The battle for oxygen during bacterial and fungal infections.

Bacterial and fungal pathogens face various microenvironmental conditions during infection. In addition to acidosis, nutrient consumption, and hypercapnia, pathogen infections are associated with hypoxia, which is induced by bacterial and fungal respiration during the formation of foci of infection or biofilms. Consequently, the in vivo interaction between host immune cells and pathogens is anticipated to occur mainly under low-oxygen conditions. Various infectious disease models have reported that pathogens benefit from hypoxia, which dampens the oxygen-dependent antimicrobial activities of m

Targeting ClpP Rewires the Metabolic-Immune Axis in Acute Myeloid Leukemia.

Acute myeloid leukemia (AML) critically depends on oxidative phosphorylation (OXPHOS). Mitochondrial targeted therapies are advancing into c … hondrial respiration and oxygen consumption in AML cells while exerting minimal direct effects on T-cell respiration. By reducing AML oxygen consumption and metabolic demand, IMP125 alleviated hypoxia and metabolic competition within the leukemic niche. In this remodeled microenvironment, T cells exhibited recovery of OXPHOS, effector function, and memory-associat …

Heightened sensitivity to adverse effects of metformin in mtDNA mutant patient cells.

AIMS: Metformin (Met) is a widely used, cost-effective, and relatively safe drug, primarily prescribed for diabetes, that also exhibits beneficial effects in other conditions, such as in cardiovascular diseases, neurological disorders, and cancer. Despite its common use, the safety of Met in patients with primary mitochondrial disease remains uncertain, as both Met and mitochondrial dysfunction increase the risk of lactic acidosis. Here we have examined the effects of Met in patient cells with m.3243A>G mitochondrial DNA mutation. MATERIALS AND METHODS: We utilized induced pluripotent stem ce

miR-125b Mediates Tau-induced Mitochondrial Dysfunction in a Cellular Model of Alzheimer's Disease.

MicroRNA (miRNA) dysregulation contributes to the pathogenesis of neurodegenerative diseases, including Alzheimer's disease (AD), but the role of specific miRNAs in tau-mediated toxicity remains unclear. Here, we investigated the contribution of hsa-miR-125b to tau-induced cellular dysfunction using a stable neuronal cell line overexpressing mutant tau (P301L). We found that hsa-miR-125b levels were significantly elevated in tau-expressing cells compared to controls. Using bioinformatic tools, we identified a strong enrichment of predicted hsa-miR-125b targets involved in mitochondrial functio

Metabolic Response to CDK4/6 Inhibition in ER+ Breast Cancer Creates a Therapeutic Vulnerability in Drug-Tolerant Persister Cells.

Although endocrine therapies prevent recurrence and progression of estrogen receptor alpha (ER)-positive breast cancer, approximately one-third of patients experience recurrent disease that is rarely cured in the advanced/metastatic setting. A subpopulation of endocrine-tolerant breast cancer cells persists as residual disease that confers risk for the eventual emergence of drug resistance. An analysis of persisters that continue to proliferate despite endocrine therapy revealed the activation of pathways related to metabolism and E2F transcription factor signaling. E2F signaling is driven by

Characterization of a UQCRC1 variant in a patient with progressive weakness, pain and sleep issues reveals a functional mitochondrial defect restored by mitochondrial transplantation.

Primary mitochondrial defects underlie the heterogeneity of many rare inherited disorders. Pathogenic variants that disrupt the function of the multi-subunit protein complexes of the mitochondrial respiratory chain contribute to a range of neurological phenotypes and other clinical manifestations. These variants are also thought to contribute to the onset and progression of numerous more common neurodegenerative conditions such as Parkinson's and Alzheimer's disease. Here we describe an individual affected with progressive muscle weakness and pain harboring a paternally inherited missense vari

Energized mitochondria increase the dynamic range over which inositol 1,4,5-trisphosphate activates store-operated calcium influx.

