Comparative in vitro activity of new beta-lactam antibiotics against anaerobic bacteria.

Several new beta-lactam antimicrobial agents have been introduced in the last few years. In this investigation, the in vitro activities of several recently introduced cephalosporins (cefoperazone, cefotaxime, ceftazidime, and ceftizoxime), moxalactam, and N-formimidoyl thienamycin were compared with those of cefoxitin, clindamycin, and metronidazole against 203 strains of anaerobic bacteria. At achievable serum levels, all of the antimicrobial agents were active against essentially 100% of the strains of anaerobic gram-positive cocci, Clostridium perfringens, Leptotrichia buccalis, and species

Multi-aspect therapeutic effects of a polyphenolic herbal formulation Cirsium japonicum, Scutellaria baicalensis, Paeonia japonica, and Glycyrrhiza uralensis on ulcerative colitis: inflammation modulation, gut microbiota remodeling, and metabolite profiling.

Ulcerative Colitis (UC) is a chronic illness that commonly demands the use of medication, sometimes for long term. In a DSS mouse model, we examined 5-aminosalicylic acid (5ASA) in comparison to a defined polyphenol-rich herbal mixture CSPG: Cirsium japonicum, Scutellaria baicalensis, Paeonia japonica, and Glycyrrhizae radix, using a two-phase approach. In phase 1 (days 1-14, without DSS stimulation), the herbal formula CSPG produced a more gut-friendly preventive profile compared to 5ASA in non-inflammatory condition:Unlike 5-ASA, which decreases microbial diversity as previously reported, CS

Dihydroartemisinin alleviates ulcerative colitis by modulating gut microbiota and butyrate to restore Treg/Th17 balance and intestinal barrier function.

Accumulating evidence suggests that dihydroartemisinin (DHA), a natural compound known for its anti-inflammatory and immunomodulatory properties, holds therapeutic potential for ulcerative colitis (UC). However, its mechanism of action, particularly concerning the gut microbiota and its metabolites, remains incompletely understood. In this study, we evaluated the therapeutic effects of dihydroartemisinin in a dextran sulfate sodium (DSS)-induced murine model of UC. Dihydroartemisinin was administered at doses of 10, 20, and 50 mg/kg, with 5-aminosalicylic acid (5-ASA, 50 mg/kg) serving as a po

Based on untargeted metabolomics and metagenomics: a study on the mechanism of Miao ethnomedicine Zingiber mioga (Thunb.) Rosc. in treating slow transit constipation.

INTRODUCTION: Slow transit constipation (STC) is a prevalent gastrointestinal disorder characterized by impaired intestinal motility, metabolic dysregulation, and gut microbial dysbiosis. Zingiber mioga (Thunb.) Rosc. (RH), a traditional medicinal-edible plant, is empirically used to alleviate gastrointestinal dysfunction, but its therapeutic mechanisms in STC remain unclear. Herein, we investigated the laxative efficacy and mechanism of RH in a rat STC model via integrated untargeted metabolomic and metagenomic analyses, providing experimental evidence for its clinical use. METHODS: A rat ST

Effects of Compound Yeast Culture and Yeast Cell Wall Polysaccharide on Intestinal Barrier Function in Mongolian Ram Lambs.

During the early growth stage, lambs are highly susceptible to pathogenic microbial invasion due to an underdeveloped intestinal structure, unstable microbial colonization, and immature mucosal immune function, leading to diarrhea, growth retardation, and elevated mortality factors that severely constrain the production efficiency and economic viability of the sheep industry. This study aimed to compare the regulatory effects of compound yeast culture (CYC) and yeast polysaccharides (YPs) on intestinal barrier function in Mongolian male lambs and clarify their underlying molecular mechanisms.

Washed microbiota transplantation relieves atopic dermatitis via gut-skin microbiome rebalancing.

BACKGROUND: Atopic dermatitis (AD) is a chronic, relapsing inflammatory skin disease in which dysbiosis of gut and skin microbiota contributes to pathogenesis and severity. Washed microbiota transplantation (WMT)-an improved form of fecal microbiota transplantation with enhanced safety and microbiota quality control-has shown efficacy in a single reported adolescent case. However, clinical data on WMT in AD and its effects on the skin and gut microbiota remain limited. METHODS: Twenty-three patients with moderate-to-severe AD received at least two courses of WMT between January 2022 and Decem

Causal relationships between oral-gut microbiome and bone neoplasm-related phenotypes: Insights from bidirectional Mendelian randomization.

The human oral and gut microbiota are the 4 largest microbial communities in the body and play crucial roles in maintaining homeostasis and influencing disease. Observational studies have suggested links between these microbiota and bone neoplasm-related phenotypes, but establishing causality has been challenging due to confounding factors and reverse causality. We conducted a bidirectional, 2-sample Mendelian randomization (MR) study to investigate evidence consistent with a potential causal association between the saliva and gut microbiota and various bone neoplasm-related phenotypes. Bile Acids Modulate Hepatic Glycolipid Metabolism via the Microbiota-Gut-Liver Axis in Lambs.

