Comparison of three methods for enamel protein extraction in different developmental phases of rat lower incisors.

Protein extraction methods [urea, trichloroacetic acid (TCA), and acetic acid] were compared for protein recovery from rat incisor developing enamel in the S phase (intermediate/late secretion), M1 phase (early maturation), M2 phase (intermediate maturation), and M3 phase (final maturation). We compared the protein recoveries with the percentage of enamel matrix dry weight burnt off by incineration. Our results indicate that TCA and urea were equally efficient for the extraction of S-stage proteins (85% and 90% recovery, respectively), while urea was the best for M1-stage proteins (92% recover

In vitro development of lower first molars from the osteopetrotic microphthalmic (mi) mouse.

While the precise mechanism of tooth eruption remains unknown it has been established that for tooth eruption to occur a functioning dental follicle is essential and normal bone resorption is required. In the osteopetrotic microphthalmic (mi) mouse, teeth fail to erupt and development is affected. In the present study, an organ culture technique was established to culture successfully lower first molars from normal and affected mi mice so as to determine if developmental defects were intrinsic to the dental tissue or whether such defects were a result of local environmental factors. Tissue was

Dental enamel in relation to ionized calcium and parathyroid hormone. Studies of human primary teeth and rat maxillary incisors.

The aims of this thesis were to evaluate the role of lowered calcium values in blood on enamel formation and mineralization, and further to analyze whether the calcium regulating parathyroid hormone (PTH) would have any effects on the forming and mineralizing enamel. DENTAL ENAMEL AND IONIZED CALCIUM: Human primary teeth from two groups of infants were clinically and histologically examined. One group (n = 25) was born with optimal perinatal conditions. The infants had low, but not hypocalcemic values on day 1 when compared with days 3 and 5 postpartum. The second group of infants (n = 11) was

Development defects of enamel in humans with hereditary epidermolysis bullosa.

Developmental defects of enamel are often reported as a feature of the more severe forms of epidermolysis bullosa (EB). The purpose of this investigation was to determine the prevalence and character of enamel defects in each of the major hereditary EB types. Clinical evaluations were made on 237 individuals representing all of the major EB types and 58 unaffected individuals. All EB cases were categorized by phenotype, mode of inheritance and skin biopsy. The frequency of individuals having developmental enamel defects ranged from 8.6% in recessive dystrophic EB to 100% in junctional EB; 27.5

Prevalence of dental anomalies on panoramic radiographs in a population of the state of Pará, Brazil.

INTRODUCTION: Dental anomalies (DAs) are the result of disorders that are able to modify the shape, number, size, and structure of teeth. This study aimed to evaluate the prevalence of DAs using panoramic radiographs in a population of the City of Belém, northern Brazil. MATERIALS AND METHODS: In this study, 487 panoramic radiographs were evaluated searching for DAs. Dental records were reviewed for diagnostic confirmation. DAs related to the shape, number, size, and structure of teeth were investigated. RESULTS: Our results showed a DA prevalence of 56.9%. The most prevalent DA was taurodon

Analysis of MMP-8, MMP-20, and TGF-β1 in molar-incisor hypomineralization and assessment of periodontal health.

BACKGROUND: Gingival crevicular fluid (GCF) volume can be used to evaluate periodontal diseases, along with matrix metalloproteinase-8 (MMP-8) and transforming growth factor-beta 1 (TGF-β1) found in GCF. Conversely, matrix metalloproteinase-20 (MMP-20) is implicated in amelogenesis and serves as a potential biomarker for assessing enamel mineralization. In this context, the present study aims to investigate the effect of molar incisor hypomineralization (MIH) on periodontal health and to evaluate the relationship between MIH and GCF volume, as well as GCF biomarkers such as MMP-8, MMP-20, and …

Heimler syndrome with a complaint of blurred vision caused by compound heterozygous variants in PEX1.

INTRODUCTION: Heimler syndrome (HS) is a rare disorder that includes sensorineural hearing loss (SNHL), nail abnormalities, and enamel hypoplasia. Patients with this syndrome can also exhibit ocular manifestations. At present, only a few cases of HS have been reported, existing knowledge of this syndrome is limited, and many cases have been misdiagnosed or even missed. This is the first report of Heimler syndrome with blurred vision as the first complaint, which was diagnosed by genetic analysis in the ophthalmology department. CASE DESCRIPTION: An 8-year-old girl complained of bilateral visu

Proteolysis on maturing enamel surface, as shown by gel-coating methods.

Degradation of enamel matrix proteins, and their removal during early maturation, is critical for the growth of large enamel crystals in the subsequent processes of enamel maturation. In this study, we sought to demonstrate, using in vivo zymography, the exact sites of proteolysis in maturing enamel and its relationship to the overlying ameloblasts. The maturing enamel surfaces of rat and bovine incisors were exposed and painted either with pre-exposed autoradiographic emulsion or with densely fluorescein-conjugated (DQ) gelatin. After a few hours, photographic development of the emulsion reve

Dental variables associated with differences in severity of fluorosis within the permanent dentition.

The aim of this study was to assess the influence of age at start, duration, and completion of enamel formation, as well as of tooth-eruption age and enamel thickness on the severity of dental fluorosis within the permanent dentition. The material comprised Ugandan children (n = 219), aged 10-14 years, with 28 teeth and at least 1 tooth with fluorosis. The children were permanent residents in districts with either 0.5 mg or 2.5 mg fluoride/l in the drinking water. Fluorosis was assessed on the vestibular surfaces of all teeth using the modified Thylstrup and Fejerskov (TF) index. In order to r

Laminin gamma2 expression is developmentally regulated during murine tooth morphogenesis and is intense in ameloblasts.

Mutations in the laminin gamma2 gene cause junctional epidermolysis bullosa, and enamel hypoplasias are frequently seen in these patients. Laminin gamma2 is one of the three polypeptide chains forming the basement membrane glycoprotein laminin-5. We have localized the expression of the laminin gamma2 gene by in situ hybridization during mouse tooth development from early morphogenesis to completion of crown development. The expression was restricted to epithelial cells. During the early morphogenesis of the tooth germ, laminin gamma2 was expressed by the outer dental epithelium and by the stel

Understanding MIH: definition, epidemiology, differential diagnosis and new treatment guidelines.

Molar-Incisor Hypomineralisation (MIH) is a congenital disease which increases in prevalence. It affects permanent first molars and, often to a lesser degree, permanent incisors with variable severity. The aetiology is unknown, but different hypotheses have been advanced. Differential diagnosis is mandatory not to confound MIH with other diseases. Treatment consists in a minimally invasive approach by reinforcing and protecting the existing dental structure. In more severe cases, restorative treatment may be indicated.

MMP20 Overexpression Disrupts Molar Ameloblast Polarity and Migration.

Ameloblasts responsible for enamel formation express matrix metalloproteinase 20 (MMP20), an enzyme that cleaves enamel matrix proteins, including amelogenin (AMELX) and ameloblastin (AMBN). Previously, we showed that continuously erupting incisors from transgenic mice overexpressing active MMP20 had a massive cell infiltrate present within their enamel space, leading to enamel mineralization defects. However, effects of MMP20 overexpression on mouse molars were not analyzed, although these teeth more accurately represent human odontogenesis. Therefore, MMP20-overexpressing mice ( Mmp20+/+Tg+)
