Heme versus non-heme iron-nitroxyl {FeN(H)O}⁸ complexes: electronic structure and biologically relevant reactivity.

Researchers have completed extensive studies on heme and non-heme iron-nitrosyl complexes, which are labeled {FeNO}(7) in the Enemark-Feltham notation, but they have had very limited success in producing corresponding, one-electron reduced, {FeNO}(8) complexes where a nitroxyl anion (NO(-)) is formally bound to an iron(II) center. These complexes, and their protonated iron(II)-NHO analogues, are proposed key intermediates in nitrite (NO2(-)) and nitric oxide (NO) reducing enzymes in bacteria and fungi. In addition, HNO is known to have a variety of physiological effects, most notably in the ca

Effect of ferrous ion on ε-poly-L-lysine biosynthesis by Streptomyces diastatochromogenes CGMCC3145.

ε-Poly-L-lysine (ε-PL) is known as an amino acid homopolymer occurring in nature. It is mainly produced by bacteria belonging to the family of Streptomycetaceae. In this study, the effect of ferrous ion on the metabolic pathway such as key enzyme activities and substrate consumptions relating to ε-PL production were investigated. When 0.1 mM Fe(2+) was supplemented into the culture medium of Streptomyces diastatochromogenes CGMCC3145 for 72 h of cultivation, the yield of ε-PL and biomass were enhanced by 29.2 and 41.9%, respectively, compared with those of the control. Synchronously, consumpti

Microscopic and spectroscopic characterization of Hg(II) immobilization by mackinawite (FeS).

This study investigated the solid-phase Hg formed by reacting 0.005 or 0.01 M Hg(II) with 10 g/L mackinawite (FeS) as a function of pH in 0.2 M chloride solutions using X-ray diffraction (XRD), transmission electron microscopy (TEM), and extended X-ray absorption fine structure (EXAFS) analyses. Under all experimental conditions, XRD analysis showed formation of metacinnabar (β-HgS) as a bulk-phase sorption product, in agreement with the results from high angle annular dark field-scanning transmission electron microscopy (HAADF-STEM) and selected area electron diffraction (SAED) in TEM analysi

[Cutaneous calcinosis induced by topical calciphylaxis in rats. I. Ultrastructural aspcts].

Skin calcification induced by topical calciphylaxis was provoked by a subcutaneous injection of iron chloride in rats previously sensitized by D.H.T. and studied by electron microscope. The evolution of the calciphylatic response can be resumed as follows: -- early appearance of numberous granules in the mitochondria of the cells of the connective tissue; their inorganic nature was revealed by microincineration and it is highly probable that they are a reserve of mineral ions; -- between the 6 hr, and the 18 hr, stages the interfibrillar matrix contained rounded vesicular structures, enclosed

Non-invasive assessment of normal and impaired iron homeostasis in the brain.

Strict iron regulation is essential for normal brain function. The iron homeostasis, determined by the milieu of available iron compounds, is impaired in aging, neurodegenerative diseases and cancer. However, non-invasive assessment of different molecular iron environments implicating brain tissue's iron homeostasis remains a challenge. We present a magnetic resonance imaging (MRI) technology sensitive to the iron homeostasis of the living brain (the r1-r2* relaxivity). In vitro, our MRI approach reveals the distinct paramagnetic properties of ferritin, transferrin and ferrous iron ions. In th

Ferroptosis-Driven Anticancer Activity of 1,3,4-Oxadiazole-Benzimidazole/Acetamide Hybrids: Design, Synthesis, and Biological Profiling.

A series of 1,3,4-oxadiazole-benzimidazole/acetamide derivatives (7a-r) was designed, synthesized, and evaluated for their anticancer activity. NCI-screening results at 10 µM against the 60 human tumor cell line panel revealed a broad-spectrum of antiproliferative effects of the tested compounds. Compounds 7b, 7e, 7f, 7g, 7h, 7k, 7l, and 7r were further tested in a five-dose assay, and they exhibited half-maximal growth inhibitory (GI50) values ranging from 0.90 to 43.50 µM. Focused studies in MDA-MB-231 triple-negative breast cancer (TNBC) cells demonstrated that compounds 7b, 7g, 7h, 7k, and

Structural and Spectroscopic Characterization of a High-Spin {FeNO}(6) Complex with an Iron(IV)-NO(-) Electronic Structure.

Although the interaction of low-spin ferric complexes with nitric oxide has been well studied, examples of stable high-spin ferric nitrosyls (such as those that could be expected to form at typical non-heme iron sites in biology) are extremely rare. Using the TMG3 tren co-ligand, we have prepared a high-spin ferric NO adduct ({FeNO}(6) complex) via electrochemical or chemical oxidation of the corresponding high-spin ferrous NO {FeNO}(7) complex. The {FeNO}(6) compound is characterized by UV/Visible and IR spectroelectrochemistry, Mössbauer and NMR spectroscopy, X-ray crystallography, and DFT c

Mitigating dithiothreitol interference to gold/thiol interface in electrochemical detection of cathepsin B activity toward multiplex protease analysis.

