Aspirin cooperates with p300 to activate the acetylation of H3K9 and promote FasL-mediated apoptosis of cancer stem-like cells in colorectal cancer.

Cancer stem-like cells (CSCs) have been proposed as a key driving force of tumor growth and relapse in colorectal cancer (CRC), and therefore, they are promising targets for cancer therapy. Epidemiological evidence has suggested that the daily use of aspirin reduces overall mortality of CRC and the risk of distant metastasis. We investigated the effect and mechanism of aspirin on CSCs in CRC. Methods: The ratio of CSCs was analyzed after aspirin treatment both in a cell model and patient samples. Chemically modified aspirin and immunoprecipitation were adopted to detect the target proteins of

Revisiting fluoroquinolone prophylaxis in haematological malignancies and hematopoietic cell transplantation: A decade of evidence and gaps in understanding the impact of gut colonization with fluoroquinolone-resistant Enterobacterales.

OBJECTIVE: Rising antimicrobial resistance necessitates reevaluation of fluoroquinolone (FQ) prophylaxis (FQP) in patients with hematologic malignancies undergoing intensive chemotherapy or hematopoietic cell transplantation (HCT). This review evaluated the clinical efficacy of FQP and the modifying effect of gut colonization with FQ-resistant Enterobacterales (FQRE). METHODS: A structured narrative review of studies published between 2015 and 2025 was conducted. Studies comparing FQP with no prophylaxis and studies evaluating outcomes according to baseline colonization with FQRE were reviewe

The growing burden of myopia in Europe: evidence from population-based cohorts.

Myopia is a childhood-onset condition driven by excessive axial ocular elongation, associated with increased risk of sight-threatening complications and irreversible visual impairment. Despite rising global prevalence, epidemiological data on long-term trends in Europe remain limited. This study examines temporal trends in myopia prevalence and axial length (AL) across European adult birth cohorts. We conducted a comparative birth-cohort analysis using harmonised data from 28 population-based studies (n = 81,828) across nine countries participating in the European Eye Epidemiology (E3) Consort

Rethinking gastric carcinogenesis: a multiscale ecological model of risk beyond Helicobacter pylori.

Gastric cancer remains one of the leading causes of cancer mortality worldwide, characterized by marked geographic disparities that cannot be fully explained by the distribution of Helicobacter pylori infection alone. Although H. pylori is recognized as the principal etiological agent, reductionist models centered exclusively on infection prevalence and eradication fail to account for the complex heterogeneity of gastric cancer risk across populations. In this narrative review, we propose a multiscale ecological framework that integrates infectious, host, environmental, microbial, and socioeco

Beyond Compensation: Hyperinsulinemia as an Early Driver of Cardiovascular-Kidney-Metabolic Syndrome.

Background. Cardiovascular-kidney-metabolic (CKM) syndrome encompasses obesity, type 2 diabetes, cardiovascular disease, and chronic kidney disease. Insulin resistance has traditionally been considered the primary driver, with hyperinsulinemia viewed as a compensatory response. However, emerging evidence suggests that hyperinsulinemia may precede insulin resistance in specific phenotypes and contribute to early disease mechanisms. Methods. We conducted a narrative review of the role of hyperinsulinemia in CKM syndrome. Literature searches were performed in PubMed/MEDLINE, Embase, and Scopus th

Trends and burden of colon and rectum cancer attributable to low physical activity in China: a comprehensive analysis from 1990 to 2021.

BACKGROUND: Colon and rectum cancer (CRC) is an increasing public health concern in China due to rising incidence and disability, particularly among aging populations. Despite medical advancements, low physical activity remains a major modifiable risk factor. This study evaluates CRC burden trends from 1990 to 2021, focusing on demographic and temporal effects. METHODS: Using age-standardized rates for mortality, disability-adjusted life years (DALYs), years lived with disability (YLDs), and years of life lost (YLLs), this retrospective analysis employed Joinpoint regression, decomposition, a

Hearing Loss and Dementia: Risk Factor, Early Marker, or Both?

