BACKGROUND AND AIMS: SCO2 encodes a mitochondrial copper chaperone required for cytochrome c oxidase (COX) assembly and is classically associated with severe multisystem mitochondrial disease. We characterize a motor-predominant axonal neuropathy presentation associated with biallelic SCO2 variants.
METHODS: Clinical, genetic, and functional studies were performed in a 15-year-old female presenting with axonal neuropathy. Functional studies were conducted in patient-derived fibroblasts, including Western blot analysis and spectrophotometric cytochrome c oxidation assay. Structural modeling wa

Invasive carnivores such as the raccoon (Procyon lotor) and raccoon dog (Nyctereutes procyonoides) are widespread in Europe and may contribute to the circulation of nematodes of veterinary and zoonotic relevance at the wildlife-domestic animal interface. Molecular data on nematode taxa infecting these hosts in Central Europe remain scarce, particularly those derived from faecal DNA. We analysed 212 post-mortem faecal samples collected in western Poland (97 raccoons, 115 raccoon dogs) using PCR-based screening followed by mitochondrial cytochrome c oxidase subunit 1 (cox1) sequencing for molecu

Posthodiplostomum minimum (MacCallum, 1921) Dubois, 1936 (Digenea, Diplostomidae) was described from adult worms from a great blue heron (Ardea herodias; Ardeidae) in New York. Metacercariae have been widely reported from diverse freshwater fishes, despite early experimental work and recent molecular data indicating exclusive infection of minnows (Leuciscidae) in North America. We compared the type specimens of P. minimum to adults taken from domestic chicks (Gallus gallus domesticus) fed metacercariae from fathead minnow (Pimephales promelas; Leuciscidae), as well as adults in naturally infec

The etiological agents of cystic echinococcosis comprise a species complex, Echinococcus granulosus sensu lato (s. l.). Despite high endemicity, the disease remains neglected in most parts of the globe, including India. The present study aimed at investigating the genotypes of Echinococcus granulosus s. l. circulating in sheep and goats in two states (Haryana and Himachal Pradesh) and a union territory (Chandigarh) of north India. Out of 1049 slaughtered small ruminants (n = 314, 394, and 341 from Chandigarh, Himachal Pradesh, and Haryana, respectively), 85 (n = 53, 17, and 15 from Chandigarh,

Dengue virus (DENV) is an expanding global health threat, yet its ecological and entomological drivers remain poorly defined in sub-Saharan Africa. In Kenya, recurrent outbreaks along the coastal region contrast sharply with the persistent absence of outbreaks in ecologically suitable western Kenya. Understanding the spatial heterogeneity of vector populations, viral circulation, and ecological context is essential for identifying under-recognized determinants of dengue emergence. We conducted a comparative entomological and virological survey across outbreak-prone coastal Kenya and the low-ou

COXFA4L3 is a testis-specific cytochrome c oxidase subunit that enhances mitochondrial complex IV activity during spermatogenesis. From the analysis of Coxfa4l3 knockout mice, the isoform switch from COXFA4 to COXFA4L3 may increase the potential COX activity, although this activity does not appear in the testis. This latent enhancement becomes evident in sperm, where COXFA4L3 promotes higher respiratory capacity, increasing sperm motility and ATP production. These findings indicate that COXFA4L3 is a key regulator of mitochondrial energy metabolism and may provide insights into the mechanisms

Taenia talicei is a cestode endemic to South America that was originally described based on its metacestodes. The adult strobilus was described after being obtained through experimental infection in dogs. Despite this, the natural definitive host for this cestode, as well as its genetic information, remained unknown. The objective of this study is to investigate the role of the domestic dog, the Andean fox (Lycalopex culpaeus), and other species of rodents in the life cycle of T. talicei. Molecular analysis of the mitochondrial genes of cytochrome c oxidase subunit 1 (cox1) and NADH dehydrogen

Tick infestation represents a significant constraint on livestock productivity in Saudi Arabia; however, there remains a substantial gap in research addressing tick species diversity, distribution, and their direct effects on milk production. This study aimed to morphologically and molecularly identify tick species infesting dairy cattle, quantify the impact of tick infestation on milk yield and composition, and contribute to the limited understanding of tick ecology and its economic implications in the region. Ticks were collected from infested cows and identified morphologically using taxono

