PURPOSE: To investigate the effect of hypoxia on proliferation and expression of HIF-1alpha and Caspase-3 in human periodontal ligament cells (PDLCs).
METHODS: Human PDLCs were exposed to cobalt chloride in order to mimic hypoxia. Cell viability of PDLCs was determined by MTT methods. Substitution of Co(III)-bound water by cyanide allows the rapid colorimetric detection of micromolar amounts of cyanide with cobalt corrinoids. Negatively charged side chains at the periphery of these metal-based sensors increase the selectivity of cyanide versus thiocyanate, the most disturbing anion, by a factor of 30 through additional supramolecular interactions in water.

BACKGROUND: We previously demonstrated that chronic hypoxia has pivotal roles in the progression of tubulointerstitial injury from the early stage of the uninephrectomized Thy1 nephritis model. We have also shown that pretreatment of cobalt confers renoprotection in the ischemia/reperfusion (I/R) injury, in association with the up-regulation of hypoxia-inducible factor (HIF)-regulated genes. Multiple sensitivity to metals is often seen in humans. This may be due to cross-reactivity or to multiple sensitization. The pattern of cross-reactivity to some metal compounds was studied, using the guinea pig maximization test method. Animals were induced with K2Cr2O7 and CoCl2, respectively, and then challenged with K2Cr2O7, CoCl2, NiSO4, PdCl2 and RhCl3. No simultaneous reactivity was recorded. Two chemical etching solutions were capable of providing micromechanical retention in two nickel-chromium-beryllium alloys and in a chromium-cobalt alloy. A resin matrix was used to verify the quality of etching on the metal surfaces. The chemical etching solutions created high microretentive surfaces in nickel-chromium-beryllium alloy but the chromium-cobalt alloy surfaces after etching were less retentive. Improved chemical etching technique should encourage expanded use of the resin-bonded retainers.

The aggregation of normal human platelets in vitro induced by ADP was severely inhibited after preincubation of platelet-rich plasma (PRP) with CoCl2. Platelets from a patient with congenital afibrinogenaemia did not aggregate until fibrinogen was added. This recovered response was also inhibited by CoCl2. In approaching the question as to which test preparations are most suitable for patch testing of metals, 25 chromium sensitive patients were tested with 0.5% potassium dichromate either in water or petrolatum. The aqueous solution proved better than the petrolatum preparation at a 48 h reading with Neodermtest Roc. No difference in sensitivity was recorded between 4 hexavalent chromium salts. 3 patients who were only sensitive to chromium solution in water also reacted positively to a 1% cobalt chloride solution and a 5% nickel sulfate solution in petrolatum.

The authors used the 57 Co urinary excretion test to assess the intestinal absorption of iron in anemia associated with hypoferrinemia and in iron overload states. Among the anemia associated with hypoferrinemia, a high elimination of 57 Co had been found in all patients (15) with post-haemorrhagic iron deficiency anemia.
