Dietary β-Glucan From the Marine Diatom Chaetoceros muelleri Enhances Immune Responses and Resistance of Nile Tilapia to Tilapia Lake Virus Infection.

This study evaluated the immunomodulatory effects of dietary β-glucan derived from the marine diatom Chaetoceros muelleri on immune responses and survival of Nile tilapia (Oreochromis niloticus) following Tilapia Lake Virus (TiLV) infection. Juvenile tilapias were fed diets supplemented with 0.1% or 0.2% β-glucan for 14 days prior to viral challenge. Fish were subsequently challenged with TiLV and immune-related gene expression was analysed in spleen and liver tissues. Fish fed the 0.1% β-glucan diet showed significantly lower cumulative mortality (26.67%) compared with the control group (55.5

IL-21 controls CD8 + T cell immunity and antibacterial defense in fish via STAT3 signaling.

CD8 + T cells are indispensable effectors of adaptive immunity. While cytokine-mediated regulation of CD8 + T cell responses has been extensively characterized in mammals, the underlying mechanisms in fish remain largely unexplored. Here, we identified and characterized an interleukin-21 (IL-21) gene from Nile tilapia ( Oreochromis niloticus). In vitro stimulation with IL-21 significantly upregulated the transcription and expression of key CD8 + T cell effector molecules, including Granzyme B and interferon-gamma (IFN-γ), in splenic leukocytes, indicating a crucial role of IL-21 in modulating

Ameliorative potential of Populus alba leaf powder against hexaflumuron exposure in Nile tilapia: immune-antioxidant, biochemical, histological, and transcriptomic analysis.

Contamination of the aquatic bodies with pesticides is a serious issue that hinders the aquaculture industry worldwide. Preventing aquatic pollution is a challenge, and finding eco-friendly strategies could help to overcome such a problem. Herein, we studied the antagonistic potential of dietary fortification of white poplar (Populus alba; PA) leaf powder against chronic hexaflumuron (HX) toxicity in Nile tilapia (Oreochromis niloticus). Fish (n = 200; 36.20 ± 1.55 g) were eventually grouped into four groups with five replicates and kept for 60 days. The C (control) and PA groups were fed basa

Presence of virulent Edwardsiella tarda in farmed nile tilapia and striped catfish.

The production of striped catfish (Pangasianodon hypothalamus) has increased worldwide; recently, it was farmed with Nile tilapia in polyculture farms. Polyculture systems and water temperature (25℃ and 33℃) could affect Edwardsiella tarda infection, antibiotic efficacy, and residues. Moribund fishes were collected from three Farms 1-3: Farm 1 (monoculture, Nile tilapia), Farm 2 (monoculture, striped catfish), and Farm 3 (polyculture). Four E. tarda, LAMSH1, and LAMAH2-4 were isolated, whereas LAMAH3 was isolated from both fish spp., where striped catfish were highly susceptible to infection.

The Potential Role of Gonadotropic Hormones and Their Receptors in Sex Differentiation of Nile Tilapia, Oreochromis niloticus.

Nile tilapia, as an ideal model for studying sex differentiation, is a popular farmed fish worldwide with a stable XX/XY sex-determination system. In tilapia, ovarian differentiation is triggered by estradiol-17β (E2) production in undifferentiated gonads. In a previous study, we suggested that follicle-stimulating hormone (FSH) signaling might be involved in ovarian differentiation in Nile tilapia. In this study, we further investigated the role of FSH signaling in ovarian differentiation via aromatase expression, which converts testosterone to E2. Masculinization of XX fry by aromatase inhib

Genome-wide characterization of the interleukin gene family in Nile tilapia (Oreochromis niloticus) spleen: identification, expression dynamics, and regulatory mechanism during Streptococcus agalactiae infection.

The spleen is the principal site of immune hematopoiesis and a major target of Streptococcus agalactiae (S. agalactiae) attack in Nile tilapia (Oreochromis niloticus). However, the immune-regulatory mechanisms employed by this organ against S. agalactiae remain unclear. In this study, integrated transcriptomic analysis of spleen tissue identified 27 genes-among them the core cytokines IL-6, CXCL3 and IL1R2-that responded to S. agalactiae challenge. Genome-wide mining subsequently revealed a total of 27 IL-family members in Nile tilapia. Synteny and chromosomal location analyses indicated that

Effects of dietary supplementation with fenugreek oil on growth, physiology, and resistance to A. hydrophila infection in Nile tilapia (Oreochromis niloticus) fingerlings.

