Pre-clinical safety of topically administered sunitinib-loaded lipid and polymeric nanocarriers targeting corneal neovascularization.

Three different types of sunitinib-loaded (SUN-loaded) nanocarriers were compared, aiming at the topical treatment of corneal neovascularization (CNV): polymeric nanospheres (NS), liposomes (LIP), and solid lipid nanoparticles (SLN). Three out of eleven formulations prepared for an optimization study - the best SUN-loaded nanocarrier of each assessed type (NS, LIP, and SLN) - were selected, based on their size, polydispersity index (PdI), drug load (DL), and encapsulation efficiency (EE). These three optimal formulations were further characterized by nanoparticle tracking analysis (NTA), elect

Topical pH Sensing NIR Fluorophores for Intraoperative Imaging and Surgery of Disseminated Ovarian Cancer.

Fluorescence-guided surgery (FGS) aids surgeons with real-time visualization of small cancer foci and borders, which improves surgical and prognostic efficacy of cancer. Despite the steady advances in imaging devices, there is a scarcity of fluorophores available to achieve optimal FGS. Here, 1) a pH-sensitive near-infrared fluorophore that exhibits rapid signal changes in acidic tumor microenvironments (TME) caused by the attenuation of intramolecular quenching, 2) the inherent targeting for cancer based on chemical structure (structure inherent targeting, SIT), and 3) mitochondrial and lysos

Relationship Between Atopic Dermatitis and Food Allergy in Children.

Atopic dermatitis (AD) is a chronic inflammatory skin condition characterized by a compromised skin barrier due to a variety of reasons, such as hereditary predisposition, immunological overactivity, and skin microbiome disruption. There is strong evidence linking food allergies (FA) with AD in some children, and many children with AD develop asymptomatic food sensitivity. FA and AD are two frequent childhood illnesses that are closely related. Food allergies affect 30% of kids suffering from moderate and severe eczema and can cause a variety of symptoms, including dry, cracked skin, rash, itc

Hyaluronic acid-based supramolecular medicine with polyamines sequestration capability for cooperative anti-psoriasis.

Psoriasis seriously harms physical and mental health of patients. Hyaluronic acid (HA)-based topical formulation can increase drug concentration in psoriatic skin via CD44-assisted targeting. Herein, we developed a supramolecular medicine composed of curcumin-loaded HA-cucurbit[7]uril (HA-CB[7]@Cur), which could efficiently sequester polyamines (PAs) via host-guest interactions of CB[7] and PAs to suppress RNA-PAs immunocomplex formation. Meanwhile, anti-psoriasis drug Cur could be released from HA-CB[7]@Cur by PAs. With phenotypical disease evaluation, psoriasis area measurements and severity

[Use of fluorides for caries prevention].

For a few decades, a decrease in the caries burden in all age groups has been observed in Germany. The regular use of fluoride preparations is described as the main reason for the caries decline. It is well established that topical fluoride application directly to the tooth surfaces in the oral cavity, in particular fluoride-containing toothpastes and fluoride varnishes, is responsible for the decline. Several mechanisms of action, such as the formation of a calcium fluoride layer, remineralization, and the effects on bacterial plaque, were determined to be responsible for the overall clinical

Unusual rapid resolution of postsclerectomy exudative retinal detachment with topical NSAIDs therapy in a case of nanophthalmos.

Nanophthalmos is a developmental ocular malformation that has been associated with high risks of uveal effusion syndrome and exudative retinal detachment (ERD). A variety of surgical procedures and systemic/topical steroids have been described as effective for treatment of ERD. However, the possibility of side effects should be considered. Here, we describe a patient with nanophthalmos who was treated for recurrent ERD during long-term follow-up, and we discuss non-surgical treatment options that are available in such cases. A 43-year-old woman with bilateral nanophthalmos exhibited ERD in her

Cutaneous Application of Celecoxib for Inflammatory and Cancer Diseases.

BACKGROUND: Nonsteroidal anti-inflammatory drugs (NSAIDs) and particularly selective cyclooxygenase-2 (COX-2) inhibitors such as celecoxib (Cxb) are considered promising cancer chemopreventive for colon, breast, prostate, lung, and skin cancers. However, the clinical application to the prevention is limited by concerns about safety, potential to serious toxicity (mainly for healthy individuals), efficacy and optimal treatment regimen. Cxb exhibits advantages as potent antiinflammatory and gastrointestinal tolerance compared with conventional NSAID's. Recent researches suggest that dermatologic

Targeting effector memory T cells with the small molecule Kv1.3 blocker PAP-1 suppresses allergic contact dermatitis.

The voltage-gated potassium channel Kv1.3 has been recently identified as a molecular target that allows for selective pharmacological suppression of effector memory T (T(EM)) cells without affecting the function of naïve and central memory T cells. We here investigated whether PAP-1, a small molecule Kv1.3 blocker (EC50=2 nM), could suppress allergic contact dermatitis (ACD). In a rat model of ACD, we first confirmed that the infiltrating cells in the elicitation phase are indeed CD8+ CD45RC- memory T cells with high Kv1.3 expression. In accordance with its selective effect on T(EM) cells, PA

Pseudolipomatosis cutis: superficial dermal vacuoles resembling fatty infiltration of the skin.

Empty spaces within the dermis of paraffin-embedded sections of skin have been attributed to fatty infiltration and postulated to originate from topical steroid administration or sebaceous gland rupture. We examined skin biopsy specimens exhibiting dermal vacuolation to determine whether this phenomenon was associated with specific skin diseases and to attempt to illuminate its etiology. Routine hematoxylin-eosin-stained sections from 26 formalin-fixed, paraffin-embedded biopsy specimens were examined. Histochemical stains for mucin and immunohistochemical staining for S100 protein and vascula

Topical Hypochlorous Acid for Acne Vulgaris: Mechanisms, Clinical Evidence, and Therapeutic Potential.

Acne vulgaris is driven by Cutibacterium acnes, sebaceous activity, and innate immune activation, leading to inflammation, scarring, and reduced quality of life. Although standard topical and systemic therapies are effective, their use may be limited by irritation, adverse effects, and antimicrobial resistance. Hypochlorous acid (HOCl) exhibits antimicrobial, biofilm-disruptive, and anti-inflammatory activity through inhibition of nuclear factor kappa B (NF-κB) and mitogen-activated protein kinase (MAPK) signaling. Preliminary studies suggest HOCl provides lesion reductions comparable to benzo

Case Report: Abrocitinib treatment for refractory palmoplantar pustulosis.

Palmoplantar pustulosis (PPP) is a chronic relapsing inflammatory dermatosis characterized by recurrent sterile pustules on the palms and soles and often shows inadequate or unsustained responses to conventional systemic therapies and biologics. We report a 58-year-old woman with a 3-year history of refractory PPP. Before referral, oral prednisone and acitretin provided only transient disease control, with rapid relapse during dose tapering. Apremilast combined with topical halometasone cream and 308-nm excimer laser therapy failed to adequately control the plantar lesions. After referral, the

ON101 counteracts oxidative stress and inflammation in radiation-induced dermatitis through Keap1/Nrf2 signaling.

Radiation-induced dermatitis (RID) remains a significant and challenging side effect of radiotherapy, with few effective topical interventions. This study investigates ON101 as a therapeutic candidate for RID, focusing on its modulation of the Keap1/Nrf2 signaling pathway. Using a murine model of radiation-induced skin injury, topical application of ON101 substantially reduced skin damage, as evidenced by decreased wound severity scores, preserved body weight, and improved histopathological outcomes. Histological analyses using H&E and Masson's trichrome staining revealed that ON101 and its ac
