The mGlu(2/3) receptor agonist LY379268 selectively blocks amphetamine ambulations and rearing.

We have previously reported that the specific group II metabotropic glutamate receptor agonist LY379268 inhibited phencyclidine (PCP)-induced motor activations in rats, but had mixed effects on behaviors produced by amphetamine. Here, LY379268 (1 mg/kg subcutaneous (s.c.)) attenuated amphetamine-induced ambulations and rearing but did not alter amphetamine-evoked fine motor movements. Consistent with a mechanism involving mGlu(2/3) receptors, the inhibitory actions of LY379268 on ambulations and rearing were reversed by LY341495, a mGlu(2/3) receptor antagonist. These data further suggest anti…

Long-lasting psychotomimetic consequences of repeated low-dose amphetamine exposure in rhesus monkeys.

The dopamine hypothesis of schizophrenia posits that dopamine dysregulation plays a key role in the etiology of schizophrenia. In line with this hypothesis, repeated amphetamine (AMPH) exposure has been shown to alter dopamine systems and induce behaviors reminiscent of positive-like and negative-like symptoms in both human and nonhuman primates. The mechanisms by which AMPH produces disturbances in brain function and behavior are not fully understood. The present study has employed a novel AMPH regimen, 12 weeks of intermittent escalating low doses of AMPH, to produce a nonhuman primate model…

Amphetamine infused into the ventrolateral striatum produces oral stereotypies and conditioned place preference.

The effects of amphetamine infused into the ventrolateral striatum (VLS) on locomotion, stereotypies, and conditioned place preference (CPP) were investigated. Five 2-day conditioning trials were conducted over 10 consecutive days. On 1 day of each trial, animals received an infusion of amphetamine (0, 2.5, 5, 10, or 20 mg/0.5 ml/side) and were placed into a distinct compartment for 30 min. On the other day, animals received sham intracranial infusions and were placed into a different compartment for 30 min. Locomotion and stereotypies were assessed following the first and last amphetamine inf…

Use of amphetamine-like appetite suppressants: a cross-sectional survey in Southern Brazil.

In 1994 a cross-sectional survey was undertaken in Pelotas, Southern Brazil, to assess the prevalence of amphetamine-like appetite suppressant use. 1,277 adults were interviewed, and the prevalence of anorectic drug use was 1.3% (95% CI = 0.7-1.9): 15 women and one man, mainly from higher socioeconomic groups. Most of the users (81%) had a medical prescription. Forty-one different drugs had been prescribed. Mean length of use was 8.7 months. These findings are discussed in terms of the overreliance on anorectics as aids to dieting, the dangers of polypharmacy, and the risks of long-term use.

Mixed D2/5-HT2 antagonism differentially affects apomorphine- and amphetamine-induced stereotyped behavior.

Evidence supports the hypothesis that psychostimulant stereotypy is mediated through postsynaptic dopamine receptors. Given the recent findings of behavioral, neurochemical and electrophysiological studies showing 5-HT2 modulation of dopamine systems, a series of experiments were undertaken to assess the ability of D2 and 5-HT2 antagonists to reverse apomorphine and amphetamine stereotypy in the rat. Haloperidol reduced stereotyped behavior induced by d-amphetamine (50% reduction with 0.162 mg/kg) and apomorphine (50% reduction with 0.112 mg/kg) MDL 28,133A, a mixed D2/5-HT2 antagonist, also r…

Sensitization to d-amphetamine after its repeated administration: evidence in EEG and behaviour.

After repeated administration of cocaine or d-amphetamine, a sensitization to their behavioural effects is frequently observed instead of a tolerance. In a previous study, it was shown that a moderate dose of d-amphetamine produced a pattern of EEG power spectrum which indicated a selective activation of D1-like dopamine receptors, whereas a larger dose induced a selective increase of power in the alpha-1 frequency band and, to a lesser degree, in the alpha-2 band, suggesting an additional activation of D2-like receptors. Furthermore, it was recently found that under a certain dosage and sched…

Mu opioid agonists potentiate amphetamine- and cocaine-induced rotational behavior in the rat.

