Extrapotentials and allorhythmias as an expression of experimental parasystole.

UNLABELLED: The aim of the study was to investigate the dynamics of experimental parasystole taking into consideration the peculiarities of recurrent arrhythmias recorded in clinical settings. MATERIAL AND METHODS: The experiments were conducted on isolated right atria of seven chinchilla rabbits. Parasystolic arrhythmias using periodical one-site electrostimulation were provoked in one atrium, where the sinus node was not affected, and in two atria with the spontaneous low value activity of pacemakers. The parasystolic arrhythmias by the dual-site periodical pacing were provoked in four atri…

Repair of chronic tympanic membrane perforations using epidermal growth factor: progress toward clinical application.

In an earlier study, epidermal growth factor (EGF) was shown to be effective in healing chronic tympanic membrane (TM) perforations in the chinchilla. The original protocol required rimming of the perforation's epithelial edge, application of a paper patch, placement of a Gelfoam pledget, and then administration of EGF solution. To develop a simple outpatient method of healing chronic TM perforations, an attempt was made to simplify the treatment protocol while preserving efficacy. In the modified experimental protocol, a large Gelfoam pledget was placed over the chronic perforation in contact…

Genetic control of bone marrow graft rejection. I. Determinant-specific difference of reactivity in two pairs of inbred mouse strains.

Transplantation of 5 x 10(5) DBA/2 (H-2(d)) bone marrow cells into irradiated B10 and 129-strain mice (both H-2(b)) resulted in graft failur … roup IX, A(w) (white-bellied agouti) of linkage group V, Sl (steel) of linkage group IV, and c(ch) (chinchilla) and p (pink eye, dilute) of linkage group I. There were suggestions, however, that the regulator genes of marrow graft rejection are either non-H-2 histocompatibility genes or other genetic factors closely linked with them.

Neutrophil-mediated delivery of antibiotics across the tympanic membrane for otitis media treatment in a preclinical model.

Acute otitis media (AOM) is a leading cause of pediatric antibiotic prescriptions. Systemic antibiotics cause side effects and antibiotic resistance, whereas local delivery of antibiotics (directly to the middle ear) is hindered by an impermeable biological barrier, the tympanic membrane (TM). Here, we developed hydroxylated liposomes that are internalized by neutrophils recruited during AOM to transport encapsulated ciprofloxacin across the intact TM. In mice, complement opsonization of the hydroxylated liposomes increased neutrophil uptake ex vivo and in vivo. In chinchillas with AOM induced…

Design of medical tympanostomy conduits with selective fluid transport properties.

Implantable tubes, shunts, and other medical conduits are crucial for treating a wide range of conditions from ears and eyes to brain and li … reservation and exhibited more efficient and rapid antibiotic delivery to the middle ear in healthy chinchillas compared with current tympanostomy tubes, without resulting in ototoxicity at up to 24 weeks. The design principle and optimization algorithm presented here may enable tubes to be customized for a wide range of patient needs.

Ultrasonic bone removal from the ossicular chain affects cochlear structure and function.

INTRODUCTION: Ultrasonic bone removal devices (UBD) are capable of cutting through bony tissue without injury to adjacent soft tissue. The feasibility and safety of using this technology for removal of bone from an intact ossicular chain (as might be required for otosclerosis or congenital fixation) was investigated in an animal model. METHODS: This was a prospective animal study conducted on seven anesthetised adult chinchillas. An UBD was used to remove bone from the malleus head in situ. Pre and post-operative distortion product otoacoustic emission (DPOAE) levels and auditory brainstem re…

N-Acetyl L-Cysteine does not protect mouse ears from the effects of noise*.

BACKGROUND: Noise-induced hearing loss (NIHL) is one of the most common occupational injuries in the United States. It would be extremely valuable if a safe, inexpensive compound could be identified which protects worker hearing from noise. In a series of experiments, Kopke has shown that the compound N-acetyl-L-cysteine (L-NAC) can protect the hearing of chinchillas from the effects of a single exposure to noise. L-NAC is used in clinical medicine and is very safe. Although L-NAC was reported to be promising, it has not been successful in other studies (Kramer et al., 2006; Hamernik et al., 2…

Exposure and impact assessment of emissions from mercury recycling using domestic rabbits.

