Hematological, biochemical and enzymological responses in an Indian major carp Labeo rohita induced by sublethal concentration of waterborne selenite exposure.

Selenium (Se) pollution in aquatic ecosystem is an environmental issue throughout the world. Elevated concentrations of inorganic Se from agricultural and industrial processes may cause adverse biological effects in aquatic organisms such as fish. In the present study, Labeo rohita an Indian major carp were exposed to sublethal concentration of Se (sodium selenite) for 35days and certain hematological, biochemical and enzymological parameters were estimated. The median lethal concentration of waterborne sodium selenite (Na2SeO3) to L. rohita was found to be 23.89mgL(-1) for 96h. During subleth

Massive mortality of Prussian carp Carassius gibelio in the upper Elbe basin associated with herpesviral hematopoietic necrosis (CyHV-2).

From 22 May to 10 June 2011 massive mortality of Prussian carp Carassius gibelio was observed in alluvial Lake Řehacˇka close to the Elbe River in the Czech Republic. More than 1400 kg of dead fish were collected and no other fish species were affected. Further molecular and cytogenetic investigation of fish (n = 232) revealed that the Rˇehacˇka population of Prussian carp consisted exclusively of gynogenetic triploid females. The causative agent was identified by means of molecular and electron microscopy as a herpesviral hematopoietic necrosis virus (Cyprinid herpesvirus 2, CyHV-2). This is

Photoreceptor proteins and melatonin rhythm generating AANAT in the carp pineal: Temporal organization and correlation with natural photo-thermal cues.

We studied temporal organization of both the photoreceptor (rod-like opsin, alpha subunit of the G protein transducin or alpha-TD) and melatonin generating (AANAT) proteins in the same pineal of a tropical surface dwelling free-living carp Catla catla, and analyzed possible correlation between them as well as with natural photo-thermal variables in an annual cycle. The pineal from individual fish was collected at four different time points (06.00 h, 12.00 h, 18.00 h, and 24.00 h) in a 24.00 h cycle and the same was repeated in four distinct seasons in an annual reproductive cycle to study each

Synaptic contribution of Ca2+-permeable and Ca2+-impermeable AMPA receptors on isolated carp retinal horizontal cells and their modulation by Zn2+.

Ca(2+)-permeable and Ca(2+)-impermeable AMPA receptors are co-expressed on carp retinal horizontal cells. In the present study, we examined the synaptic contribution and Zn(2+) modulatory effect of these two AMPA receptor subtypes using whole-cell patch clamp technique. Specific Ca(2+)-permeable AMPA receptor antagonist (1-naphthyl acetyl spermine, NAS) and selective Ca(2+)-impermeable AMPA receptor blocker (pentobarbital, PB) were used to separate the glutamate-response in isolated H1 horizontal cell mediated by these two subtypes of AMPA receptors respectively. Application of 100 microM NAS

Morphological specializations of the buccal cavity in relation to the food and feeding habit of a carp Cirrhinus mrigala: a scanning electron microscopic investigation.

The buccal cavity of an herbivorous fish, Cirrhinus mrigala, was investigated by scanning electron microscopy to determine its surface ultrastructure. The buccal cavity shows significant adaptive modifications in relation to food and feeding ecology of the fish. The buccal cavity of the fish is of modest size and limited capacity, which is considered an adaptation with respect to the small-sized food items primarily consumed by the fish that could be accommodated in a small space. Modification of surface epithelial cells, on the upper jaw, into characteristic structures-the unculi-is considere

Effects of iron sulfate dosage on the water flea (Daphnia magna Straus) and early development of carp (Cyprinus carpio L.).

Adult water fleas, Daphnia magna Straus, and the early life stages of carp, Cyprinus carpio L., were exposed to river water near an iron sulfate dosage installation to determine the effects of phosphate precipitation with iron(II)sulfate. Tests were conducted during two consecutive dosage periods of 3,000 and 5,000 kg/day iron sulfate (520 and 620 microg/L total Fe respectively) at the dosage site and at a reference site (60 microg/L total Fe) further downstream. Though survival remained unaffected, the filter-feeding D. magna accumulated iron and other metals at the dosage site. Viability of

In vitro cell-mediated cytotoxicity against allogeneic erythrocytes in ginbuna crucian carp and goldfish using a non-radioactive assay.

Cell-mediated cytotoxicity of clonal ginbuna crucian carp leukocytes against allogeneic erythrocytes is described using a sensitive non-radioactive in vitro assay. Hemoglobin released from target erythrocytes after cell-mediated erythrolysis was detected by tetramethylbenzidine (TMB). TMB assay showed clear correlation with a 51Cr-release assay and even exhibited higher cytotoxicity. The use of erythrocytes as target cells has several advantages over a conventional 51Cr-release assay. Erythrocytes do not have cytotoxic activity, are relatively homogeneous, are available in large numbers and er

Physiological and Pathological Responses of Grass Carp (Ctenopharyngodon idella) to Total Dissolved Gas Supersaturation During Exposure and Subsequent Recovery.

