Osteoinductive substances in middle ear surgery.

There have been a number of stages in the development of cholesteatoma surgery. After the introduction of the radical operation the disadvantages of this procedure led to the development of more conservative operative techniques, for example the anterior-posterior approach with preservation of the posterior wall of the external ear canal or the obliteration of the mastoid cavity with muscle flaps or various materials. The assumption that organic fragments of the bone enable certain unspecified mesenchymal cells to form bone led to this investigation. Allogenic bone matrix and gelatin were impl

[Cytokine and disease].

Cytokine is a generic term of biologically active molecules which are mainly produced by the immune-competent cells and regulate the immune response, inflammation and hematopoiesis. This includes interleukins (IL), colony-stimulating factors (CSF), interferons (IFN), tumor necrosis factors (TNF) and so on. These cytokines are glycoproteins with a molecular weight of 20,000-40,000 kD and work at very low concentrations of pM order. ILs and CSFs transduce their signal via specific cell-membrane receptors which usually consist of at least two subunits and belong to a newly identified superfamily

Proliferation and differentiation of the small intestinal epithelium: from Petri dish to bedside.

The small intestinal epithelium is a continuously renewing cell system in which two main compartments can be defined: the crypt compartment and the villus compartment. The crypt compartment houses the proliferative cells (stem cells) that give birth to the four types of differentiated enterocytes (absorptive enterocytes, goblet cells, enteroendocrine cells, Paneth cells) that, with the exception of Paneth cells, populate the villus compartment. Enterocyte proliferation, differentiation and migration are regulated by a complex network of cell-cell and cell-matrix information, and modulated by g

Regulation of synovial cell growth by polypeptide growth factors.

In vitro, rheumatoid arthritis (RA) synovial cells display several of the characteristics of neoplastic and virally transformed cells. The recent observation that synovial cell cultures, derived from collagenase digests of synovial membranes from RA patients, proliferate in serum-free medium suggests that these cells have the capacity to synthesize those factors essential for their growth. Direct immunocytochemical staining and Western analysis have identified transforming growth factor-beta (TGF-beta) band and basic fibroblast growth factor (FGF) in the cytoplasm of RA and normal synovial cel

Peptic ulcer pathophysiology.

Despite extensive research, the etiology of peptic ulcer disease remains unclear. Given the multiple processes that control acid and pepsin secretion and defense and repair of the gastroduodenal mucosa, it is likely that the cause of ulceration differs between individuals. Acid and pepsin appear to be necessary but not sufficient ingredients in the ulcerative process. It is clear that the majority of gastric ulcers and a substantial number of duodenal ulcers do not have increased gastric acid secretion. Recent research has focused more on protection and repair of the stomach and duodenum. NSAI

Presence of an endothelioid tubelike structure at the interface of the amniotic membranes in twins with single and double placenta. Growth factors involvement.

A histomorphological study of the amniotic membranes in full-term twins with double and single placenta was carried out by means of the silver impregnation staining technique suitably modified. Specimens of interface of amniotic membranes were prepared by means of sections. The constant presence of a tubelike structure was observed. Proceeding from the amniotic cavity, the following histological layers were noted. 1) single layer of amniotic cells; 2) amorphic substance with fibrocytes; 3) single layer of endothelial cells. The same order of single layer is present in the amniotic membrane of

CXCL14-like immunoreactivity in growth hormone-containing cells of urodele pituitaries.

Immunohistochemical techniques were employed to investigate the distribution of a chemokine, namely, CXCL14-like immunoreactivity in the axolotl (Ambystoma mexicanum) and Japanese black salamander (Hynobius nigrescens) pituitaries. CXCL14-immunoreactive cells concentrated at an area of the pars distalis adjacent to the pars intermedia. We found that these cells correspond to the cells immunoreactive to an antibody against rat growth hormone (GH). Immunoelectron microscopy indicated that the CXCL14-like substance and GH coexisted on the secretory granules in the axolotl pituitary. Western blot

Oxyresveratrol, a Stilbene Compound from Morus alba L. Twig Extract Active Against Trichophyton rubrum.

Morus alba L. (mulberry) twig is known to have an inhibitory effect on pathogens in traditional Chinese medicine. In the present study, the dermophytic fungus, Trichophyton rubrum, was used to evaluate the inhibitory effect of total M. alba twig extract and extracts obtained using solvents with different polarities by the method of 96-well MTT colorimetry. The main active substance was isolated and identified by tracking its activity. In addition, the inhibitory effects of active extracts and a single active substance were investigated in combination with miconazole nitrate. Our data indicated

Amphibian studies to investigate the endocrine-disrupting properties of chemicals through the thyroid modality: a comparison of their statistical power.

