Low genetic risk for coronary artery disease underlies multigenerational longevity and healthy aging.

Aging is a major risk factor for chronic diseases. Unlike the general population, members of long-lived families maintain exceptional health as they age, with over 10 years delayed onset of their first chronic disease. We therefore hypothesize that one of the key features explaining healthy survival up to high ages (longevity) is the absence of chronic disease risk alleles. We investigated this hypothesis in the Leiden Longevity Study, a cohort with data from more than 420 long-lived families in three generations and the Leiden 85-plus study. To analyze our data, we constructed a set of polyge

Association of neurofilament light chain, CXCL9, IL-6, and TNFRSF11B with intrinsic capacity in community-dwelling older adults.

Intrinsic capacity (IC) reflects multidomain functional reserve and a measurable phenotype of healthy ageing, but its biological correlates remain incompletely understood. We examined associations of neurofilament light chain (NfL), CXCL9 and inflammatory biomarkers with IC in community-dwelling older adults. We conducted a cross-sectional analysis within the Screening and Prevention of Intrinsic Capacity Decline in Elders (SPICE) programme. IC was derived from locomotion (SPPB), cognition (MoCA), psychological health (PHQ-9), and vitality (MNA-SF), rescaled to a 0-100 composite and analysed a

Adherence to the Eatwell Guide and associations with markers of physical function: a prospective analysis within the UK Biobank cohort.

The Eatwell Guide represents the UK's principal healthy eating model and understanding whether adherence to UK dietary recommendations can attenuate age-related functional decline is essential to inform healthy ageing strategies. In up to 157,457 participants from the UK Biobank, we explored cross-sectional and prospective associations between adherence to the Eatwell Guide and markers of physical function (grip strength, fat-free mass percentage, self-reported walking pace, and falls). Eatwell Guide adherence scores were derived from 24-h dietary recall data (Oxford WebQ), and quantified usin

Prioritizing clinical utility to use allostatic load as a biomarker of the consequences of stress.

Allostatic load measures the physiological consequences of stress and is associated with disease outcomes. Clinical use of allostatic load is limited, in part, because its calculation requires biomarkers not routinely measured in clinical care. Presently, we evaluated associations between high allostatic load and depression as co-occurring consequences of stress among a cohort representing population subgroups largely excluded from existing literature. We emphasize clinical implementation potential by employing both an established and a recent consensus streamlined allostatic load score. Data

Circulating factors induced by time-restricted eating drive metabolic reprogramming in endothelial cells.

Age-related endothelial dysfunction in the cerebral microcirculation contributes significantly to the pathogenesis of vascular cognitive impairment and dementia (VCID). Time-restricted eating (TRE) has emerged as a promising lifestyle intervention with beneficial effects on metabolic and vascular health; however, the mechanisms by which TRE influences the brain microvasculature remain incompletely understood. In particular, the role of circulating factors induced by TRE in modulating endothelial function has not been systematically investigated. Here, we tested the hypothesis that circulating

Higher burden of neuropsychiatric symptom-like behaviors associated with canine cognitive dysfunction compared to normal aging in the Dog Aging Project.

Non-cognitive, neuropsychiatric symptoms (NPS) are nearly universal in Alzheimer's disease (AD), but investigation of their underlying biology is complicated by comparative medicine approaches that incompletely capture spontaneous disease, primarily using transgenic rodent models. The aged companion dog, which spontaneously develops an AD-like disease called canine cognitive dysfunction (CCD), may help fill this translational gap. Using data from the Dog Aging Project with > 10,000 aged dogs (> 8 years old), we identify numerous behaviors in dogs "at-risk" for and with CCD that mirror NPS in h

Task difficulty and limb dominance modulate the effects of ageing on neuromuscular function.

Neuromuscular function is critical for independence in ageing, yet asymmetries between dominant and non-dominant limbs, arising from central or peripheral mechanisms, are not well understood. This study examined age- and limb-related differences and motor unit (MU) firing behaviour of the vastus lateralis under tasks of varying difficulty. Twenty-one young (22 ± 4 years; 15 M, 6F) and seventeen older adults (74 ± 5 years; 12 M, 5F) performed constant and variable force unilateral isometric knee extensions. In both limbs, high-density surface electromyography signals were decomposed into MU spi

Beyond gait speed: a multidimensional motor signature of motoric cognitive risk syndrome identified through domain-specific anomaly detection.

