alzheimer's Disease (LEARN-AD) study and Latin American SuperAgers Study (LASAS) establish the first harmonized frameworks to characterize resilience and exceptional cognitive aging phenotypes in the region. METHODS: The shared protocol integrates cognitive, neurological, psychosocial, and genetic data. SuperAgers are deeply phenotyped, while successful cognitive

An integrated analytical method for the separation, identification and quantification of carotenoids in Rhodobacter capsulatus.

The analysis and quantification of microbial carotenoids remain challenging due to the complexity of biological matrices and the high cost or limited availability of commercial standards. Herein, we present a cost-effective and reproducible workflow for the extraction, isolation, identification, and quantification of carotenoids in Rhodobacter capsulatus cultures. The method integrates light-protected pigment extraction, screening by thin-layer chromatography, and column chromatography for the isolation of individual carotenoids. Structural assignment is supported by UV-Vis spectroscopy, nucle

A modified Ising model of Barabási-Albert network with gene-type spins.

The central question of systems biology is to understand how individual components of a biological system such as genes or proteins cooperate in emerging phenotypes resulting in the evolution of diseases. As living cells are open systems in quasi-steady state type equilibrium in continuous exchange with their environment, computational techniques that have been successfully applied in statistical thermodynamics to describe phase transitions may provide new insights to the emerging behavior of biological systems. Here we systematically evaluate the translation of computational techniques from s

Single-threshold-guided adaptive cancer therapy with partial-cycle treatment: A mechanistic and reinforcement learning analysis.

Adaptive cancer therapy seeks to modulate aggressive treatment to preserve drug-sensitive tumor cells that suppress resistant populations, but existing strategies often rely on frequent treatment decisions enabled by intensive surveillance, limiting clinical feasibility. Here, we propose a clinically motivated alternative that shortens the treatment window within a fixed and relatively long surveillance cycle, thereby avoiding the need for frequent monitoring. Based on this idea, we develop a mechanistic modeling framework for single-threshold-guided adaptive therapy with partial surveillance-

Beyond species-level differences: A comprehensive review of intraspecific venom plasticity in spiders.

Spider venoms represent a diverse array of biological cocktails, crucial for predator-prey dynamics and ecological interactions. While differences in venom composition between species (interspecific variation) are well-characterized, the extent and drivers of intraspecific venom plasticity remain a critical, underexplored area. This comprehensive review synthesizes current research on venom variation within species, emphasizing the multifaceted factors that govern this dynamic phenotype. Venom composition, yield, and bioactivity are significantly influenced by both endogenous and exogenous ele

A heavyweight early whale pushes the boundaries of vertebrate morphology.

The fossil record of cetaceans documents how terrestrial animals acquired extreme adaptations and transitioned to a fully aquatic lifestyle1,2. In whales, this is associated with a substantial increase in maximum body size. Although an elongate body was acquired early in cetacean evolution3, the maximum body mass of baleen whales reflects a recent diversification that culminated in the blue whale4. More generally, hitherto known gigantism among aquatic tetrapods evolved within pelagic, active swimmers. Here we describe Perucetus colossus-a basilosaurid whale from the middle Eocene epoch of Per

Retortamonads from vertebrate hosts share features of anaerobic metabolism and pre-adaptations to parasitism with diplomonads.

Although the mitochondria of extant eukaryotes share a single origin, functionally these organelles diversified to a great extent, reflecting lifestyles of the organisms that host them. In anaerobic protists of the group Metamonada, mitochondria are present in reduced forms (also termed hydrogenosomes or mitosomes) and a complete loss of mitochondrion in Monocercomonoides exilis (Metamonada:Preaxostyla) has also been reported. Within metamonads, retortamonads from the gastrointestinal tract of vertebrates form a sister group to parasitic diplomonads (e.g. Giardia and Spironucleus) and have als