BACKGROUND: Aeromonas hydrophila is an opportunistic pathogen that causes Motile Aeromonad Septicemia (MAS) with significant losses in Nile tilapia farms. Because of its bioactive compounds that enhance immunity and antioxidant capacity, dietary supplementation with fenugreek oil can effectively mitigate A. hydrophila-associated losses. AIM: This study aimed to explore the hematobiochemical, immune, antioxidant, and histopathological impacts of dietary fenugreek oil (FO) in Nile tilapia under normal culturing plus A. hydrophila infection. METHODS: Approximately 300 Nile tilapia fingerlings (

Isolation and characterization of Streptococcus agalactiae inducing mass mortalities in cultured Nile tilapia (Oreochromis niloticus) with trials for disease control using zinc oxide nanoparticles and ethanolic leaf extracts of some medicinal plants.

BACKGROUND: Streptococcus agalactiae (Group B streptococcus, GBS) induces a serious infection that can harm not only aquatic life but also humans and other animals. In a fish farm in southern Egypt, Nile tilapia (Oreochromis niloticus) has developed an epidemic with clinical symptoms resembling piscine streptococcosis. RESULTS: Initial microscopic inspection of the affected fish brain and kidney indicated the presence of Gram-positive cocci. S. agalactiae was effectively isolated and identified using nucleotide homology of the 16S rRNA and species-specific PCR. The partial 16S rRNA sequence w

Aquatic assessment of the chelating ability of Silica-stabilized magnetite nanocomposite to lead nitrate toxicity with emphasis to their impact on hepatorenal, oxidative stress, genotoxicity, histopathological, and bioaccumulation parameters in Oreochromis niloticus and Clarias gariepinus.

BACKGROUND: In recent years, anthropogenic activities have released heavy metals and polluted the aquatic environment. This study investigated the ability of the silica-stabilized magnetite (Si-M) nanocomposite materials to dispose of lead nitrate (Pb(NO3)2) toxicity in Nile tilapia and African catfish. RESULTS: Preliminary toxicity tests were conducted and determined the median lethal concentration (LC50) of lead nitrate (Pb(NO3)2) to Nile tilapia and African catfish to be 5 mg/l. The sublethal concentration, equivalent to 1/20 of the 96-hour LC50 Pb(NO3)2, was selected for our experiment. F

Preparation of the monoclonal antibody against Nile tilapia Igλ and study on the Igλ+ B cell subset in Nile tilapia.

Immunoglobulins (Igs) are important effector molecules that mediate humoral immunity. A typical Ig consists of two heavy and two light chains. In teleosts, three Ig heavy chain isotypes (Igμ, Igδ and Igτ) and three Ig light chain isotypes (Igκ, Igλ and Igσ) have been identified. Compared to the heavy chains, teleost Ig light chains have been poorly studied due to the lack of antibodies. In this study, a mouse anti-Nile tilapia Igλ monoclonal antibody (mAb) was prepared, which could specifically recognize Igλ in serum and Igλ+ B cells in tissues. Further, the composition of IgM+ and Igλ+ B cell

Deep evolutionary origin of limb and fin regeneration.

Salamanders and lungfishes are the only sarcopterygians (lobe-finned vertebrates) capable of paired appendage regeneration, regardless of the amputation level. Among actinopterygians (ray-finned fishes), regeneration after amputation at the fin endoskeleton has only been demonstrated in polypterid fishes (Cladistia). Whether this ability evolved independently in sarcopterygians and actinopterygians or has a common origin remains unknown. Here we combine fin regeneration assays and comparative RNA-sequencing (RNA-seq) analysis of Polypterus and axolotl blastemas to provide support for a common

Aripiprazole prevents stress-induced anxiety and social impairment, but impairs antipredatory behavior in zebrafish.