Background/Objectives: Hearing loss and dementia are highly prevalent conditions in older adults and represent a growing public health challenge. Over the past decade, a substantial body of epidemiological evidence has demonstrated a consistent association between age-related hearing loss and cognitive dysfunction, including incident dementia. However, the nature of this relationship remains incompletely understood. Methods: This narrative review provides a structured overview of current evidence, focusing on epidemiological findings, mechanistic pathways, and clinical implications. Hearing lo

Preventing childhood obesity: what works?

Rates of overweight in North American children and adolescents have increased dramatically since the 1970s. Childhood obesity has reached epidemic proportions and calls for prevention and treatment programs to reverse this trend have been made. However, the evidence base needed for effective action is still incomplete, especially for childhood obesity prevention programs. This paper focuses on primary prevention of childhood obesity and has three aims: (1) to briefly describe current primary prevention approaches for childhood obesity and the evidence for their impact; (2) to elucidate promisi

What's happening? The expanding role of apheresis platelet support in neonatal alloimmune thrombocytopenia: current status and future trends.

Despite some unresolved problems that may be associated with platelet transfusion, such as alloimmunisation, refractoriness, bacterial contamination, and the potential side effects related to the development of some biological response modifiers during storage, platelet therapy remains the most effective treatment for the management and prevention of severe thrombocytopenia and hemorrhage [Seghatchian J, Snyder EL, Krailadsiri P, editors. Effect modification of maternal concentrations of per- and polyfluoroalkyl substances on vaccine effectiveness in children up to 6 years of age: The Japan Environment and Children's Study.

Epidemiological evidence suggests that per- and polyfluoroalkyl substances (PFAS) exposure may attenuate antibody responses to vaccination. However, vaccine effectiveness as an outcome has not been investigated. This study assessed whether maternal PFAS concentrations during pregnancy modified childhood vaccine effectiveness. We analyzed mother-child pairs from the Japan Environment and Children's Study. Children's vaccination status for varicella, mumps, and influenza was collected via self-reported questionnaires at 2 years of age. Subsequent infection incidence was similarly assessed until

Periodontitis and Parkinson's disease: a narrative review of associations, proposed mechanisms, and clinical considerations.

Periodontitis is a common chronic inflammatory disease, and oral health problems are frequent in people with Parkinson's disease (PD). Observational studies consistently indicate poorer periodontal status in PD, but evidence that periodontitis increases the incidence or progression of PD is inconsistent and does not establish causality. This narrative review summarizes current evidence on epidemiological associations, biologically plausible mechanisms, and dental management considerations. Human data support an association between PD and greater periodontal disease burden, plausibly related to

Loneliness and cognitive aging: a brain-heart axis perspective on a modifiable risk.

Loneliness, the subjective perception of inadequate social connection, is increasingly recognized as a significant and modifiable determinant of health with implications extending beyond psychosocial well-being to cardiovascular and cognitive outcomes. A growing body of evidence links loneliness to increased risk of cardiovascular disease, cognitive decline, and dementia; however, these domains have often been studied in parallel. This review advances a brain-heart axis framework to integrate these literature, conceptualizing loneliness as a chronic psychosocial stressor that contributes to co

Habitat-Mediated Spillover Risk of Andes Hantavirus in Oligoryzomys longicaudatus: A Mechanistic Eco-Epidemiological Model.

Andes hantavirus (ANDV) is a rodent-borne orthohantavirus associated with hantavirus cardiopulmonary syndrome in southern South America. Its maintenance and spillover risk depend on the ecology of its principal reservoir, Oligoryzomys longicaudatus, and on environmental changes that alter habitat availability, host abundance, and human-rodent interfaces. We developed a mechanistic eco-epidemiological model that couples effective habitat cover to an SIR framework for ANDV transmission in O. longicaudatus. Habitat degradation and compensatory restoration modify rodent carrying capacity, natality

F-erythrocytes promote Plasmodium falciparum proliferation in sickle cell disease.

