Facile Preparation of Crosslinked PEG-Polypeptide Drug Nanocarriers Through Biphasic Polymerization of Crude N-Carboxyanhydrides.

Poly(ethylene glycol) (PEG)-polypeptide drug nanocarriers have garnered increasing attention during last decades. However, the tedious polymer synthesis and the stability issue greatly limit their further development. Herein the biphasic polymerization platform is reported for the easy access of a series of crosslinked PEG-polypeptide nanoparticles for enhanced drug loading and selective drug release. The accelerated polymerization allows for the preparation of a library of PEG-polypeptides from crude monomers with different polypeptide segments for structural optimization. Nanocarriers with r

Bioorthogonal Photocatalytic Protein Labeling and Cross-Linking Enabled by Stabilized Ketyl Radicals.

Radical reactions offer transformative potential in biological contexts but remain constrained by poor selectivity and off-target reactivity. We address these limitations through visible-light photocatalytic generation of diaryl ketyl radicals from benzophenones. This strategy circumvents traditional UV excitation pathways by suppressing triplet diradical formation─which drives nonspecific [2 + 2] cycloadditions and H atom abstraction─in favor of bioorthogonal radical-radical coupling. Our platform enables precise live-cell protein labeling with minimal cytotoxicity, including in sensitive pri

Calcium Cross-Linked Cellulose Nanofibrils: Hydrogel Design for Local and Controlled Nitric Oxide Release.

Nitric oxide (NO) holds promise for wound healing due to its antimicrobial properties and role in promoting vasodilation and tissue regeneration. The local delivery of NO to target cells or organs offers significant potential in numerous biomedical applications, especially when NO donors are integrated into nontoxic viscous matrices. This study presents the development of robust cellulose nanofibril (CNF) hydrogels designed to control the release of nitric oxide (NO) generated in situ from a NO-donor molecule (S-nitrosoglutathione, GSNO) obtained from the nitrosation of its precursor molecule

Prediction of Pentacam image after corneal cross-linking by linear interpolation technique and U-NET based 2D regression model.

Keratoconus is a common corneal disease that causes vision loss. In order to prevent the progression of the disease, the corneal cross-linking (CXL) treatment is applied. The follow-up of keratoconus after treatment is essential to predict the course of the disease and possible changes in the treatment. In this paper, a deep learning-based 2D regression method is proposed to predict the postoperative Pentacam map images of CXL-treated patients. New images are obtained by the linear interpolation augmentation method from the Pentacam images obtained before and after the CXL treatment. Augmented

Corneal Crosslinking: Present and Future.

Keratoconus is a progressive corneal thinning disorder that can lead to vision loss. In the last 2 decades, corneal crosslinking (CXL) has emerged as an effective method to halt the progression of keratoconus and reduce the number of patients requiring keratoplasty. The procedure has been adopted globally and has evolved to become a part of combination treatments to regularize the cornea and improve visual outcomes. CXL has even been extrapolated in managing other ocular pathologies such as progressive myopia, infectious keratitis, and bullous keratopathy. This review aims to summarize the cur

The efficiency and safety of oxygen-supplemented accelerated transepithelial corneal cross-linking.

PURPOSE: To investigate the impact of oxygen delivery on the clinical outcomes of accelerated transepithelial corneal cross-linking (A-TE CXL). METHODS: Fifty-seven eyes of 44 progressive keratoconus (KCN) patients were randomly separated into two age-sex-matched groups. Twenty-nine eyes of 23 KCN patients that underwent oxygen-supplemented A-TE CXL formed the study group and 28 eyes of 21 patients treated with the same procedure but under room air conditions formed the control group. All patients were examined preoperatively, one, six and twelve months after the procedure. The logMAR spectac

Update on corneal collagen crosslinking for ectasia.

PURPOSE OF REVIEW: Corneal collagen crosslinking (CXL) is a minimally invasive treatment that can stabilize corneal ectatic disorders including keratoconus, pellucid marginal degeneration, or postrefractive surgery ectasia. The benefits of CXL have been well documented. New research is focused on modifying current treatment protocols with the goals of maximizing corneal stability while also shortening overall procedure time. RECENT FINDINGS: Accelerated CXL protocols have the goal of delivering the same ultraviolet A intensity as conventional protocols, but over a shorter time period. Acceler

Changes and quantitative characterization of hyper-viscoelastic biomechanical properties for young corneal stroma after standard corneal cross-linking treatment with different ultraviolet-A energies.