In eukaryotic cells, activation of cell surface receptors that couple to the phosphoinositide pathway evokes a biphasic increase in intracellular free Ca2+ concentration: an initial transient phase reflecting Ca2+ release from intracellular stores, followed by a plateau phase due to Ca2+ influx. A major component of this Ca2+ influx is store-dependent and often can be measured directly as the Ca2+ release-activated Ca2+ current (I(CRAC)). Under physiological conditions of weak intracellular Ca2+ buffering, respiring mitochondria play a central role in store-operated Ca2+ influx. They determine

[Respiratory chain of Candida mycoderma].

The respiration chain of the yeast Candida mycoderma was studied during its growth on glucose. Electrons can be transported from the pyridine nucleotide (PN) pool to oxygen by two pathways, as was shown in experiments with intact cells on the basis of inhibitory analysis of respiration and the extent of reduction of electron carriers. The first pathway is inhibited by cyanide and antimycin A; it includes PN, flavoprotein, (FP) and cytochromes b, c and a + a3. The second pathway of electron transport is resistant to cyanide (CrPET), includes PN and FP, lacks cytochromes, and is blocked by salic

New insights into the genic and metabolic characteristics of induced pluripotent stem cells from polycystic ovary syndrome women.

BACKGROUND: Polycystic ovary syndrome (PCOS) is a common endocrine and metabolic disorder that affects female fertility. However, with the lack of a corresponding research model, the pathology mechanism of PCOS is poorly understood. Induced pluripotent stem cell (iPSC) technology has been recognized as means to generate patient-specific stem cells for disease modeling. METHODS: The mRNA abundance of iPSCs was analyzed by RNA microarray and real-time polymerase chain reaction (RT-PCR). Karyotyping of iPSCs was performed with cytogenetic analysis. The mitochondrial respiration ability and glyco

Humidity and Deposition Solution Play a Critical Role in Virus Inactivation by Heat Treatment of N95 Respirators.

Supply shortages of N95 respirators during the coronavirus disease 2019 (COVID-19) pandemic have motivated institutions to develop feasible and effective N95 respirator reuse strategies. In particular, heat decontamination is a treatment method that scales well and can be implemented in settings with variable or limited resources. Prior studies using multiple inactivation methods, however, have often focused on a single virus under narrowly defined conditions, making it difficult to develop guiding principles for inactivating emerging or difficult-to-culture viruses. We systematically explored

Differentiating the in vitro toxicity of solid-surface composite dust: A comparison of material components and abrasive particles.

BACKGROUND: Workers fabricating solid-surface composite (SSC) materials like Corian® are exposed to airborne particulate matter (PM) containing aluminum trihydrate (ATH) and potentially abrasive particles from sanding tools, leading to concerns about respiratory health effects like pulmonary fibrosis. However, the relative toxicological contributions of the SSC material versus the sandpaper abrasives remain unclear. OBJECTIVE: This study aimed to compare the in vitro cellular responses induced by respirable dust generated from sanding SSC with different commercial sandpapers (aluminum oxide [

A novel mechanism of cannabidiol in suppressing ovarian cancer through LAIR-1 mediated mitochondrial dysfunction and apoptosis.

Cannabidiol (CBD) is a nonpsychoactive cannabinoid compound. It has been shown that CBD can inhibit the proliferation of ovarian cancer cells, but the underlying specific mechanism is unclear. We previously presented the first evidence for the expression of leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1), a member of the immunosuppressive receptor family, in ovarian cancer cells. In the present study, we investigated the mechanism by which CBD inhibits the growth of SKOV3 and CAOV3 ovarian cancer cells, and we sought to understand the concurrent role of LAIR-1. In addition to indu

Metabolic profile of leukemia cells influences treatment efficacy of L-asparaginase.

BACKGROUND: Effectiveness of L-asparaginase administration in acute lymphoblastic leukemia treatment is mirrored in the overall outcome of patients. Generally, leukemia patients differ in their sensitivity to L-asparaginase; however, the mechanism underlying their inter-individual differences is still not fully understood. We have previously shown that L-asparaginase rewires the biosynthetic and bioenergetic pathways of leukemia cells to activate both anti-leukemic and pro-survival processes. Herein, we investigated the relationship between the metabolic profile of leukemia cells and their sen

Development of a high throughput assay for indirectly measuring phage growth using the OmniLog(TM) system.