Motoric cognitive risk (MCR) syndrome, defined by subjective cognitive complaints and slow gait speed, identifies older adults at increased risk of major neurocognitive disorders (NCDs). Yet, gait speed reflects a composite output shaped by heterogeneous neuromusculoskeletal and cognitive processes, limiting its clinical specificity. This study aimed to refine the motor signature of MCR by quantifying domain-specific gait deviations relative to a normative reference cohort using an anomaly-detection approach. Ninety-seven adults (≥ 55 years) completed two 3-min 30-s treadmill walking bouts at

Identifying cognitive subgroups in older adults from community data with hierarchical cluster analysis.

This study applied Ward's hierarchical clustering to detailed neuropsychological data from a community-recruited sample of 180 older adults to identify naturally occurring cognitive subgroups and characterize multidomain cognitive heterogeneity beyond conventional mild cognitive impairment (MCI) criteria. Twenty-two demographically adjusted z-scores spanning memory, language, executive function, and attention/processing speed were submitted to Ward's method, and the resulting four-cluster solution was cross-validated using an independent k-means analysis. The identified subgroups included an a

The impact of cumulative obesity and genetic susceptibility on biological aging in Chinese older adults.

The detrimental impacts of cumulative obesity exposures and their interactions with genetics on biological aging remain poorly understood. We included 687 older adults (median age: 79.6 years) from the Rugao Longitudinal Ageing Study (RLAS), a community-based cohort study conducted in Rugao, Jiangsu, China. The cumulative exposures of body mass index (cBMI) and waist circumference (cWC) were calculated as the excessive exposure of BMI and WC relative to their reference values (BMI ≥ 24 kg/m2; WC ≥ 90 cm for males and ≥ 80 cm for females) multiplied by their durations across the 2014, 2017, and

Spatial memory in Alzheimer's disease 5XFAD mice is enhanced by XPO1 inhibitor KPT-330.

The proteostatic decline in Alzheimer's disease is well established, and improvement in proteostasis could potentially delay cognitive impairment. One emerging entry point to modulate proteostasis is the regulation of nucleo-cytoplasmic partitioning of proteins across the nuclear pore via karyopherins. The nuclear exportin XPO1 is a key regulator of proteostasis by driving the assembly of ribosomes and by modulating the process of autophagy. We recently found that the XPO1 inhibitor KPT-330 (Selinexor), an FDA-approved drug against multiple myelomas, enhances proteostasis, leading to benefits

Sleep disturbances and Alzheimer's disease: a multiscale approach from exposome to neurobiology and precision medicine.

Sleep disturbances and Alzheimer's disease (AD) are interconnected public health challenges. However, the underlying mechanisms of their complex relationships remain elusive. Here, we propose a hypothetical integrative, stream-like model outlining how external and internal exposome factors accelerate brain aging, thereby exacerbating circadian dysregulation, orexin-mediated hyperexcitability, metabolic imbalance, and inflammaging. These changes can lead to increased sleep fragmentation and reduced slow-wave sleep, triggering widespread neuroinflammation, glymphatic dysfunction, and the accumul

Combined effects of ageing and type 2 diabetes mellitus on human dental mechanics.

Dental collagen, a protein subject to essentially zero turnover, is highly susceptible to the accumulation of advanced glycation end-products (AGEs) due to elevated blood glucose in type 2 diabetes mellitus (T2DM). Progressive loss of collagen's structural stability likely increases the risk of tooth decay and fracture complicating the proper fixation of dental restorations. Our study aimed at investigating the structural and mechanical alterations in T2DM-affected teeth by measuring the stiffness and detecting micro-traumas and fractures. By using computed tomography (CT) and static compressi

Cardiovascular risk factors and vascular neuropathological findings in primary age-related tauopathy versus Alzheimer's disease.

