Genetically predicted circulating polyunsaturated fatty acids and Crohn's disease susceptibility: A two-sample mendelian randomization study.

Crohn disease (CD) remains an increasing global health burden. Omega-3 and omega-6 polyunsaturated fatty acids participate in lipid metabolism and inflammatory regulation, but previous observational and interventional studies have produced inconsistent findings. This study aimed to assess whether genetically predicted circulating omega-3, omega-6, and the omega-6/omega-3 ratio are associated with CD susceptibility using a two-sample Mendelian randomization (MR) design. Genome-wide association study summary data for circulating omega-3, omega-6, and the omega-6/omega-3 ratio were obtained from

Mendelian Randomization to Examine the Causal Effects of Cystatin on Ovarian Lesions.

BACKGROUND: Cystatin, a superfamily of cysteine protease inhibitors, are implicated in extracellular matrix remodeling, immune modulation, and tumor progression. Observational studies have reported associations between specific cystatin and gynecological pathologies, including ovarian cancer. However, whether these associations reflect causality remains uncertain due to potential confounding and reverse causation. METHODS: We performed a two-sample Mendelian randomization (MR) study to investigate the causal relationships between genetically predicted levels of seven cystatin subtypes (Cystat

Analysis of multi-trait evolution across independently evolved cavefish populations reveals shared and independent evolution of suites of traits.

Environmental perturbations often lead to the evolution of multiple traits. Determining whether shared genetic factors underlie multi-trait evolution is a central question in evolutionary biology. In the Mexican tetra, Astyanax mexicanus, cave-dwelling populations have repeatedly evolved multiple traits. The repeated evolution of these traits, paired with robust environmental differences between the surface and cave habitats, provide an opportunity to investigate the genetic basis of multi-trait evolution. Here, we investigate the extent to which shared genetic mechanisms underlie the repeated

Causal Relationship Between 38 Dietary Factors, Including Sugar and Alcohol Intakes, and Polycystic Ovary Syndrome: A Two-Sample Mendelian Randomization Study.

PURPOSE: Emerging evidence suggests that diets high in sugar consumption may be implicated in the development of polycystic ovary syndrome (PCOS), but the causal nature of these associations remains unclear. This study aimed to explore the potential causal links between 38 specific dietary factors (including alcohol and added sugar), particularly sugar-sweetened beverage intake, and the risk of PCOS. PATIENTS AND METHODS: A two-sample Mendelian randomization (MR) approach was employed using genome-wide association study summary statistics. The inverse-variance weighted (IVW) method served as

Causal effects of endometriosis on cancer risk: A Mendelian randomization study.

Endometriosis has been reported in epidemiological studies to be associated with certain types of cancer. However, the presence of reverse causality and residual confounding due to common risk factors introduces uncertainty regarding the extent to which endometriosis itself contributes to the development of cancer. We performed the Mendelian randomization (MR) to investigate the causal associations between endometriosis and 34 different types of cancers. The results of the inverse-variance-weighted (IVW) model suggested that genetic predisposition to endometriosis was causally associated with

Pleiotropic Effects of Polymorphisms in the BCL11A Gene on Laboratory Parameters in Sickle Cell Anemia.

Sickle cell anemia (SCA) is characterized by hematological events that lead to vaso-occlusion and the onset of clinical manifestations. Fetal hemoglobin (HbF) has been shown to positively influence the clinical outcomes of individuals with SCA. Genetic polymorphisms are known to modulate clinical phenotypes by increasing HbF levels, with the BCL11A gene being an important marker in this regard for future therapies. However, while the BCL11A gene plays a role in the regulation of several genes during hematopoiesis, its various effects are not yet fully understood. The study aimed to investigate

Causal Effects Between Antibody-Mediated Immune Responses and Parkinson's Disease: Insights from Genetic Studies.

BACKGROUND: Parkinson's disease (PD) is a multifactorial neurodegenerative disorder with a complex etiology. Increasing evidence suggests that antibody-mediated immune responses elicited by prior infectious exposures may contribute to its pathogenesis; however, whether these associations reflect true causal relationships remains unresolved, largely due to confounding and other inherent limitations of observational study designs. METHODOLOGY: We conducted a two-sample Mendelian randomization (MR) analysis to interrogate the causal associations between 46 antibody-mediated immune responses and

Genetic Evidence for the Associations of Alcohol Intake Frequency and Dietary Salt Intake with Non-Alcoholic Fatty Liver Disease Risk and Hepatic Steatosis: A Mendelian Randomization Study.