BACKGROUND: Sickle cell disease (SCD) remains prevalent because heterozygous carriers (HbAS) are partially resistant to Plasmodium falciparum malaria. Sickle hemoglobin (HbS) polymerization in low and intermediate oxygen (O2 ) conditions is the main driver of HbAS-driven resistance to P. falciparum malaria. However, epidemiological studies have reported mixed malaria morbidity and mortality outcomes in individuals with sickle cell disease (SCD). While maximum-tolerated dose hydroxyurea has been shown to lower malaria incidence, fetal hemoglobin (HbF), an inhibitor of HbS polymerization that is

Pathophysiologic Mechanisms of Tobacco Smoke Producing Atherosclerosis.

INTRODUCTION: Despite the convincing epidemiologic association between smoking and vascular disease, the pathophysiologic mechanisms by which smoking initiates and contributes to the progression of atherosclerosis remain incompletely understood. A precise dose-dependent correlation has never been demonstrated, suggesting that the biological relationship is complex and influenced by individual genetic and possibly environmental factors. Although endothelial dysfunction and intimal damage appear to be central to atherogenesis, how tobacco products cause this effect has not been established. The

Thyroid Eye Disease: How A Novel Therapy May Change The Treatment Paradigm.

Thyroid eye disease (TED) is a complex, debilitating autoimmune disease that causes orbital inflammation and tissue remodeling, resulting in proptosis, diplopia, and in severe cases, loss of vision. TED can lead to facial disfigurement and severely impact patients' quality of life. Although the course of TED was identified over 60 years ago, effective treatment options have proved to be challenging. Current treatments such as glucocorticoid therapy and orbital radiation focus on reducing orbital inflammation. However, these therapies fail to modify the disease outcomes, including proptosis and

The Influence of Age on the Relationship Between Allergic Rhinitis and Otitis Media.

PURPOSE OF REVIEW: To examine the relationship between otitis media, allergic rhinitis, and age. RECENT FINDINGS: Otitis media and allergic rhinitis are prevalent conditions with a controversial relationship. Some data suggest that these entities are significantly associated, either through allergic rhinitis inducing Eustachian tube dysfunction or through allergic pathophysiology simultaneously occurring intranasally and in the ear. Other studies, however, have refuted this relationship. For example, treatment with antihistamines does not reliably improve OME, making causation and association

The Impact of Ventilation and Early Diagnosis on Tuberculosis Transmission in Brazilian Prisons.

Prisoners have among the highest incidence of tuberculosis (TB) globally. However, the contribution of the prison environment on transmission is not well understood and structural characteristics have received little attention as effective epidemiological interventions in TB control. We evaluated architectural characteristics and estimated ventilation rates in 141 cells in three prisons in central west Brazil using steady-state exhaled carbon dioxide (CO2) levels. We used a modified Wells-Riley equation to estimate the probability of infection for inmates sharing a cell with an infectious case

Accuracy and limitations of the diagnosis of malnutrition in dialysis patients.

Uremic malnutrition, also known as protein-energy wasting (PEW), is a common phenomenon in maintenance dialysis patients and a risk factor for poor clinical outcomes including worse quality of life and increased hospitalization and mortality. The paradoxical association between traditional cardiovascular risk factors and better outcomes in dialysis patients also referred to as "reverse epidemiology," is a good example of the powerful effect-modifying impact of the nutritional status in this population. Measures of food intake, body composition tools, nutritional scoring systems, and laboratory

HIV/hepatitis C virus co-infection in drug users: risk behavior and prevention.

Studies of HIV-positive patients have consistently shown that drug users, in particular injection drug users (IDU), are far more likely to have hepatitis C virus (HCV) infection than other patient groups. HIV incidence and prevalence in IDU has declined in recent years, but HCV remains endemic in this population. HCV antibody prevalence among non-injection users of drugs such as heroin and cocaine is between 5 and 30%, although there are scant data on specific transmission risk behavior. The control of HIV/HCV co-infection must address HCV prevention. Epidemiological studies have suggested tha