Amphibians are the current model species for investigating the endocrine-disrupting (ED) properties through the thyroid modality in non-mammalian species. A recurrent question in the European Union (EU) regulatory endocrine assessment of pesticide active substances (2018/605) is whether the positive results from an in vivo screening test, that is, Amphibian Metamorphosis Assay (AMA), can be considered sufficient to conclude on the ED properties of a pesticide active substance or whether the larval amphibian growth and developmental assay (LAGDA) is a necessary step to further clarify the conce

Understanding the Molecular Basis of Pathological Retinal Angiogenesis: Roles of Lipids, Non-Coding RNAs, and Cellular Crosstalk.

Pathological retinal neovascularization is a prevalent cause of blindness, impacting millions of individuals across all age groups, from infants to older adults. It is a key clinical feature of several retinal diseases, such as diabetic retinopathy (DR), retinopathy of prematurity (ROP), age-related macular degeneration (AMD), and Coats disease. Researchers have extensively investigated the involvement of vascular endothelial growth factor (VEGF) signaling, hypoxia inducible factor (HIF) signaling, and inflammation in the regulation of pathological retinal neovascularization. Recent breakthrou

Isolation and characterization of Lactobacillus plantarum LP2301 from bovine milk: acid and bile tolerance, antibacterial activity, and genomic safety evaluation.

The long-term and non-standard use of antibiotics poses a significant threat to food safety and human health. Probiotics are a promising alternative to antibiotics, effectively combating bacterial infections while reducing the risks associated with antibiotic resistance. Gram staining confirmed LP2301 as a Gram-positive bacterium, and species identification as Lactobacillus plantarum was established by 16S rRNA gene sequencing and whole-genome sequencing. In this study, LP2301 was isolated from the milk of a healthy dairy cow, which exhibits characteristics of acid (pH = 2.5) and 0.3% bile sal

Herbivore pest under combined plant extract and cadmium effect, the case of Spodoptera exigua from control and cadmium strain.

Secondary plant substances like flavonoids found in host plant tissues or/and environmental contaminants, like cadmium, can pose a common challenge for polyphagous herbivore pests. Earlier pre-exposure experience, with specific stressor, lasting even through many generations may affect the response to the other stressor. In this study, combined the effect of cadmium and extracts of some common spices (ordered according to total flavonoid content: lovage > mentha > basilium), on Spodoptera exigua larvae from two strains, control, and cadmium (selected with a sublethal concentration of cadmium f

Antioxidant system alterations and oxidative stress caused by polyfluoroalkyl substances (PFAS) in exposed biota: a review.

Contamination of aquatic and terrestrial organisms by Perfluoroalkyl substances (PFAS), emerging contaminants, is widespread, as these compounds are present in water, soil, air, and food, owing to their environmental persistence. PFAS exposure induces biochemical process alterations associated with the disruption of the antioxidant defense system in several species. This review aims to discuss how PFAS-induced antioxidant system alterations lead to changes in biochemical processes in different organisms exposed to these pollutants. This disruption, then leads to an imbalance in antioxidant def

Key metabolites and mechanistic insights in forchlorfenuron controlling kiwifruit development.

Forchlorfenuron (CPPU) is a plant growth regulator widely applied on kiwifruit to improve yield, however, there are rarely reports on its effects on the nutrients of kiwifruits. Based on UHPLC-Q-TOF-MS, the effects of CPPU on metabolism profile and nutrient substances of two kiwifruit varieties during development were investigated by non-targeted metabolomics. A total of 115 metabolites were identified, and 29 differential metabolites were confirmed and quantified using certified reference standards. Metabolic profile indicated that CPPU promoted kiwifruit development during the main expansion

Sustainable advances on phosphorus utilization in soil via addition of biochar and humic substances.

The intervention of human in phosphorus pool seems to be a vicious circle. The rapid population growth leads to the global food shortage, which leads to the massive use of phosphate fertilizer and the continuous exploitation of phosphate rocks. With the massive loss and fixation of phosphate fertilizer in the soil, the unavailable phosphorus in the soil becomes superfluous, while the phosphate mineral resources turn to scarce. Interestingly, exogenous carbonaceous materials, notably, biochar and humic substances, have been widely used as soil conditioners in agricultural production up to date,

Targeted tumor therapies at a glance.

Conventional tumor therapy is usually based on surgery, radiation and chemotherapy. Treatment with chemotherapeutics is often impeded by dose-limiting toxicities. Therefore, medical scientists sought for tools to improve chemotherapy by directly coupling targeting molecules to cytotoxic substances. This review provides a general overview on the development of targeted drugs designed for tumor therapy. Further carrier-based delivery systems of anti-tumorigenic drugs will not be described here. The targeting moiety is usually an antibody or a fragment thereof. Growth factors, cytokines and ligan
