The effect of vitamin A deficiency and fluoride on glycosaminoglycan metabolism in bone.

The effects of fluoride intake and vitamin A deficiency on glycosaminoglycan metabolism in vivo were investigated. Weanling female rats were fed either a vitamin A deficient diet ad libitum, a vitamin A supplemented diet pair-fed to the deficient animals, or the vitamin A supplemented diet ad libitum. Additionally, each vitamin A dietary group was divided into three subgroups with the animals receiving water containing 0, 10 or 50 ppm fluoride. The results showed that the groups receiving 10 and 50 ppm fluoride at all dietary levels of vitamin A had significantly higher in vivo 35SO4 incorpora

Interactions of fluoride and guanine nucleotides with thyroid adenylate cyclase.

The activation of bovine thyroid adenylate cyclase (ATP pyrophosphate-lyase (cyclizing), EC 4.6.1.1) by Gpp(NH)p has been studied using steady-state kinetic methods. This activation is complex and may be characterized by two Gpp(NH)p binding sites of different affinities with measured constants: Ka1 = 0.1 micro M and Ka2 = 2.9 micro M. GDP beta S does not completely inhibit the Gpp(NH)p activation: analysis of the data is consistent with a single GDP beta S inhibitory site which is competitive with the weaker Gpp(NH)p site. Guanine nucleotide effects upon F- activation of adenylate cyclase hav

The pathophysiological profile of the acute cardiovascular toxicity of sodium fluoride.

The intravenous infusion of sodium fluoride (2 mg/kg X min) into anesthetized rats caused a progressive fall in arterial blood pressure, cardiac output, heart rate and peripheral resistance. Respiratory rate increased during the first 20 min of infusion resulting in increased oxygen and decreased carbon dioxide blood concentrations. Total body oxygen consumption decreased after 30 min of NaF infusion by 29%, whereas the respiratory quotient (RQ) increased from 0.8 to 1.06. Death occurred after a mean dose of 79.6 +/- 4.6 mg/kg NaF. The terminal cardiac event before death was atrioventricular b

Fluoride stimulation of the rat parathyroid gland: an ultrastructural study.

The parathyroid glands of rats given 150 ppm fluoride in the drinking water for 10 weeks are evaluated ultrastructurally and compared to the parathyroid glands of untreated rats. As a result of fluoride ingestion, the majority of the parathyroid cells are dark chief cells, indicating that these cells are in the active stages of the secretory cycle. More significantly, in the fluoride-treated rats, the cytoplasmic organelles of the dark chief cells are even more developed that those seen in the dark chief cells of untreated rats. The dark cells contain an electron-dense cytoplasm with abundant

Effect of sodium fluoride on concentrating and diluting ability in the rat.

Mechanisms for the concentrating defect produced by fluoride were examined in the rat. Free-water clearance at all levels of delivery was normal after 5 days of chronic fluoride administration in the hereditary hypothalamic diabetes insipidus rat. In the Sprague-Dawley rats, during moderate fluoride administration (120 micronmol/kg per day), urine osmolality and cyclic AMP excretion decreased and urine volume increased, but after exogenous vasopressin, volume decreased and osmolality and cyclic AMP increased appropriately. During larger daily doses of fluoride (240 micronmol/kg per day) urinar

Effect of matrix bound phosphate and fluoride on mineralization of dentin.

Completely demineralized bovine dentin mineralized to a high degree, but only after extraction of about half of its organic phosphate. Initiation of mineral formation depended on the concentrations of Ca2 plus, Pi, HCO3 minus and H plus ions in the incubation solution. After nucleation, mineral accretion by the dentin continued in solutions that did not, themselves, initiate mineralization. The nucleation phase, but not the subsequent accretion of mineral, was sensitive to small changes in temperature. Inclusion of 0.05 mM NaF in the solution markedly reduced the amount of mineral deposited du

On topical application of fluorides and its inhibiting effect on caries.

The observations of interest made in this investigation may be summarized as follows: 1. Conventional fluoride application by mouthrinsing with neutral NaF solution (0.025% daily for three weeks or three months; or 0.05% solution daily; or 0.2% solution weekly for three weeks) is found to result in only moderate and insignificant uptake of F in facial enamel surfaces. The gain is in the order of a hundred ppm within 10 to 15 microns' depth (as determined by an acid etching technique) and a few hundred ppm at 0.02 to 0.05 microns (according to pilot ion probe analysis). 2. Painting, at one week

Evaluation of SMART technique using Nano-silver fluoride and Silver Diamine Fluoride; evaluation of tooth discoloration and shear bond strength in primary molars.

The aim of this study was to evaluate the shear bond strength and tooth discoloration of resin-modified glass ionomer cement (RMGIC) and high-viscosity glass ionomer cement (HGIC) restorations in primary teeth treated with either nano-silver fluoride (NSF) or silver diamine fluoride (SDF). Sixty-six primary molars with dentin caries were randomly assigned to six groups of 11 teeth each: two NSF-treated groups (one RMGIC, one HGIC) and two SDF-treated groups (one RMGIC, one HGIC). Two additional control groups received direct RMGIC and HGIC restorations, respectively. All samples were tested fo

Impact of explicit fluoride-related language in prefectural dental health ordinance on fluoride mouth-rinse programs dissemination in Japan: a quasi-experimental study.

OBJECTIVES: To estimate the impact of explicit fluoride-related language in prefectural dental health ordinance on the dissemination of school-based fluoride mouth-rinse (S-FMR) programs. METHODS: A longitudinal ecological design was employed to analyze prefectural-level panel data from Japan spanning 2007 to 2018. Ordinances enacted between 2010 and 2014 were categorized by the presence of explicit fluoride-related terms: 1) explicit mention of "fluoride mouth-rinse" (FMR group); 2) explicit mention of "fluoride application" (FA group); and 3) no fluoride-related policy language (NF group).

Arrest of interproximal caries lesion in primary molars with 38% silver diamine fluoride solution and 5% sodium fluoride varnish: A randomized clinical trial.

BACKGROUND: Dental caries is a global public health concern, with interproximal caries presenting a particular challenge due to their limited accessibility. Fluoride has long been employed in the prevention and management of early-stage carious lesions. AIM: To evaluate and compare the effectiveness of 38% silver diamine fluoride (SDF) and 5% sodium fluoride varnish (NaF) in arresting interproximal caries in primary molars. SETTINGS AND DESIGN: A split-mouth randomized clinical trial was conducted following the CONSORT guidelines. METHODS: Children aged 5-9 years with at least two noncavita