The conventional and most accepted method of measuring the lytic activity of a phage against its bacterial host is the plaque assay. This me … e phage growth using the OmniLog(TM) system. The OmniLog(TM) system uses redox chemistry, employing cell respiration as a universal reporter. During active growth of bacteria, cellular respiration reduces a tetrazolium dye and produces a color change that is measured in an automated fashion. On the other hand, successful phage infection and subsequent growth of th …

Reversible blockade of electron transport during ischemia protects mitochondria and decreases myocardial injury following reperfusion.

Cardiac mitochondria sustain damage during ischemia and reperfusion, contributing to cell death. The reversible blockade of electron transport during ischemia with amobarbital, an inhibitor at the rotenone site of complex I, protects mitochondria against ischemic damage. Amobarbital treatment immediately before ischemia was used to test the hypothesis that damage to mitochondrial respiration occurs mainly during ischemia and that protection of mitochondria during ischemia leads to decreased cardiac injury with reperfusion. Langendorff-perfused Fischer-344 rat hearts were treated with amobarbit

Impact of geometrical uncertainties on 3D CRT and IMRT dose distributions for lung cancer treatment.

PURPOSE: To quantify the effect of set-up errors and respiratory motion on dose distributions for non-small cell lung cancer (NSCLC) treatment. METHODS AND MATERIALS: Irradiations of 5 NSCLC patients were planned with 3 techniques, two (conformal radiation therapy (CRT) and intensity modulated radiation therapy (IMRT1)) with a homogeneous dose in the planning target volume (PTV) and a third (IMRT2) with dose heterogeneity. Set-up errors were simulated for gross target volume (GTV) and organs at risk (OARs). For the GTV, the respiration was also simulated with a periodical motion around a vary

Transcriptomic and functional profiling reveal autophagy inhibition and persistent bioenergetic collapse following parallel photodamage to lysosomes and mitochondria.

Photodynamic therapy (PDT) using 1,9-dimethyl methylene blue (DMMB) induces coordinated mitochondrial and lysosomal damage and results in strong cellular death induction. However, the underlying transcriptional regulation in response to DMMB remains elusive. We compared the transcriptome response of photoactivated DMMB (paDMMB) to the gene signature triggered by autophagy-modulating agents: rapamycin (an autophagy activator) and bafilomycin A1 (an autophagy inhibitor). Transcriptome analysis revealed a pronounced transcriptomic response to paDMMB, with 884 differentially expressed genes (DEGs)

Cellular energetics and mitochondrial uncoupling in canine aging.

The first domesticated companion animal, the dog, is currently represented by over 190 unique breeds. Across these numerous breeds, dogs have exceptional variation in lifespan (inversely correlated with body size), presenting an opportunity to discover longevity-determining traits. We performed a genome-wide association study on 4169 canines representing 110 breeds and identified novel candidate regulators of longevity. Interestingly, known functions within the identified genes included control of coat phenotypes such as hair length, as well as mitochondrial properties, suggesting that thermor

Mitochondria poor but capacity rich? The paradox of podocyte respiration.

Campbell et al. show that podocyte mitochondria, isolated using a cell-specific MITO-Tag approach, possess high intrinsic respiratory capacity despite their low abundance. The study demonstrates that conventional culture conditions suppress mitochondrial respiration and increase oxidative stress, suggesting that current experimental systems may underestimate mitochondrial function in podocytes. The authors provide evidence that mitochondrial respiratory capacity in podocytes declines in male mice with aging, indicating that mitochondria could play a role in age-associated glomerular injury.

Exercise training normalizes mitochondrial respiratory capacity within the striatum of the R6/1 model of Huntington's disease.

Huntington's disease (HD) is a neurodegenerative disorder characterized by progressive cell loss in the striatum and cerebral cortex, leading to a decline in motor control and eventually death. The mechanisms promoting motor dysfunction are not known, however loss of mitochondrial function and content has been observed, suggesting that mitochondrial dysfunction may contribute to HD phenotype. Recent work has demonstrated that voluntary wheel running reduces hindlimb clasping in the R6/1 mouse model of HD, which we hypothesized may be due to preservation of mitochondrial content with exercise.