Cardiovascular risk factors (CVRFs) are prevalent and modifiable contributors to dementia risk; however, their contributions to vascular pathologies across dementia etiologies has not been examined extensively. This study assessed differences in the prevalence of CVRFs and vascular neuropathologies between people with primary age-related tauopathy (PART) and Alzheimer's disease neuropathology (ADNP), and whether CVRFs associated differentially with vascular co-neuropathologies and antemortem cognitive status between groups. 5144 individuals from the National Alzheimer's Coordinating Center wit

Intestinal T cells in aging: implications for gut barrier integrity and inflammaging.

Aging is accompanied by a progressive decline in intestinal barrier integrity, resulting in increased permeability to luminal microbes and microbial products and contributing to chronic low-grade inflammation ("inflammaging"). While epithelial and microbial changes have been extensively studied, the role of intestinal T cells as active regulators of barrier homeostasis during aging remains underappreciated. The gut harbors the largest population of T cells in the body, including diverse conventional and unconventional subsets that directly shape epithelial differentiation, mucus production, an

TTV virome marks immune exhaustion, α7nAChR alteration, and mortality in elderly patients with severe COPD.

Torque teno virus (TTV) viremia, part of the human virome, is emerging as a dynamic marker of immune competence, yet its links to immune exhaustion, cholinergic dysfunction, and poor outcomes in elderly patients with severe COPD remain unclear. In 102 elderly COPD patients (age 72.6 ± 8.8 years, GOLD stage 3-4) undergoing a 3-week pulmonary rehabilitation program, TTV load was measured, and peripheral blood mononuclear cells were analyzed for T-cell subsets and α7 nicotinic acetylcholine receptor (α7nAChR) expression. Patients with high TTV load (≥ 4 log10/mL) exhibited profound T-cell depleti

Aging effects on nigrostriatal structure, hemodynamics, and connectivity: implications for Parkinson's disease.

Aging is an important risk factor for Parkinson's disease (PD). Characterizing age-related alterations in the nigrostriatal system may help identify early vulnerability prior to overt neurodegeneration. We aimed to delineate aging trajectories of structure and hemodynamics of the nigrostriatal system and examine their associations with motor and cognitive functions. We analyzed 486 healthy adults from Human Connectome Project-Aging dataset, stratified into younger (≤ 60 years) and older (> 60 years) groups. Motor, cognitive, and motor cognition functions were assessed. Multiparametric MRI incl

Whole-brain cholinergic modulation following computerized cognitive training in healthy older adults: a [18F]FEOBV PET study.

The cholinergic system is central to attention, learning, and neuroplasticity and undergoes progressive degeneration with aging. Neuroplasticity-based computerized cognitive training is a scalable, non-pharmacological strategy to support cognitive health in older adults, yet its neurobiological mechanisms remain unclear. [18F]Fluoroethoxybenzovesamicol (FEOBV) positron emission tomography (PET) quantifies presynaptic cholinergic terminals via vesicular acetylcholine transporter (VACht) binding, providing an index of cholinergic integrity and neurotransmission capacity. This offers a unique opp

Epileptiform discharges in neurodegenerative diseases linked to atrophy but not associated with iron depositions.

Epileptiform discharges in Alzheimer's disease (AD) and Lewy body dementia (DLB) may be associated with cognitive decline. Iron accumulation has been implicated in neurodegenerative processes; however, its relationship to epileptiform discharges remains unknown. In addition, the relationship between iron deposition, structural brain changes, and epileptiform discharges remains unclear. The objective is to investigate whether iron deposition and structural brain atrophy are associated with epileptiform discharges in patients with AD, DLB, and healthy controls (HCs). A total of 25 patients with

Cognitive trajectories and mortality risk in older adults: a 9-year follow-up study.

Cognitive decline is a central feature of aging and a major determinant of disability and mortality. Understanding cognitive trajectories is essential for identifying individuals at risk and informing preventive strategies. In this study, we estimated transition probabilities across cognitive states and examined predictors of cognitive deficit and mortality among 1,413 community-dwelling older adults for nine years. Cognitive status was assessed at baseline and at two follow-up waves (2006, 2010, and 2015) using the Mini-Mental State Examination and categorized into four trajectories: stable,