Corneal collagen cross-linking (CXL) treatment can restore vision in patients suffering from keratoconus and corneal injury, by improving the mechanical properties of the cornea. The correlation between ultraviolet-A (UVA) irradiant energies of standard CXL (SCXL) and corneal visco-hyperelastic mechanical behavior remains unknown. In this study, SCXL with four different UVA irradiant energy doses (0-5.4 J/cm2) were administered as part of quantitative treatments of corneal stromal lenticules extracted from young myopic patients via small incision lenticule extraction (SMILE) corneal refractive

Cross-Linked Cyclodextrins Bimetallic Nanocatalysts: Applications in Microwave-Assisted Reductive Aminations.

The optimization of sustainable protocols for reductive amination has been a lingering challenge in green synthesis. In this context, a comparative study of different metal-loaded cross-linked cyclodextrins (CDs) were examined for the microwave (MW)-assisted reductive amination of aldehydes and ketones using either H2 or formic acid as a hydrogen source. The Pd/Cu heterogeneous nanocatalyst based on Pd (II) and Cu (I) salts embedded in a β-CD network was the most efficient in terms of yield and selectivity attained. In addition, the polymeric cross-linking avoided metal leaching, thus enhancin

The efficacy of standard versus accelerated epi-off corneal cross-linking protocols: a systematic review and sub-group analysis.

PURPOSE: To compare the one-year efficacy of accelerated and standard 5.4 J/cm2 protocols of cross-linking (CXL) in the treatment of progressive keratoconus. METHODS: In this systematic review, two members of the research team searched Scopus, Pubmed, ISI, Ovid, Science Direct, and Cochrane databases independently for publications between January 2010 and December 2016. The majority of retrieved studies were not randomized clinical trials (RCT), or the second arm of the RCT was either untreated or customized CXL. The outcomes of interest were uncorrected distance visual acuity, corrected visu

Collagen cross-linking impact on keratoconus extracellular matrix.

BACKGROUND: Keratoconus (KC) is a common multifactorial ectatic corneal disease with unknown onset. KC most commonly appears in adolescence and affects approximately 1:400 people worldwide. Treatment options, for advanced KC cases, are collagen cross-linking (CXL) and corneal transplants. CXL is a new KC treatment that helps arrest the disease. Unfortunately, only a fraction of KC patients will qualify for CXL treatment. Our goal, in this study, was to begin to understand how CXL affects the corneal microenvironment and pave the way towards a more patient-driven CXL treatment. METHODS: Primar

Optimization of an innovative vinylimidazole-based monolithic stationary phase and its use for pressured capillary electrochromatography.

A novel polymer monolith based on the dicationic crosslinker 3,3'-(hexane-1,6-diyl)bis(1-vinylimidazolium) bromide, the monomer 1-vinylimidazole and a ternary porogen mixture (1-propanol, decan-1-ol and water) was developed and optimized for capillary electrochromatography. This aim was accomplished by adjusting the composition of individual constituents in the polymerization mixture and monitored based on several relevant parameters (e.g. pore structure by scanning electron microscopy, generation of electroosmotic flow, or permeability of material). The ultimately selected composition yielded

[Impact of crosslinking on corneal epithelial healing].

INTRODUCTION AND AIM: To analyse the effect of crosslinking (CXL) on corneal epithelial healing. PATIENTS AND METHOD: 39 keratoconus patients following CXL and 11 patients following PRK were enrolled in this study. 21, 45 and 69 hours after CXL and 16, 37 and 61 hours following PRK slitlamp photographs were taken using fluorescein staining and the size of the epithelial defect and the time of epithelial closure was determined. As the observation time-points differed in both groups, the size of the epithelial defect following PRK was calculated for the time points used for analysis of CXL pati

Epithelium-off photochemical corneal collagen cross-linkage using riboflavin and ultraviolet a for keratoconus and keratectasia: a systematic review and meta-analysis.

This report presents the results of a systematic review and meta-analyses of studies on epithelium-off photochemical corneal collagen cross-linkage for the management of keratoconus and secondary ectasia. The literature search identified 3,400 records of which 49 were considered for inclusion in the meta-analyses. Eight papers reported 4 unique randomized controlled trials, 29 studies were prospective, and 12 were retrospective studies. The majority of the studies (39/49) were graded as very low quality evidence. Twenty-six studies described adverse events and were included in the safety analy

Differentiating untreated and cross-linked porcine corneas of the same measured stiffness with optical coherence elastography.

Structurally degenerative diseases, such as keratoconus, can significantly alter the stiffness of the cornea, directly affecting the quality of vision. Ultraviolet-induced collagen cross-linking (CXL) effectively increases corneal stiffness and is applied clinically to treat keratoconus. However, measured corneal stiffness is also influenced by intraocular pressure (IOP). Therefore, experimentally measured changes in corneal stiffness may be attributable to the effects of CXL, changes in IOP, or both. We present a noninvasive measurement method using phase-stabilized swept-source optical coher
