Tetrix: A novel Tetris-based paradigm for neuroimaging research and clinical applications.

Tetris is a widely used computer game, not only for entertainment purposes but increasingly in cognitive and clinical neuroscience research. Despite its frequent application in experiments, the neural mechanisms underlying gameplay remain insufficiently understood. Prior work suggests that cognitive control during complex visuospatial tasks, like Tetris, engages the attention network as well as regions supporting visuospatial working memory, mental imagery, and motor planning. However, researchers currently lack a standardized paradigm to investigate these processes. To address this gap, we in

Fear Learning and Memory in the Aging Brain: Neural Mechanisms of Posttraumatic Stress Disorder Risk and Resilience in Older Adults.

With a rapidly growing population of older adults worldwide, understanding how aging shapes mental and cognitive health is an urgent public health priority. Although older age has been considered protective against the development of posttraumatic stress disorder (PTSD), this pattern is far from universal. Emerging evidence indicates that older adults with a history of trauma may experience delayed-onset PTSD or a reemergence of symptoms after years of dormancy. In this review, we examine how age-related changes in the brain intersect with known PTSD mechanisms to influence trajectories of ris

Proposal for a Novel Classification of Patients With Enlarged Ventricles and Cognitive Impairment Based on Data-Driven Analysis of Neuroimaging Results in Patients With Psychiatric Disorders.

One of the challenges in diagnosing psychiatric disorders is that the results of biological and neuroscience research are not reflected in the diagnostic criteria. Thus, data-driven analyses incorporating biological and cross-disease perspectives, regardless of the diagnostic category, have recently been proposed. A data-driven clustering study based on subcortical volumes in 5604 subjects classified into four brain biotypes associated with cognitive/social functioning. Among the four brain biotypes identified in controls and patients with schizophrenia, bipolar disorder, major depressive diso

The Long and Winding Road to Real-Life Experiments: Remote Assessment of Executive Functions with Computerized Games-Results from 8 Years of Naturalistic Interventions.

Mate Marote is an open-access cognitive training software aimed at children between 4 and 8 years old. It consists of a set of computerized games specifically tailored to train and evaluate Executive Functions (EF), a class of processes critical for purposeful, goal-directed behavior, including working memory, planning, flexibility, and inhibitory control. Since 2008, several studies were performed with this software at children's own schools in interventions supervised in-person by cognitive scientists. After 2015, we incorporated naturalistic, yet controlled, interventions with children's ow

Triangulation of the neurocomputational architecture underpinning reading aloud.

The goal of cognitive neuroscience is to integrate cognitive models with knowledge about underlying neural machinery. This significant challenge was explored in relation to word reading, where sophisticated computational-cognitive models exist but have made limited contact with neural data. Using distortion-corrected functional MRI and dynamic causal modeling, we investigated the interactions between brain regions dedicated to orthographic, semantic, and phonological processing while participants read words aloud. We found that the lateral anterior temporal lobe exhibited increased activation …

Comparison of psychophysical, electrophysiological, and fMRI assessment of visual contrast responses in patients with schizophrenia.

Perception has been identified by the NIMH-sponsored Cognitive Neuroscience Treatment Research to Improve Cognition in Schizophrenia (CNTRICS) group as a useful domain for assessing cognitive deficits in patients with schizophrenia. Specific measures of contrast gain derived from recordings of steady-state visual evoked potentials (ssVEP) have demonstrated neural deficits within the visual pathways of patients with schizophrenia. Psychophysical measures of contrast sensitivity have also shown functional loss in these patients. In the current study, functional magnetic resonance imaging (fMRI) …

Revised associative inference paradigm confirms relational memory impairment in schizophrenia.

OBJECTIVE: Patients with schizophrenia have widespread cognitive impairments, with selective deficits in relational memory. We previously reported a differential relational memory deficit in schizophrenia using the Associative Inference Paradigm (AIP), a task suggested by the Cognitive Neuroscience Treatment Research to Improve Cognition in Schizophrenia (CNTRICS) initiative to examine relational memory. However, the AIP had limited feasibility for testing in schizophrenia because of high attrition of schizophrenia patients during training. Here we developed and tested a revised version of the…

Radiation-induced Brain Injury and the Radiation Late Effects Cohort (RLEC) of Rhesus Macaques (Macaca mulatta).

In the event of a nuclear accident or attack, thousands of people could receive high doses of total-body irradiation (TBI). Although retrospective analyses of atomic bomb and nuclear disaster survivors have been conducted, the long-term outcomes on the brain and cognitive function are conflicting. Radiation-induced brain injury (RIBI) is characterized by inflammation, vascular injury, deficits in neuronal function, and white matter (WM) injury, but the molecular mechanisms by which this occurs remain unknown. Animal models are crucial for evaluating radiation effects on the brain and have prov

Hippocampal pyramidal cells of the CA1 region are not a major target of the thalamic nucleus reuniens.