BACKGROUND: Bile acids are essential molecules that facilitate lipid emulsification and function as signaling molecules mediating host-microbiota interactions. They shape the gut microbial structure and function, playing a critical role in metabolic regulation via the gut-liver axis. OBJECTIVES: This study aimed to investigate the effects of exogenous bile acids, primarily hyocholic acid (HCA), on the microbiota-gut-liver metabolism in male Tan-lambs fed a high-grain diet. METHODS: Thirty six-mo-old male Tan lambs (Ovis aries) were randomly allocated into either a control group or an HCA-sup

Sex-Specific Differences in Gut Microbial Composition and Metabolism of Jiangshan Black Pigs.

The gut microbiota plays a vital role in regulating the host's physiological functions, including metabolism and immunity. The microbial composition and metabolism are modulated by multiple factors; host sex is an important yet under-explored determinant. To investigate the sex-dependent differences in the gut microbiota within the small and large intestine, sixteen somatic mature Jiangshan black pigs (eight males and eight females) were analyzed. The ileal and colonic microbial community and metabolites were profiled using 16S rRNA gene high-throughput sequencing and gas chromatography. Disti

Causal link between gut microbiota and obsessive-compulsive disorder: A two-sample Mendelian randomization analysis.

BACKGROUND: Previous studies have indicated a potential link between the gut microbiota and obsessive-compulsive disorder (OCD). However, the exact causal relationship remains uncertain. In this study, we employed a two-sample Mendelian randomization (MR) analysis to evaluate the causal connection between gut microbiota and OCD. METHODS: We collected Genome-Wide Association Study (GWAS) summary data on gut microbiota (n = 18, 340) and OCD (n = 199, 169), using single nucleotide polymorphisms (SNPs) as the instrumental variable. SNPs with an F-statistic of <10 were deemed weak instrumental var

Multi-omics analysis reveals the genetic basis for rapid CO2 utilization in the acetogenic bacterium Sporomusa sphaeroides KIAC.

Sporomusa sphaeroides KIAC is a novel acetogen isolated from cattle feces that exhibits rapid CO2 utilization. To investigate the molecular basis of this phenotype, we performed a comprehensive multi-omics analysis, including Genome-seq, RNA-seq, dRNA-seq, and Term-seq, to map its transcriptome architecture. We identified 2,158 transcription start sites and 2,275 transcript 3' ends, enabling high-resolution reconstruction of the transcriptional landscape and associated regulatory features. This analysis uncovered key cis-regulatory elements and an expanded regulatory role for the alternative s

Causal Associations Between 141 Gut Microbiota Taxa and 14 Rheumatic Diseases: A Mendelian Randomization Study.

OBJECTIVE: Gut microbiota has been increasingly recognized as important and novel targets for rheumatic diseases. However, previous studies mainly examined the associations, leaving causality largely unknown. This knowledge gap hinders the application of gut microbiota in preventing and treating rheumatic diseases. METHODS: We performed Mendelian randomization (MR) analysis to evaluate causal associations between gut microbiota and rheumatic diseases. Genetic instruments for 141 gut microbiota taxa were extracted from the published genome-wide association study (GWAS) (N = 18 340). Summary st

Gut microbiome dysbiosis in hepatocellular carcinoma patients with persistent HCV viremia versus viral clearance: a cross-sectional study.

BACKGROUND: Hepatocellular carcinoma (HCC) remains a lethal complication of chronic hepatitis C virus (HCV) infection, even after successful direct-acting antiviral (DAA) therapy. The gut microbiome influences hepatocarcinogenesis through the gut‒liver axis; however, the microbial signatures associated with HCC in DAA-treated patients are poorly defined. This study aimed to elucidate the patterns of microbiomes in HCV-treated patients who developed HCC, with a focus on bacterial diversity, differentially represented taxa, and their associations with clinical markers (FIB-4) and metabolic profi

Effects of dietary supplementation of fish oil plus vitamin D3 on gut microbiota and fecal metabolites, and their correlation with nonalcoholic fatty liver disease risk factors: a randomized controlled trial.

We previously reported that fish oil plus vitamin D3 (FO + D) could ameliorate nonalcoholic fatty liver disease (NAFLD). However, it is unclear whether the beneficial effects of FO + D on NAFLD are associated with gut microbiota and fecal metabolites. In this study, we investigated the effects of dietary supplementation of FO + D on gut microbiota and fecal metabolites and their correlation with NAFLD risk factors. Methods: A total of 61 subjects were randomly divided into three groups: FO + D group (2.34 g day-1 of eicosatetraenoic acid (EPA) + docosahexaenoic acid (DHA) + 1680 IU vitamin D3)