Opioids modulate brain dopaminergic function in various experimental paradigms. This study used the rotational model of behavior in rats with unilateral 6-hydroxydopamine-induced lesions of the nigrostriatal pathway to investigate this interaction. Doses of two presynaptically acting dopaminergic drugs, amphetamine and cocaine, were coadministered with several doses of the mu opioid agonist, morphine. Morphine, at 3.0 mg/kg, potentiated rotational behavior induced by each dose of the stimulants. To determine the receptor specificity of the actions of morphine, the mu opioid agonists buprenorph…

Does dizocilpine (MK-801) selectively block the enhanced responsiveness to repeated amphetamine administration?

In order to examine the hypothesis that N-methyl-D-aspartate (NMDA) receptors are selectively involved in the development of stimulant sensitization, we characterized the interaction between acute and chronic dizocilpine (MK-801) + amphetamine using a detailed behavioral analysis, including concurrent assessment of the locomotor and stereotype components of the stimulant response and continuous monitoring of all the various phases of the emergent sensitization. The results showed that MK-801 (0.125 mg/kg) significantly affected the acute response to amphetamine (0.5, 1.75, 4.0 mg/kg), increasi…

Estradiol enhances behavioral sensitization to cocaine and amphetamine-stimulated striatal [3H]dopamine release.

Locomotor activity and stereotypy induced by cocaine is increased or 'sensitized' after repeated cocaine administration. This behavioral sen … ated rats. When striatal [3H]DA release was measured in vitro 1 or 7 days after the last injection, amphetamine-stimulated release was greater in vehicle-treated rats 7 days after cocaine injections but not 1 day after injections. In contrast, release was enhanced in group E both 1 and 7 days after cocaine.(ABSTRACT TRUNCATED AT 250 WORDS)

Interaction between amphetamine and alpha 2-postsynaptic adrenoreceptors in the rat submaxillary gland.

1 The interaction between amphetamine and the alpha 2-adrenoreceptor agonists, clonidine and guanabenz, was studied in the submaxillary gland of anaesthetised rats. 2 Low doses of clonidine (10 micrograms/kg) and guanabenz (10 micrograms/kg) inhibited the secretory responses induced by methacholine and substance P, respectively. 3 Amphetamine (300 micrograms/kg) antagonized the inhibitory effects of both alpha 2-agonists. This dose of amphetamine alone did not show sialagogic effects. 4 Atropine (1 micrograms/kg) diminished the secretory responses to methacholine as much as clonidine (10 micro…

Supersensitivity of d-amphetamine-induced hyperthermia in rats following continuous treatment with neuroleptics.

Following short term treatments with the neuroleptic drugs haloperidol, chlorpromazine and oxypertine, there was an increase in the sensitivity of rats to the hyperthermic effects of d-amphetamine. There was no significant difference in the body temperature of control and neuroleptic-treated rats prior to challenge with d-amphetamine. No change in the maximal hyperthermic response was observed. It is possible that these responses may be related to the neuroleptic malignant syndrome occasionally observed in human patients following short term neuroleptic treatments.

Correlation between anorectic potency and affinity for hypothalamic (+)-amphetamine binding sites of phenylethylamines.

The potencies of a number of phenylethylamines and related compounds to inhibit food intake were compared with their relative affinities for hypothalamic (+)-amphetamine binding sites in vitro. There was a positive correlation (r = 0.78, P less than 0.001, n = 19) between potencies of these compounds to inhibit food intake and their potencies to inhibit [3H](+)-amphetamine binding. Individual enantiomers of amphetamine, ephedrine, norephedrine, pseudoephedrine and norpseudoephedrine all inhibited food intake with a two- to five-fold greater potency than that of their respective optical isomers…

Amphetamine and other non-opioid drug users attending a special drug dependence clinic.

