Different Efficacy of Five Soluble Dietary Fibers on Alleviating Loperamide-Induced Constipation in Mice: Influences of Different Structural Features.

Different dietary fibers have distinct structures, leading to significant variations in their laxative effects. To explore how these structural differences impact constipation intervention, a 14-day study was conducted on loperamide-induced constipated mice using five dietary fibers: soluble dietary fiber from steamed sweet potato (SDF-S), oat β-glucan (OB), polydextrose (PD), arabinogalactan (AG), and inulin (IN). The results showed that four fibers, excluding PD, significantly improved gastrointestinal (GI) transit rate (p < 0.05), although PD had the highest fecal moisture, it was significa

Impact of fermented foods consumption on gastrointestinal wellbeing in healthy adults: a systematic review and meta-analysis.

OBJECTIVE: In recent years, the consumption of fermented foods (FFs) has been linked with gastrointestinal health and wellbeing. Here, we systematically review and meta-analyse the currently available evidence relating to this as part of the COST Action PIMENTO and guided by the European Food Safety Authority (EFSA) health claim dossiers. METHODS: MEDLINE, Scopus and Cochrane CENTRAL bibliographic libraries were searched for relevant literature up to 31st January 2025. All eligible studies were included for narrative review as per EFSA guidelines, but only randomised controlled trials (RCTs)

Anti-muscarinic 3 antibodies associate with a severe clinical phenotype in patients with systemic sclerosis.

OBJECTIVES: Functional antibodies play a role in SSc gastrointestinal (GI) disease, but their clinical relevance is unclear. We examined GI and extraintestinal features associated with anti-muscarinic-3 receptor (M3R) antibodies in SSc patients. METHODS: In a cohort enriched for GI symptoms, SSc patients were tested for anti-M3R antibodies using an ELISA. High-titres were defined a priori based on the top quartile of anti-M3R titres to enhance specificity. Clinical features, including high GI severity (Medsger GI Severity Score of ≥2), whole-gut transit study data and UCLA Scleroderma Clinica

The cannabinoid-1 receptor inverse agonist taranabant reduces abdominal pain and increases intestinal transit in mice.

BACKGROUND: Constipation-predominant irritable bowel syndrome (IBS-C) is a common functional gastrointestinal (GI) disorder with abdominal pain and decreased motility. Current treatments of IBS-C are insufficient. The aim of this study was to evaluate the potential application of taranabant, a cannabinoid type 1 (CB1) inverse agonist using mouse models mimicking the symptoms of IBS-C. METHODS: Changes in intestinal contractile activity were studied in vitro, using isolated mouse ileum and colon and intracellular recordings. In vivo, whole gastrointestinal transit (WGT) and fecal pellet output…

The effect of physical exercise on parameters of gastrointestinal function.

Exercise decreases splanchnic bloodflow. Therefore exercise may induce alterations in gastrointestinal (GI) function. In the present study w … urements during exercise. Cycling at 70% Wmax does not lead to differences in reflux, gastric pH or gastrointestinal transit in healthy trained individuals. The distal oesophageal pressure decreases and peristaltic velocity increases. The lactulose/rhamnose ratio and jejunal glucose absorption are decreased during exercise.

Synergistic interaction between fentanyl and a tramadol: paracetamol combination on the inhibition of nociception in mice.

Multimodal analgesic approaches to manage acute and chronic pain are commonly used in humans. Here, we attempted to characterize a synergistic interaction between fentanyl, tramadol, and paracetamol on the inhibition of nociception in a model of visceral pain in mice. The three-drug combined treatment displayed a potent synergistic antinociceptive effect, together with a significant reduction of gastrointestinal transit inhibition. Furthermore, selective µ- and κ-opioid receptor antagonists reversed these synergistic antinociceptive effects, thus suggesting a pivotal role of the opioid system.…

Lactiplantibacillus plantarum NCHBL-004 improves intestinal motility and modulates serotonergic gene expression in a loperamide-induced constipation model.

Slow-transit constipation (STC) is characterized by reduced colonic motility, impaired smooth muscle function, and altered mucosal signaling. Among these pathways, mucosal serotonin (5-HT), mainly produced by enterochromaffin cells, contributes to intestinal motility regulation. In a loperamide-induced constipation model in male C57BL/6 J mice, we evaluated the effects of Lactiplantibacillus plantarum NCHBL-004 on bowel function and associated molecular changes. Compared with loperamide-treated mice, live NCHBL-004 at 1 × 10⁹ CFU improved fecal output and gastrointestinal transit and partially…

Lycopene regulates intestinal dysmotility and behavioral disorders by regulating enteric neurons and the cholinergic system.