The prefrontal-hippocampal-entorhinal system is perhaps the most widely-studied circuit in cognitive and systems neuroscience, due to its role in supporting cognitive functions such as working memory and decision-making. Disrupted communication within this circuit is a key feature of disorders such as schizophrenia and dementia. Nucleus reuniens (NRe) is a midline thalamic nucleus that sits at the nexus of this circuit, linking these regions together. As there are no direct projections from prefrontal cortex to hippocampus (HPC), the accepted model is that the NRe mediates prefrontal drive of

A transdisciplinary view on curiosity beyond linguistic humans: animals, infants, and artificial intelligence.

Curiosity is a core driver for life-long learning, problem-solving and decision-making. In a broad sense, curiosity is defined as the intrinsically motivated acquisition of novel information. Despite a decades-long history of curiosity research and the earliest human theories arising from studies of laboratory rodents, curiosity has mainly been considered in two camps: 'linguistic human' and 'other'. This is despite psychology being heritable, and there are many continuities in cognitive capacities across the animal kingdom. Boundary-pushing cross-disciplinary debates on curiosity are lacking,

Individual predictors and electrophysiological signatures of working memory enhancement in aging.

A primary goal of translational neuroscience is to identify the neural mechanisms of age-related cognitive decline and develop protocols to maximally improve cognition. Here, we demonstrate how interventions that apply noninvasive neurostimulation to older adults improve working memory (WM). We found that one session of sham-controlled transcranial direct current stimulation (tDCS) selectively improved WM in older adults with more education, extending earlier work and underscoring the importance of identifying individual predictors of tDCS responsivity. Improvements in WM were associated with

The neuroecology of the water-to-land transition and the evolution of the vertebrate brain.

The water-to-land transition in vertebrate evolution offers an unusual opportunity to consider computational affordances of a new ecology for the brain. All sensory modalities are changed, particularly a greatly enlarged visual sensorium owing to air versus water as a medium, and expanded by mobile eyes and neck. The multiplication of limbs, as evolved to exploit aspects of life on land, is a comparable computational challenge. As the total mass of living organisms on land is a hundredfold larger than the mass underwater, computational improvements promise great rewards. In water, the midbrain

The Human Dynamic Clamp Reveals the Fronto-Parietal Network Linking Real-Time Social Coordination and Cognition.

How does the brain allow us to interact with others? Social neuroscience has already provided some answers to these questions but has tended to treat high-level, cognitive interpretations of social behavior separately from the sensorimotor mechanisms upon which they rely. The goal here is to identify the underlying neural processes and mechanisms linking sensorimotor coordination and intention attribution. We combine the human dynamic clamp, a novel paradigm for studyingrealistic social behavior, with high-resolution electroencephalography. The collection of humanness and intention attribution

Chapter 17: cognitive assessment in neurology.

Modern interests in cognitive assessment began with Franz Gall's early 19th century theory of mental organology and Paul Broca's reports in the 1860s on patients with focal brain injury and aphemia. These workers spurred interest in assessing delimited mental abilities in relation to discrete cerebral areas. With roots in experimental and educational psychology, the intelligence testing movement added assessment tools that could be applied to neurological patients. Early- to mid-20th-century landmarks were Alfred Binet and Theodore Simon's intelligence scale, Howard Knox's nonverbal performanc

Interoception Primes Emotional Processing: Multimodal Evidence from Neurodegeneration.

Recent frameworks in cognitive neuroscience and behavioral neurology underscore interoceptive priors as core modulators of negative emotions. However, the field lacks experimental designs manipulating the priming of emotions via interoception and exploring their multimodal signatures in neurodegenerative models. Here, we designed a novel task that involves interoceptive and control-exteroceptive priming conditions followed by post-interoception and post-exteroception facial emotion recognition (FER). We recruited 114 participants, including healthy controls (HCs) as well as patients with behav…

The insula: Leveraging cellular and systems-level research to better understand its roles in health and schizophrenia.

Schizophrenia is a highly heterogeneous disorder characterized by a multitude of complex and seemingly non-overlapping symptoms. The insular cortex has gained increasing attention in neuroscience and psychiatry due to its involvement in a diverse range of fundamental human experiences and behaviors. This review article provides an overview of the insula's cellular and anatomical organization, functional and structural connectivity, and functional significance. Focusing on specific insula subregions and using knowledge gained from humans and preclinical studies of insular tracings in non-human