This paper reports the findings in 104 users of nonopioid drugs (mainly amphetamines) attending a drug dependence clinic in London between March 1968 and February 1969 in relation to demographic characteristics, forensic, psychiatric, and medical histories, clinical groups, management policy, and the use of biochemical tests for the presence of drugs in the urine.Nearly one-third gave a history of starting amphetamine misuse while at school (13.5%) or within a year of leaving school (16.3%). Amphetamine psychosis had occurred in 35% of cases and was a more frequent complication of intravenous …

In vitro inhibition of monoamine transport by amphetamine-like pre-workout supplement ingredients.

Phenethylamine (PEA) and alkylamine (AA) analogues are a prominent group of pre-workout food supplement ingredients. They are structurally related to the stimulant amphetamine and to the endogenous catecholamines noradrenaline and dopamine, implying potential cardiovascular and psychological effects. This study systematically investigated the inhibitory potential of 12 PEAs and 4 AAs identified in pre-workout supplements on the human dopamine transporter (hDAT), human noradrenaline transporter (hNET) and human serotonin transporter (hSERT) that are stably overexpressed in HEK 293 cells. All PE…

Explainable AI molecularly imprinted polymer electrochemiluminescence biosensor for discriminative methamphetamine and amphetamine detection.

Independent forensic quantification of methamphetamine (MA) and amphetamine (AMP) remains challenging due to their near-identical structures and overlapping electrochemical signatures. We report an end-to-end AI-integrated MIP-ECL biosensing framework employing target-specific MIP-NP/MWCNT/Nafion/Ir(pq)2(acac)/SPCE platforms. The Ir(III) luminophore within an MWCNT/Nafion matrix produced markedly superior ECL emission over the [Ru(bpy)3]2+ benchmark (40 mM DBAE, pH 8.0). MIP nanoparticles conferred high selectivity against structurally related interferents, validated in saliva, urine, and bloo…

Amphetamine-Associated Reversible Biventricular Cardiomyopathy Complicated by Acute Respiratory Failure: A Case Report.

Amphetamine overdose can cause severe cardiovascular toxicity, including rarely reversible cardiomyopathy. A 26-year-old man presented approximately 33 hours after intentionally ingesting 25 20-mg extended-release mixed amphetamine salt tablets with lorazepam. He developed chest pain, tachycardia, hypotension, hypoxemia, lactic acidosis, rhabdomyolysis, and markedly elevated cardiac biomarkers. Electrocardiography showed diffuse ST-segment depressions, and chest imaging demonstrated bilateral pulmonary edema and acute lung injury without pulmonary embolism. Echocardiography revealed severe glo…

Limitations and Future Directions in Pharmacological Treatment for Amphetamine-Type Stimulant Use Disorder.

Plain Language SummaryWhy Current Treatments for Stimulant Addiction Often Fall Short: Insights and Future Directions Stimulant addiction, such as addiction to drugs like amphetamines, is a growing public health concern, but there are very few effective medications to treat it. A recent study reviewed a drug called modafinil and found it didn't help reduce drug use, cravings, or improve treatment outcomes. This article discusses why treatments that work for other addictions, like opioids, are less effective for stimulants. It also highlights the challenges researchers face, such as differences…

Solriamfetol and m-chlorophenylpiperazine cause false positive amphetamine results on urine drug screening.

Urine drug screening by immunoassay is a common method to quickly identify drug exposures in the emergency setting and to detect unexpected drug exposures in a variety of patient care and occupational health settings. Although they provide rapid results, immunoassays are susceptible to cross-reactivity with other medications and metabolites. Herein we evaluate the performance of the Thermo Scientific DRI Amphetamines immunoassay for reactivity with trazodone, aripiprazole, atomoxetine, solriamfetol and relevant metabolites. Each of these compounds were spiked into drug-free urine across a rang…