Environmental pollution caused by antipsychotic residues is a relevant ecological problem. Studies revealed that residues of these drugs are present in a wide range of different ecosystems and can have adverse effects on non-target organisms even in low environmental concentrations. Among these antipsychotic drugs, aripiprazole (APPZ) is a second-generation atypical antipsychotic that is a partial agonist of dopaminergic and serotoninergic receptors. APPZ is used to treat schizophrenia, bipolar disorder, autism, obsessive-compulsive disorder, and anxiety or panic disorders. Thus, in this study

Expression of immune-related genes of Nile tilapia Oreochromis niloticus after Gyrodactylus cichlidarum and Cichlidogyrus sclerosus infections demonstrating immunosupression in coinfection.

Gyrodactylus cichlidarum and Cichlidogyrus sclerosus, two monogenean ectoparasite species commonly found on the body surface and gills of Nile tilapia (Oreochromis niloticus) respectively, inflicted considerable economic losses in intensive tilapia farming. In order to explore the immune response of tilapia against these two species of monogeneans, expression patterns of five immune-related genes were studied after singular G. cichlidarum or C. sclerosus infection and their coinfection. The transcription levels of IL-1β were up-regulated in the skin after G. cichlidarum infection, reaching a p

Serum biomarker levels alter following nanoparticle (Al2O3, CuO, TiO2) exposures in freshwater fish (Oreochromis niloticus).

Nanoparticles (NPs) are used in diverse field of technology and consequently are released to the environment, most ending up in water bodies. Because NPs have potential to cause adverse effects on the metabolisms of animals, the present study was carried out to help understanding their effects on fish metabolism. In this study, freshwater fish (Oreochromis niloticus) were exposed to aluminum oxide (Al2O3) (40 nm), copper oxide (CuO) (40 nm) and titanium dioxide (TiO2) (21 nm) NPs in differing concentrations (0, 1, 5, 25 mg/L) for 14 d. Gene transfer and mutagenesis mediated by Sleeping Beauty transposon in Nile tilapia (Oreochromis niloticus).

The success of gene transfer has been demonstrated in many of vertebrate species, whereas the efficiency of producing transgenic animals remains pretty low due to the random integration of foreign genes into a recipient genome. The Sleeping Beauty (SB) transposon is able to improve the efficiency of gene transfer in zebrafish and mouse, but its activity in tilapia (Oreochromis niloticus) has yet to be characterized. Herein, we demonstrate the potential of using the SB transposon system as an effective tool for gene transfer and insertional mutagenesis in tilapia. A transgenic construct pT2/tiH

Estimation of biodiesel cytotoxicity by using acid phosphatase as a biomarker of lysosomal integrity.

Biodiesel is promoted as environmentally less harmful than diesel fuel. Nevertheless its water-soluble-fraction (WSF) may contain methanol, which appears by a reversion of the transesterification reaction, when biodiesel contacts water. This paper evaluated the loss of the lysosomal membrane integrity in liver homogenate of juvenils Tilapia exposed to biodiesels-WSF, through the increase of the acid phosphatase activity, as an evidence of citotoxicity. Differences in the enzyme activity levels (3.4, 2.3 and 0.8 mU mg(-1) total protein over the control value, which was 1.6 mU mg(-1) total prote

Impacts of encapsulated azadirachtin (Azadirachta indica) in the diet of Nile tilapia (Oreochromis niloticus).

Various plant-based products are currently undergoing testing in aquaculture as growth and/or health enhancers for fish, serving as substitutes for chemicals and antibiotics that may be detrimental to animal and consumer well-being. Azadirachtin is a bioactive compound extracted from the Azadirachta indica plant, with great microbicidal and immunostimulant potential. The aim of this study was to evaluate the effects of dietary azadirachtin supplementation on the zootechnical performance and health of juvenile Nile tilapia (Oreochromis niloticus) reared in a recirculating aquaculture system. Cocaine- and amphetamine-regulated transcripts in two percomorphs: evolutionary conservation and energy-status dependent responses.

Cocaine and amphetamine-regulated transcript (Cart) is a pleiotropic neuropeptide involved in appetite regulation and maintenance of energy homeostasis, among other processes. Although Cart is widely studied in vertebrates, its multigenic nature in fish raises questions about which functions are affected by specific Cart peptides. This study compared two important aquaculture percomorph species with distinct habitats and feeding behaviors: Nile tilapia (Oreochromis niloticus) and gilthead seabream (Sparus aurata). Bioinformatics revealed seven cart genes in tilapia and six in seabream that sho