The total dissolved gas (TDG) supersaturation induced by discharged floods severely threatens fish survival during the flood season. However, investigations into the physiological and pathological effects of TDG supersaturation on fish remain limited, and even less is known about the recovery from TDG-induced injuries at the physiological and pathological levels. The biochemical and histopathological responses of fish injured by TDG exposure, and whether injured fish can return to normal conditions, remain unclear. In this study, on the basis of a combination of in situ investigations and labo

A Survey of the Species Origins of Common Fish Cell Lines-With a Particular Focus on the Grass Carp Ovarian Cell Line (GCO).

Cell line misidentification or contamination undermines the reliability of research outcomes. In this study, we verified the species origin of commonly used fish cell lines, with a particular focus on the grass carp ovarian cell line (GCO). RNA-seq data from 146 samples of 11 widely used fish cell lines were collected from public databases (NCBI-SRA) and were mapped to the reference genomes of eight species. GCO RNA-seq samples from three Chinese laboratories showed a mapping rate of 21.64% ± 3.61% to the grass carp genome, but 89.81% ± 4.83% to the fathead minnow genome. Further transcriptomi

Differences in Susceptibility to Cyprinid Herpesvirus 3 (CyHV-3) Infection Among Carp (Cyprinus carpio L.) Strains and Hybrids.

Cyprinid herpesvirus 3 (CyHV-3) is a pathogen that causes high mortality in common carp (Cyprinus carpio) and koi. Common carp breeding lines with different genetic backgrounds exhibit different resistance levels to viral pathogens. This study aimed to determine the differences in CyHV-3 disease resistance performance between the hybrid offspring (Y × M and M × Y) of the mirror carp 'Longke 11' (resistant to CyHV-3) and Yellow River carp, as well as the self-crossed offspring (M and Y). The M, Y × M, M × Y and Y groups were infected with CyHV-3 by immersion. The order of mortality and the dura

Unraveling the interplay of microbial succession, flavor deterioration, and physicochemical dynamics in vacuum-packed marinated silver carp during storage.

This study systematically investigated the quality and flavor deterioration and microbial succession of marinated silver carp (MSC) during storage. After 60 days of storage, the samples showed significant morphological deterioration, decreased sensory acceptability, while moisture content and pH value initially decreased and then increased. Synergistic protein-lipid oxidation accelerated deterioration, as reflected by rising TVB-N and TBARS levels, along with sulfhydryl group depletion, and carbonyl accumulation. Flavor analysis confirmed sulfides, aldehydes, and ketones as major volatile comp

Sox9a and amh indirectly cooperate to promote spermatogonia differentiation and gonadal development in Cyprinus carpio var. koi(koi carp).

In teleost fish, sex differentiation is co-regulated by genetic factors and environmental signals. Genetically, Wnt4, Rspo1, Foxl2, and cyp19a dominate female differentiation, while dmrt1, Sox9, SF-1, and amh govern male differentiation. Among these, amh (TGF-βsuperfamily domain) and Sox9a (HMG-box domain) are conserved male-specific genes critical for testicular development in zebrafish and goldfish. Notably, their functional interactions under long-term artificial selection in koi carp remains unelucidated at the molecular level. Additionally, female koi carp have higher market value in aquac

Thirty-week exposure reveals the damaging effects of microplastics: Histology, immunity and intestinal microbiota of grass carp (Ctenopharyngodon idella).

Microplastic (MP) pollution has become a global aquatic environmental issue, yet its long-term toxic effects on aquatic organisms remain poorly understood. In this study, grass carps (Ctenopharyngodon idella) were subjected to thirty-week dietary exposure to polystyrene microplastics (PS-MPs) to systematically evaluate the effects on histology, immune signaling, and intestinal microbiota. Histopathological examination revealed that PS-MPs exposure induced focal necrosis, increased melanomacrophage centers, and sinusoidal congestion in the spleen, while the intestine exhibited villus fusion, di…

Identification and analysis of lncRNA50877, which is involved in KRT8-mediated mitochondrial homeostasis and GCRV replication in grass carp (Ctenopharyngodon idella).

Grass carp (Ctenopharyngodon idella) reovirus (GCRV), the causative agent of hemorrhagic disease, causes substantial economic losses in the grass carp industry every year. However, the biological regulatory roles of long non-coding RNAs (lncRNAs) during GCRV infection remain largely unclear. In this study, high-throughput sequencing technology was used to analyze the expression profiles of lncRNAs in GCRV-infected and mock-infected liver and intestinal tissues of grass carp. In this study, compared with those in the control group, the transcriptomic profiles of grass carp after GCRV genotype I

Genome to single-cell characterization of the MAPK family in bighead carp (Hypophthalmichthys nobilis) spleen during Aeromonas hydrophila challenge.

INTRODUCTION: Mitogen-activated protein kinases (MAPKs) are central to stress and innate immune signaling in fish, but their genomic composition and infection related transcriptional responses remain unclear in bighead carp (Hypophthalmichthys nobilis). METHODS: We performed a comparative genomic survey across 24 species and characterized MAPK family members in bighead carp based on conserved motifs, gene structure, and synteny. Tissue expression profiling, transcriptome sequencing, qRT-PCR, weighted gene co-expression network analysis (WGCNA), and single-cell RNA sequencing were used to inve
