Muscarinic receptors in the rabbit carotid body were studied utilizing the specific muscarinic antagonist. [3H]quinuclidinyl benzilate. Biochemical and autoradiographic assays of ligand binding showed that receptor numbers and distributions remained unchanged following degeneration of axons and terminals of the carotid sinus nerve. The data suggest that muscarinic receptors are associated with carotid body parenchymal cells and are absent on afferent nerve terminals.

Exonuclease III digests DNA sequentially from the 3' end. This enzyme is used to analyse the location of nucleosomes on DNA fragments containing a particular 145 base-pair (bp) sequence. When one of these fragments is assembled into chromatin and digested with exonuclease, a strong and persistent pause in digestion is detected at a single location. The spontaneous evolution of molluscum contagiosum was studied on autoradiograms after incorporation of tritiated thymidine. Three patterns of distribution and intensity of labeling were distinguished and interpreted as three successive stages in the evolution of the lesions, corresponding to growth, steady-state, and regression. Controversy exists whether the initiating events in psoriasis are primarily epidermal or dermal (vascular). To study this point, serial biopsies from 6 patients were taken from the periphery of individual plaques before, and at 1 to 3 day intervals during, Goeckerman and PUVA treatments. Part of the biopsy was studied by electron microscopy to determine the fine structure of the capillary loops and part was incubated with tritiated thymidine to determine the labeling index (LI) of the basal cells. The non-A beta component (NAC) of Alzheimer's disease amyloid is a newly discovered 35 amino acid peptide found to be closely linked to the beta-amyloid fibrils in senile plaques. Apolipoprotein E (apoE) is another prominent constituent of senile plaques. In vitro studies have shown that apoE binds beta-amyloid (A beta) with high avidity, but it is unknown to what extent apoE interacts with NAC. The effects of the anti-schistosomal drug, niridazole, on the migration of Schistosoma mansoni larvae, biosynthetically radioisotope-labelled with 75[Se]-selenomethionine, was evaluated by autoradiography of compressed tissues of mice treated daily from Days 6 to 10 post-infection with 200 mg kg-1 niridazole. Osteophyte formation in a partial meniscectomy model of osteoarthritis (OA) in rabbits was studied. Serial sacrifices were performed at 1 day, 3 days, and 1, 4, 6, 8, 12 weeks in partial meniscectomy (15), nonoperated (14) and sham-operated (7) animals. H and E, safranin-O/fast green and toluidine blue stains were performed on coronal sections and pathologic findings correlated with 3H-thymidine autoradiography. Electron microscopic autoradiographic studies on the dynamics and location of DNA synthesis by means of incorporation of 8H-thymidine during the replication of duck plague virus (DPV) revealed that the duration of DNA synthesis of DPV was rather long. The replication of viral DNA occurred simultaneously with the assembly procedure of nucleocapsids, the maturation and release of viruses. DNA synthesis of DPV occurred in the matrix with lower electron density in the nucleus. A biopsy case of endolymphatic stromal myosis, a rare neoplasm of the uterus, which occurred in a 33-year-old female was described. The histology was typical and resembled stromal cells of proliferative phase endometrium. The tumor extended in the endometrium and diffusely infiltrated into the myometrium and the parametrium. However, 3H-thymidine labeling index was low suggesting slow growth of the tumor.

Tritiated thymidine radioautography was employed to study the effect of cortisol and other glucocorticoids on cellular proliferation in the aorta and pulmonary artery of rabbits with cholesterol atherosclerosis. Labelled cell counts showed that glucocorticoids, even after one day and at a relatively low dose, decrease sharply the deoxyribonucleic acid synthesis in the intimal plaques. The hormonal influence on [3H]thymidine uptake seems to be a dose-dependent process.