Proteases are overexpressed at various stages of conditions such as cancers and thus can serve as biomarkers for disease diagnosis. Electrochemical techniques to detect the activity of extracellular proteases have gained attraction due to their multiplexing capability. Here we employ an electrochemical approach based on a 3 × 3 gold (Au) microelectrode array (MEA) functionalized with (2-aminoethyl)ferrocene (AEF) tagged specific peptide substrates to monitor cathepsin B (CB) protease activity. Cleavage of these peptide substrates by proteases leads to an exponential decay in the alternating cu

Proton-linked effects on the axial coordination in ferric and ferrous horse heart cytochrome c.

The effect of pH (between pH 7.0 and 0.0) on the spectroscopic properties of ferric and ferrous derivatives of horse heart cytochrome c (hhcytc-Fe(III) and hhcytc-Fe(II), respectively) in 1.0M phosphate buffer is reported. This study further deepens the investigation of spectroscopic properties of a heme-protein to very low pH (i.e., pH ≈ 0) showing relevant pH-dependent variations of the heme surroundings, involving the heme-Fe atom coordination. For both hhcytc-Fe(III) and hhcytc-Fe(II) a severe weakening of the Fe-Met80 has been detected with a similar pKa, ranging between 2.5 and 3.0, sugg

Natural pyrite-stimulative periodate activation: efficiency and mechanism study.

Natural pyrite (NP) is an alternative catalyst for wastewater purification via advanced oxidation processes (AOPs). However, the activation performance and mechanism of periodate (PI) by NP have not yet been revealed. Herein, this work examines the activation performance of NP towards PI and its application in the degradation of antibiotic wastewater. Interestingly, 95.69% of chlortetracycline (CTC) was degraded by NP within 20 min via PI activation. Besides, NP shows effective degradation of various pollutants such as rhodamine B (65.81%), sulfamethoxazole (89.04%), and sodium butylxanthate (

A redox switch allows binding of Fe(II) and Fe(III) ions in the cyanobacterial iron-binding protein FutA from Prochlorococcus.

The marine cyanobacterium Prochlorococcus is a main contributor to global photosynthesis, whilst being limited by iron availability. Cyanobacterial genomes generally encode two different types of FutA iron-binding proteins: periplasmic FutA2 ABC transporter subunits bind Fe(III), while cytosolic FutA1 binds Fe(II). Owing to their small size and their economized genome Prochlorococcus ecotypes typically possess a single futA gene. How the encoded FutA protein might bind different Fe oxidation states was previously unknown. Here, we use structural biology techniques at room temperature to probe

Assay of Fe(III) Chelate Reductase Activity in Arabidopsis thaliana Root.

A sensitive FerroZine assay is used to measure the membrane-bound ferric-chelate reductase activity in the Arabidopsis thaliana roots. In Arabidopsis, FRO2 (FERRIC CHELATE REDUCTASE 2) encodes the Fe(III) chelate reductase and its expression is induced by iron deficiency. As FRO2 reduces Fe(III) to soluble Fe(II), the resulting Fe(II) forms a purple-colored complex with the dye FerroZine. The concentration of the Fe(II)-FerroZine is directly proportional to the absorbance at 562 nm.

Deep-dive into iron-based co-precipitation of arsenic: A review of mechanisms derived from synchrotron techniques and implications for groundwater treatment.

The co-precipitation of Fe(III) (oxyhydr)oxides with arsenic (As) is one of the most widespread approaches to treat As-contaminated groundwater in both low- and high-income settings. Fe-based co-precipitation of As occurs in a variety of conventional and decentralized treatment schemes, including aeration and sand filtration, ferric chloride addition and technologies based on controlled corrosion of Fe(0) (i.e., electrocoagulation). Despite its ease of deployment, Fe-based co-precipitation of As entails a complex series of chemical reactions that often occur simultaneously, including electron-

α-Fe2O3 based nanotherapeutics for near-infrared/dihydroartemisinin dual-augmented chemodynamic antibacterial therapy.

Due to the negligible bacterial resistance, chemodynamic therapy (CDT) is a promising treatment for bacterial infection. However, it is severely impeded by the constant body temperature, shortage of Fe(Ⅱ) ions and insufficient H2O2 level in infected tissue. To enhance the therapeutic efficiency of CDT, improved strategies are urgently needed to tackle these problems. Herein, we exploited an infection microenvironment-responsive nanotherapeutics for near-infrared (NIR)/dihydroartemisinin (DHA) dual-augmented antibacterial CDT. The convenient encapsulation of DHA-loaded α-Fe2O3 nanorods with met

WS2 significantly enhances the degradation of sulfachloropyridazine by Fe(III)/persulfate.

The use of antibiotics has become an indispensable part of the production and life of human society. Among them, sulfonamide antibiotics widely used in humans and animals are considered to be one of the most crucial antibiotics. However, antibiotics are difficult to degrade naturally, leading to an accumulation in the environment and a potential hazard to human health. In this paper, WS2 as a co-catalyst could reduce trace Fe(III) to Fe(II) which exhibited a great activating ability to PS through the exposed W(IV) active sites, and formed the Fe(III)/Fe(II) cycle to degrade sulfachloropyridazi