A biomonitoring study using domestic rabbits (Chinchilla) aimed at the exposure and impact risks assessment of emissions released into the ambient air from a mercury-recycling plant has been carried out. Groups of rabbits were exposed to the emissions during 6 months at biomonitoring stations built up in two localities (Rudnany and Matejovce) in the distance of about 3 and 6 km around and downwind from the mercury-producing plant. The aim of the biomonitoring was to trace the translocation of inhaled inorganic Hg in body tissues and the immunotoxic impact of the emissions in the exposed mammal…

The effect of 'conditioning' exposures on hearing loss from impulse noise.

Research has shown that prior noise exposures or 'conditioning' can moderate the amount of permanent threshold shift (PTS) from subsequent high intensity noise exposures. The aim of this experiment was to study the effect of 'conditioning' on subsequent exposure to high intensity impulse noise. The subjects were seven experimental and 14 control monaural chinchillas. Evoked potential (EVP) thresholds were measured before and after the noise exposures. Experimental animals received 10 days of exposure to an octave band noise (OBN) centered at 0.5 kHz OBN at 95 dB SPL (6 h on/18 h off) and allow…

Cochlear mechanics: implications of electrophysiological and acoustical observations.

Implications of the spatial distribution of distortion products (2f1--f2) and (f2--f1) observed from populations of cochlear nerve fibers for cochlear mechanics are reviewed (the terms f1 and f2 represent the primary stimulus frequencies; f1 < f2). Characteristics of the distortion products (2f1--f2) and (f2--f1) in the ear-canal sound pressure of the cat and the chinchilla are investigated. Physiological origin of the acoustic distortion product (2f1--f2) is supported by demonstrations of the vulnerability of the distortion product to anoxia, to overstimulation and to cyanide perfusion of the…

Temporary Unilateral Hearing Loss Impairs Spatial Auditory Information Processing in Neurons in the Central Auditory System.

Temporary conductive hearing loss (CHL) can lead to hearing impairments that persist beyond resolution of the CHL. In particular, unilateral CHL leads to deficits in auditory skills that rely on binaural input (e.g., spatial hearing). Here, we asked whether single neurons in the auditory midbrain, which integrate acoustic inputs from the two ears, are altered by a temporary CHL. We introduced 6 weeks of unilateral CHL to young adult chinchillas via foam earplug. Following CHL removal and restoration of peripheral input, single-unit recordings from inferior colliculus (ICC) neurons revealed the…

The colonic groove or furrow: a comparative morphological study of six species of African mole-rats (Rodentia, Bathyergidae).

Herbivorous mammals such as nutrias, guinea pigs, chinchillas, and mole-rats have a longitudinal mucosal colonic groove (furrow) in their ascending colon, which is thought to play a role in the colonic separation mechanism (CSM). It is not known whether this groove is structurally modified to adapt to this function in mole-rat species. The morphology of this groove was studied in 32 mol-rats, four species, one of which consisted of three subspecies, endemic to southern Africa and two species found in eastern Africa. The macroscopic morphology of the groove was documented, and samples for histo…

Divergent Auditory Nerve Encoding Deficits Between Two Common Etiologies of Sensorineural Hearing Loss.

Speech intelligibility can vary dramatically between individuals with similar clinically defined severity of hearing loss based on the audiogram. These perceptual differences, despite equal audiometric-threshold elevation, are often assumed to reflect central-processing variations. Here, we compared peripheral-processing in auditory nerve (AN) fibers of male chinchillas between two prevalent hearing loss etiologies: metabolic hearing loss (MHL) and noise-induced hearing loss (NIHL). MHL results from age-related reduction of the endocochlear potential due to atrophy of the stria vascularis. MHL…

A novel gel patch for minimally invasive repair of tympanic membrane perforations.

OBJECTIVE: Evaluate the efficacy of a photocrosslinkable gel patch for repairing tympanic membrane (TM) perforations using a minimally invasive procedure. METHODS: 38 adult male chinchillas underwent bilateral TM perforation via CO2 laser (n = 76 TMs). Eight weeks post-perforation induction, either a gel patch (n = 26) or EpiDisc (n = 12) was applied to the perforation through the ear canal. Perforation margins were not abraded prior to gel patch application in order to make the procedure minimally invasive. During the study, the application process was refined, and 9 of 26 gel-treated TMs re…