Gut dysmotility is a prevalent gastrointestinal disorder characterized by disrupted defecation and often accompanied by depression and anxiety. Lycopene (LYC) is a type of carotenoid with strong antioxidant and anti-inflammatory properties. However, the effects of LYC on gut dysmotility and related behavioral disorders remain elusive. Herein, we found that 100 mg kg-1 d-1 LYC notably improved the gut transit time and intestinal transit rate as well as concurrently alleviated depression- and anxiety-like behaviors in a diphenoxylate (Dip, 5 mg kg-1 d-1)-induced constipated mouse model. LYC pret

Barber Pole Sign in CT Angiography, Adult Presentation of Midgut Malrotation: A Case Report.

Adult midgut volvulus is a challenging diagnosis because of its low incidence and nonspecific symptoms. Diagnostic delay and long-term complaints are frequent in this clinical scenario. We reported a patient referred to our diagnostic imaging unit with intermittent abdominal pain, bloating and episodic vomiting for several years. He underwent barium gastrointestinal transit and abdominal ultrasound, which revealed severe gastric dilatation, food retention and slow transit until a depressed duodenojejunal flexure, with malrotation of the midgut and jejunal loops being located in the right upper…

Evidence for the involvement of 5-HT2A receptors in mild mesenteric ischemia/reperfusion dysfunctions in mice.

In this study, the involvement of 5-HT2A receptors on mesenteric ischemia-reperfusion injury was examined in mice. Intestinal ischemia produced by 45 min occlusion of superior mesenteric artery was followed by 24h reperfusion (I/R). The 5-HT2A selective antagonist, ketanserin (0.5 mgkg(-1)) or the 5-HT2A agonist DOI (0.25 mgkg(-1)) was intravenously administered before ischemia and 8h after the beginning of reperfusion. The effects were compared with those obtained in sham operated animals (S). Ketanserin prevented the upper gastrointestinal transit delay induced by I/R (P<0.01), protected int…

Cisapride. Drug interactions of clinical significance.

Cisapride is a prokinetic agent which restores motility of the gastrointestinal tract in conditions of decreased bowel transit. It may also alter the absorption of coadministered drugs. The absorption of morphine, diazepam, cyclosporin, alcohol (ethanol) and levodopa are increased. Initial absorption of cimetidine and raniditine is also increased, but overall absorption is lower due to increased bowel transit. The absorption of digoxin, propranolol and the anticoagulants warfarin and phenprocoumon appears unaffected by cisapride, although increase thrombotest values were seen with acenocoumaro

The ageing gastrointestinal tract.

PURPOSE OF REVIEW: This article reviews the impact of ageing on the gastrointestinal tract, including effects on the absorption of nutrients and drugs and the gastrointestinal tract defence system against ingested pathogens. RECENT FINDINGS: Recent publications support earlier observations of an age-related selective decline in gut function including changes in taste, oesophageal sphincter motility, gastric emptying, and neurons of the myenteric plexus related to gut transit which may impact the nutritional status. Ageing is also associated with structural and functional mucosal defence defec

Effects on gastrointestinal functions and symptoms of serotonergic psychoactive agents used in functional gastrointestinal diseases.

The effects of antidepressants on the gastrointestinal tract may contribute to their potential efficacy in functional dyspepsia and irritable bowel syndrome; buspirone, a prototype 5-HT1A agonist, enhances gastric accommodation and reduces postprandial symptoms in response to a challenge meal. Paroxetine, a selective serotonin reuptake inhibitor, accelerates small bowel but not colonic transit, and this property may not be relevant to improve gut function in functional gastrointestinal disorders. Venlafaxine, a prototype serotonin norepinephrine reuptake inhibitor, enhances gastric accommodati

Gastrointestinal involvement in insulin-requiring diabetes mellitus.

Gastrointestinal (GI) dysfunction in diabetes mellitus has never been evaluated systematically in all parts of the digestive system in a group of diabetics. Therefore, we have evaluated the frequency, extent, and clinical significance of GI complications in 75 consecutive, male, insulin-requiring diabetics (46 with neuropathy). Nineteen percent of the 75 patients and 30% of those with neuropathy had one or more GI symptoms. Esophageal, gastric, gallbladder, and small intestinal functions were studied in 30 patients using radionuclide esophageal and gastric emptying, postprandial gallbladder em

Electrophysiological Mechanisms of Gastrointestinal Arrhythmogenesis: Lessons from the Heart.

Disruptions in the orderly activation and recovery of electrical excitation traveling through the heart and the gastrointestinal (GI) tract can lead to arrhythmogenesis. For example, cardiac arrhythmias predispose to thromboembolic events resulting in cerebrovascular accidents and myocardial infarction, and to sudden cardiac death. By contrast, arrhythmias in the GI tract are usually not life-threatening and much less well characterized. However, they have been implicated in the pathogenesis of a number of GI motility disorders, including gastroparesis, dyspepsia, irritable bowel syndrome, mes