OBJECTIVE: Non-Alcoholic Fatty Liver Disease (NAFLD) poses a significant global health burden, yet causal relationships with modifiable dietary factors remain incompletely defined. This Mendelian Randomization (MR) study evaluated the causal associations between dietary exposures and NAFLD risk, as well as related hepatic traits. METHODS: We performed univariable, Bayesian Weighted MR (BWMR), and Multivariable MR (MVMR) analyses using genetic variants as instrumental variables. Data from 335,394 to 462,630 European-ancestry individuals (IEU OpenGWAS) were analyzed for 20 dietary exposures. Ou

Erythema nodosum, malignant melanoma and non-melanoma skin cancer in relation to inflammatory bowel disease: a Mendelian randomization study.

Inflammatory bowel disease (IBD) is a multisystem condition that could affect the cutaneous systems, namely cutaneous extraintestinal manifestations (EIMs). It has been suggested that IBD is associated with erythema nodosum (EN), malignant melanoma (MM) and non-melanoma skin cancer (NMSC). However, the potential causal relationship between IBD and the mentioned above cutaneous EIMs is still unclear. This study aims to determine the effect of IBD on EN, MM and NMSC within a Mendelian randomization (MR) design. Summary-level data for IBD, EN, MM, NMSC were obtained from large-scale genome-wide a

Gene discovery and biological insights into anxiety disorders from a large-scale multi-ancestry genome-wide association study.

We leveraged information from more than 1.2 million participants, including 97,383 cases, to investigate the genetics of anxiety disorders across five continental groups. Through ancestry-specific and cross-ancestry genome-wide association studies, we identified 51 anxiety-associated loci, 39 of which were novel. In addition, polygenic risk scores derived from individuals of European descent were associated with anxiety in African, admixed American and East Asian groups. The heritability of anxiety was enriched for genes expressed in the limbic system, cerebral cortex, cerebellum, metencephalo

The Genetic Architecture of Domestication in Animals.

Domestication has been essential to the progress of human civilization, and the process itself has fascinated biologists for hundreds of years. Domestication has led to a series of remarkable changes in a variety of plants and animals, in what is termed the "domestication phenotype." In domesticated animals, this general phenotype typically consists of similar changes in tameness, behavior, size/morphology, color, brain composition, and adrenal gland size. This domestication phenotype is seen in a range of different animals. However, the genetic basis of these associated changes is still puzzl

Shared Genetic Architecture Between Atopic Dermatitis and Autoimmune Diseases.

Atopic dermatitis (AD) and autoimmune diseases exhibit epidemiological comorbidity, yet the shared genetic architecture remains incompletely understood. We investigated the genetic overlap between AD and three autoimmune disorders including inflammatory bowel disease (IBD), rheumatoid arthritis (RA), and vitiligo, leveraging genome-wide association data. Despite modest evidence for global genetic correlations, we found 113 independent pleiotropic loci shared among AD and autoimmune diseases, with 11 displaying a concordant effect across all 3 pairwise comparisons. Genetic Interactions Affect Lung Function in Patients with Systemic Sclerosis.

Scleroderma, or systemic sclerosis (SSc), is an autoimmune disease characterized by progressive fibrosis of the skin and internal organs. The most common cause of death in people with SSc is lung disease, but the pathogenesis of lung disease in SSc is insufficiently understood to devise specific treatment strategies. Developing targeted treatments requires not only the identification of molecular processes involved in SSc-associated lung disease, but also understanding of how these processes interact to drive pathology. One potentially powerful approach is to identify alleles that interact gen

Identification of variants in pleiotropic genes causing "isolated" premature ovarian insufficiency: implications for medical practice.

Next-generation sequencing (NGS) is increasingly being used in a clinical setting for the molecular diagnosis of patients with heterogeneous disorders, such as premature ovarian insufficiency (POI). We performed NGS of ~1000 candidate genes in four unrelated patients with POI. We discovered the genetic cause of "isolated" POI in two cases, both of which had causative variants in surprising genes. In the first case, a homozygous nonsense variant in NBN was causative. Recessive function-altering NBN variants typically cause Nijmegen breakage syndrome characterized by microcephaly, cancer predisp
