BACKGROUND: Experimental data suggest that there is an imbalance between Th1 and Th2 cells in atopic dermatitis (AD) skin compared to allergic contact dermatitis (ACD). This imbalance (Th2 and Th1 predominance, respectively) implies the production of different cytokines in these two conditions leading to different expression of adhesion molecules on skin endothelial cells.
OBJECTIVE: The expression of VCAM-1 (IL-4/Th2-dependent) and ICAM-1 (INF-gamma/IL-1) on dermal vessels was compared in six patients with AD and six patients with ACD. The effect of cetirizine, a highly selective H1-receptor

Cutaneous lymphocyte-associated antigen (CLA) is a cell surface glycoprotein which has been implicated in the homing of lymphocytes to cutaneous sites. It is postulated to play an important role in T-cell migration to skin in atopic dermatitis; however, the expression of CLA in both normal children and children with atopic dermatitis has not been extensively studied. If CLA expression on T cells were important in the traffic of lymphocytes to atopic dermatitis skin lesions, it might be expected that the proportion of CLA+ T cells in unstimulated peripheral blood from children with atopic derma

BACKGROUND: Atopic dermatitis (AD) and allergic contact dermatitis (ACD) are inflammatory skin conditions whose association is not clearly defined.
OBJECTIVES: To identify differences in ACD profile between patients with and without AD among those referred for patch testing. Additionally, to determine the prevalence of sensitisation to standard Spanish contact allergens in both groups.
METHODS: We analysed two groups (AD and non-AD) within the Spanish Registry of Research in Contact Dermatitis and Cutaneous Allergy (REIDAC). Contact allergy, clinical relevance and epidemiological data were c

PURPOSE: To investigate the relationship between the cytokine levels in the tear fluid and the severity of ocular surface disease in patients with atopic dermatitis (AD) and atopic keratoconjunctivitis (AKC).
METHODS: Tear fluid was collected from 104 AD patients with stratified AKC severity and the healthy controls, who all underwent ophthalmological examination. Cytokine levels in tear fluid were analyzed with Luminex multiplex assays.
RESULTS: Increased levels of eotaxin-3, IFN-β, IFN-γ, IL-1β, IL-13, IL-15, IL-17A, IL-2, IL-4, IL-6, IL-8, MCP-4, MIP-1α, MIP-1β, MMP-9, MPIF-1 and MPO were

BACKGROUND: Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by persistent itching, dryness, redness, and recurrent eczematous lesions, significantly impacting patients' quality of life and psychological well-being. Despite various effective conventional treatments, innovative therapeutic approaches that simultaneously provide anti-inflammatory effects and enhanced skin hydration remain essential.
OBJECTIVE: This case-series study aimed to evaluate the clinical efficacy, patient satisfaction, and safety profile of polynucleotide (PN)-based injectable therapies in pa

INTRODUCTION: Atopic dermatitis (AD) is the most common chronic inflammatory skin disease and is clinically and biologically heterogeneous. Its diagnosis remains clinical, supported by validated criteria, with no single confirmatory test, yet the expansion of targeted systemic therapies has intensified the need for objective tools to support diagnosis and stratify patients.
AREAS COVERED: This review summarizes the established clinical framework of AD diagnosis and the emerging tools intended to complement it: circulating and skin-derived biomarkers, minimally invasive tape stripping, the ski

IMPORTANCE: The exposome has been proposed to have a key role in the development of atopic dermatitis (AD). However, whether early-life infections are associated with AD risk remains unclear.
OBJECTIVE: To investigate whether early infection burden and infection subtypes are associated with the risk of childhood AD.
DESIGN, SETTING, AND PARTICIPANTS: A cohort study using data from children aged 0 to 10 years from the 2010 Danish Copenhagen Prospective Studies on Asthma in Childhood (COPSAC2010) birth cohort who were monitored for infection diagnoses, AD diagnosis, Scoring Atopic Dermatitis (

BACKGROUND: Atopic dermatitis (AD) is primarily driven by a Type 2 immune response, with T helper (TH2) cells producing IL-4 and IL-13, thereby promoting inflammation, itch, and a compromised skin barrier. Yet, the spatial organization of pathogenic immune cells and their interactions with stromal and epithelial compartments in human AD skin remain incompletely understood.
METHODS: We performed 10× Genomics Visium spatial transcriptomics on FFPE skin biopsies from patients with AD (n = 6), psoriasis (n = 2), and healthy controls (n = 5). Data were integrated with AD single-cell RNA sequencing

BACKGROUND: Atopic dermatitis (AD) is the most common chronic inflammatory skin disease. Its pathogenesis is closely related to the imbalance of immune homeostasis regulation. Accumulating evidence demonstrated that nitric oxide (NO) serves a critical role in modulating skin immune homeostasis during the development of multiple inflammatory skin diseases. NO is mainly synthesized by inducible nitric oxide synthase (iNOS); however, the mechanism underlying iNOS in AD has not been fully elucidated yet. The present study aimed to investigate the specific mechanisms by which iNOS regulates the dev

Atopic dermatitis (AD) is a common skin disease. Although AD pathogenesis has been widely researched, inhibitory mechanisms in AD are still unclear. Sialic acid-binding immunoglobulin-like lectins (Siglecs) are a family of receptors recognising sialic acids; most Siglecs work as inhibitory receptors. Among Siglecs, Siglec-E is expressed on dendritic cells (DCs) and eosinophils, important immune cells in AD. Although Siglec-E inhibits Type 1 inflammatory diseases, how it influences AD is unknown. Thus, we investigated the role of Siglec-E in AD mouse model by using Siglec-E knockout (KO) mice.

Atopic dermatitis (AD) is a prominent inflammatory skin disorder that significantly affects daily functioning and mental health in patients. Among its psychological repercussions, besides anxiety and depression, obsessive-compulsive disorder (OCD) has emerged. Multiple studies have indicated an increased risk of OCD in patients with AD. This systematic review aims to synthesize and evaluate the current literature on the association between AD and OCD, with particular attention to mechanisms linking disease and integrative treatment approaches. Atopic dermatitis is a risk factor for venous thromboembolism which may be the first manifestation of occult cancer. We examined whether a venous thromboembolism in patients with atopic dermatitis is a marker of occult cancer. We used Danish health registries to conduct this population-based cohort study. Patients with a first-time diagnosis of venous thromboembolism and a history of atopic dermatitis were identified from the Danish National Patients Registry from 1980 through 2022. We calculated the absolute risk of cancer treating death as a competing event. As a measure of relative risk, we

Atopic dermatitis (AD), or eczema, is a common, chronic inflammatory skin condition resulting in recurrent flares of itch and rash. This review discusses current literature regarding epidemiology, pathophysiology, etiology, comorbidities, clinical presentation, and impact on quality of life with the goal of addressing common gaps in understanding. Onset is usually early in life, but it is not uncommon to persist into adulthood, despite a common assumption that children outgrow AD. The primary drivers of its pathophysiology are skin barrier dysfunction and immune dysregulation, which are comple
