Purinergic signaling: A tribute to Francesco Di Virgilio.

This Special Issue (SI) is dedicated to the memory of Francesco Di Virgilio, an eminent and extraordinarily gifted scientist who unexpectedly passed away while attending a congress two years ago. Francesco left behind a remarkable legacy of discoveries in the field of purinergic signaling, with particular emphasis on the ATP-sensitive ionotropic P2X7 receptor (P2X7R). We never anticipated that our initiative to organize this SI would generate such overwhelming interest within the purinergic research community. The SI comprises 20 articles, including both original research papers and review art

From metabolic dysfunction-associated fatty liver disease to metabolic dysfunction-associated steatotic liver disease: Controversy and consensus.

The newly released nomenclature of metabolic dysfunction-associated steatotic liver disease (MASLD) in the 2023 European Association for the Study of the Liver Congress has raised great clinical concerns. This marks the second instance of significant renaming of non-alcoholic fatty liver disease since the introduction of metabolic dysfunction-associated fatty liver disease (MAFLD) in 2020. The nomenclature and definitions of MASLD and MAFLD exhibit significant disparities as well as substantial consensus. The disparities regarding the framework of nomenclature, the definitions, the clinical ma

Prophylaxis in von Willebrand disease with von Willebrand factor concentrate and nonfactor therapies.

This manuscript summarizes the current status of prophylaxis and novel potential therapies to prevent bleeding in patients with von Willebrand disease (VWD). VWD is the most common inherited bleeding disorder, which is associated mainly with mucocutaneous bleeding and bleeding during surgical and dental interventions. More severely affected VWD patients, mostly those with type 2 and type 3, can also suffer from joint, muscle, and gastrointestinal bleeds. Most patients with mild and moderate VWD are treated with desmopressin. The majority of patients with type 2 and 3 are treated with von Wille

[In search of an effective algorithm for rhythmic transcranial magnetic stimulation in neurorehabilitation after severe traumatic brain injury].

Rehabilitation of patients with severe traumatic brain injury (sTBI) is a topical medical and social issue because this pathology is one of the main causes of mortality and disability in the young working age population [1]. The most common sTBI consequences include motor and cognitive impairment as well as depression of consciousness [2, 3]. Despite significant progress in treatment of the consequences of severe traumatic brain injury, there are no treatment and rehabilitation standards for these patients, and the used rehabilitation measures are not always effective. These circumstances subs

Asthma in childhood.

Several topics on childhood asthma were addressed in the Paediatric Clinical Year in Review session at the 2015 European Respiratory Society International Congress. With regard to the relationship between lower respiratory tract infections and asthma, it emerges that is the number of respiratory episodes in the first years of life, but not the particular viral trigger, to be associated with later asthma development. Understanding which characteristics of individual patients are associated with an increased risk for asthma exacerbation is a critical step to implement strategies preventing these

Scientific Priorities Related to the Use of Double Sequential External Defibrillation in Patients With Refractory Cardiac Arrest: Report From a Multistakeholder Thinktank.

Ventricular fibrillation (VF) is common among patients with out-of-hospital cardiac arrest. Cardiac arrest with VF has a more favorable prognosis than those with other rhythms, but about 25% of patients with VF are still fibrillating after 3 attempts at defibrillation. Use of 2 defibrillators close together in time, "double sequential external defibrillation" (DSED), has been proposed to treat patients who remain in VF. The DOSE VF (Defibrillation Strategies for Refractory Ventricular Fibrillation) trial enrolled patients with out-of-hospital cardiac arrest with refractory VF to compare DSED v

Alzheimer's Disease-Related Dementias Summit 2019: National Research Priorities for the Investigation of Traumatic Brain Injury as a Risk Factor for Alzheimer's Disease and Related Dementias.

Traumatic brain injury (TBI) is a risk factor for later-life dementia. Clinical and pre-clinical studies have elucidated multiple mechanisms through which TBI may influence or exacerbate multiple pathological processes underlying Alzheimer's disease and Alzheimer's disease-related dementias (AD/ADRD). The National Institutes of Health hosts triennial ADRD Summits to inform a national research agenda, and the 2019 ADRD Summit was the first to highlight "TBI and AD/ADRD Risk" as an emerging topic in the field. A multidisciplinary committee of TBI researchers with relevant expertise reviewed exta

Update on the molecular pathogenesis and clinical treatment of Mantle Cell Lymphoma (MCL): minutes of the 9th European MCL Network conference.

Mantle cell lymphoma (MCL) is a distinct subtype of malignant lymphoma which is characterized by the chromosomal translocation t(11;14)(q13;q32), resulting in constitutional overexpression of cyclin D1 and cell cycle dysregulation in virtually all cases. Clinically, MCL shows an aggressive clinical course with a continuous relapse pattern and a median survival of only 3-5 years. However, recently a subset of 15% long-term survivors has been identified with a rather indolent clinical course, even after conventional treatment strategies only. Advanced stage disease is usually apparent already at