The Asian blue tick, Rhipicephalus (Boophilus) microplus (Canestrini, 1888) is renowned as a notorious pest of veterinary importance globally. Its recent introduction to West Africa and subsequent eastward invasion and displacement of native tick fauna in many African countries is alarming, considering the prevalent unrestricted livestock migration in these regions. This study determined the genetic diversity of R. microplus in Africa based on the mitochondrial cytochrome oxidase subunit one (cox1) gene sequences in comparison with global sequences. Phylogenetic and genetic diversity analyses

Photobiomodulation (PBM), previously known as low-level laser light therapy, represents a noninvasive form of phototherapy that utilizes wavelengths in the red light (RL, 620-700 nm) portion of the visible light (VL, 400-700 nm) spectrum and the near-infrared (NIR, 700-1440 nm) spectrum. PBM is a promising and increasingly used therapy for the treatment of various dermatologic and nondermatologic conditions. Photons from RL and NIR are absorbed by endogenous photoreceptors including mitochondrial cytochrome C oxidase (COX). Activation of COX leads to the following changes: modulation of mitoch

BACKGROUND: Deer-derived materials (antler, venison, fetus, penis, bone, tail, and others) are some of the most valuable traditional animal-based medicinal and food materials in China. In production, processing, and trade, the quality of deer products varies. The market is confusing, and counterfeit and shoddy products are common. There is an urgent need to establish an accurate identification method.
RESULTS: Two pairs of primers suitable for identifying deer-derived medicinal materials were obtained by screening the cytochrome oxidase I (COI) sequences of 18 species from nine genera of the

Cystic echinococcosis (CE) is a disease that can be transmitted from animals to humans, caused by the metacestode of Echinococcus granulosus. The disease has significant health and economic impacts worldwide, particularly in endemic areas. The study aimed to evaluate the prevalence of hydatid cysts in ruminants (cattle and sheep) (n = 2060) from the Setif Province of Algeria using microscopy. The results showed that hydatid cysts were detected in 9.6% (198/2060) of ruminants, with a higher prevalence in cattle (16.8%; 56/333) compared to sheep (8.2%; 142/1727). Molecular techniques were used t

Although hydrogen sulfide (H2S) is an endogenous signaling molecule with antioxidant properties, it is also cytotoxic by potently inhibiting cytochrome c oxidase and mitochondrial respiration. Paradoxically, the primary route of H2S detoxification is thought to occur inside the mitochondrial matrix via a series of relatively slow enzymatic reactions that are unlikely to compete with its rapid inhibition of cytochrome c oxidase. Therefore, alternative or complementary cellular mechanisms of H2S detoxification are predicted to exist. Here, superoxide dismutase [Cu-Zn] (SOD1) is shown to be an ef

Low-level laser therapy, or photobiomodulation, utilizes red or near-infrared light for the treatment of pathological conditions due to the presence of intracellular photoacceptors, such as mitochondrial cytochrome c oxidase, that serve as intermediates for the therapeutic effects. We present an in-detail analysis of the effect of low-intensity LED red light irradiation on the respiratory chain of brain mitochondria. We tested whether low-level laser therapy at 650 nm could alleviate the brain mitochondrial dysfunction in the model of acute hypobaric hypoxia in mice. The irradiation of the mit

Assembly factors play a critical role in the biogenesis of mitochondrial respiratory chain complexes I-IV where they assist in the membrane insertion of subunits, attachment of co-factors, and stabilization of assembly intermediates. The major fraction of complexes I, III and IV are present together in large molecular structures known as respiratory chain supercomplexes. Several assembly factors have been proposed as required for supercomplex assembly, including the hypoxia inducible gene 1 domain family member HIGD2A. Using gene-edited human cell lines and extensive steady state, translation

Mosquitoes are commonly identified to species level using morphological traits, but complementary methods for identification are often necessary when specimens are collected as immature stages, stored inadequately, or when delineation of species complexes is problematic. DNA-barcoding using the mitochondrial cytochrome c oxidase subunit 1 (COI) gene is one such tool used for the morphological identification of species. A comprehensive entomological survey of mosquito species in Mexico State identified by COI DNA barcoding and morphology is documented in this paper. Specimens were collected fro

Perinatal hypoxic ischaemic (HI) encephalopathy is associated with severe neurodevelopment problems and mortality. This study uses broadband continuous-wave near-infrared spectroscopy (NIRS) to assess the early changes in cerebral oxygenation and metabolism after HI injury in an animal model using controlled anoxia events. Anoxia was induced before and 1 h after various levels of HI injury to assess the metabolic response via the changes in the oxidation state of cytochrome-c-oxidase (oxCCO), a marker of oxidative metabolism. The oxCCO responses to anoxia were classified into five categories:
