Huge symbiotic organs in giant scale insects of the genus Drosicha (Coccoidea: Monophlebidae) harbor flavobacterial and enterobacterial endosymbionts.

Giant scale insects (Drosicha: Coccoldea: Monophlebidae) were investigated for their symbiotic organs and bacterial endosymbionts. Two types of bacterial 16S rRNA gene sequences, flavobacterial and enterobacterial, were consistently detected in D. corpulenta and D. pinicola. The former sequences formed a compact clade in the Bacteroidetes, allied to the symbionts of cushion and armored scales. The latter sequences formed a robust clade in the gamma-Proteobacteria, allied to enteric bacteria like Enterobacter aerogenes and Escherichia coli. Another type of 16S sequence derived from Wolbachia wa

First experimental observations on the development of Rhabdochona denudata (Nematoda: Rhabdochonidae) in the intermediate host.

Two species of mayfly nymphs, Habroleptoides modesta (Hagen) and Habrophlebia lauta (Eaton) (Ephemeroptera: Leptophlebiidae), were found as suitable experimental intermediate hosts of the fish nematode Rhabdochona denudata (Dujardin, 1845) (Rhabdochonidae), an intestinal parasite of Palaearctic cyprinids. The parasite's eggs were obtained from nematode gravid females parasitizing the European chub, Leuciscus cephalus (L.), of the Rokytná River, Czech Republic. At the water temperature of 9 degrees C, the eggs of R. denudata were ingested by the intermediate host and the hatched, toothed (with

Genome-wide identification, expression profiling, and functional analysis of sugar transporter family genes in Diaphorina citri.

Sugar transporters (STs) are important proteins responsible for the movement of sugar through the cell membrane and play pivotal roles in the growth, development, and osmoregulation of phloem-feeding hemipterans. However, information about STs in the Asian citrus psyllid (ACP), Diaphorina citri Kuwayama (Hemiptera: Liviidae), remains unknown. In this study, a total of 61 STs were identified according to the genome and transcriptome analysis of D. citri. All the ST genes have complete open reading frames (ORFs), and most of them have 12 transmembrane regions. Chromosome localization and phyloge…

The Begomovirus Disease Tetrahedron: Weeds as the Missing Dimension in Virus Epidemiology.

Begomoviruses are among the most destructive plant viruses, causing substantial yield losses across diverse cropping systems. Their epidemiological success is driven by high genetic plasticity, broad host range, and efficient transmission by the whitefly vector Bemisia tabaci. Traditional epidemiological models based on the classical disease triangle (virus-host-vector) fail to fully capture the ecological and evolutionary complexity of begomovirus pathosystems. Increasing evidence highlights the critical role of non-cultivated plants, particularly weeds, as persistent reservoirs that maintain

Proteomic analysis may explain differences in Citrus × limon and Citrus × sinensis susceptibility to Trioza erytreae.

The psyllid Trioza erytreae is a vector for Huanglongbing, one of the most destructive citrus diseases worldwide. Citrus × limon is the preferred host, although the mechanisms underlying this preference remain poorly understood. This study compared the responses of C. × limon 'Eureka' and C. × sinensis 'Valencia' plants to T. erytreae infestation, specifically to nymph feeding. The number of successfully developed nymphs observed in C. × limon was three times that of 'Valencia' plants. The enriched vascular sap proteome of young leaves from infested and control plants was compared using nanosc

Periodical cicadas suffer legacy effects of century-old forest removal.

Periodical cicada (Magicicada spp.) ranges have continued to decline even as their habitat has rebounded from historic lows. This counterintuitive trajectory may be a consequence of Magicicada's limited dispersal capabilities combined with their requirement that individual host trees survive across decades. Indeed, we find that the presence of Magicicada breeding populations is best predicted by tree cover in 1930, not by present-day tree cover, and that cicadas have largely not recolonized restored habitat even after nearly 100 years. A model based on empirical measures of tree cover turnover

What you eat is what we need: using ants to detect spotted lanternfly (Lycorma delicatula) DNA.

BACKGROUND: Early detection of invasive species such as the spotted lanternfly (SLF, Lycorma delicatula) is critical for effective management including eradication efforts and limiting further spread. SLF excretes honeydew containing detectable DNA, providing a unique opportunity to leverage environmental DNA (eDNA) for its detection. This study introduces the ant-derived DNA (antDNA) approach, utilizing ants as 'honeydew samplers' to detect SLF DNA. We validated the effectiveness of this method through three experiments. RESULTS: Using SLF-specific polymerase chain reaction (PCR), we consist

Geographic variation and diversity of bacterial endosymbionts in Asian citrus psyllid, Diaphorina citri, from Iran.

BACKGROUND: The Asian citrus psylla (ACP, Diaphorina citri), a destructive insect, poses a significant threat to citrus industries worldwide. As the primary vector of huanglongbing (HLB), ACP infestations have caused devastating economic losses and declines in citrus production across many regions. Despite the role of endosymbionts in psyllid biology and HLB transmission, their geographic distribution in Iran remains uncharacterized. In this study, the composition of bacterial endosymbiont communities associated with ACP was examined across four geographic regions in Iran (Sarbaz, Roudan, Fary

Overexpression of CYP6CX4 contributing to field-evolved resistance to flupyradifurone, one novel butenolide insecticide, in Bemisia tabaci from China.

Bemisia tabaci is a formidable insect pest worldwide, and exhibits significant resistance to various insecticides. Flupyradifurone is one novel butenolide insecticide and has emerged as a new weapon against B. tabaci, but field-evolved resistance to this insecticide has become a widespread concern. To unravel the mechanisms of field-evolved flupyradifurone resistance, we conducted a comprehensive investigation into susceptibility of twenty-one field populations within the Beijing-Tianjin-Hebei Region of China. Alarmingly, thirteen of these populations displayed varying degrees of resistance, r

Knockdown of the salivary protein gene NlG14 caused displacement of the lateral oviduct secreted components and inhibited ovulation in Nilaparvata lugens.

Saliva plays important roles in insect feeding, but its roles in insect reproduction were rarely reported. Here we reported that the knockdown of a salivary gland-specific gene NlG14 disrupted the reproduction through inhibiting the ovulation of the brown planthopper (BPH), Nilaparvata lugens (Stål), one of the most devastating rice pests in Asia. NlG14 knockdown caused the displacement of the lateral oviduct secreted components (LOSC), leading to the ovulation disorder and the accumulation of mature eggs in the ovary. The RNAi-treated females laid much less eggs than their control counterpart

Extracts of Euphorbia nivulia Buch.-Ham. showed both phytotoxic and insecticidal capacities against Lemna minor L. and Oxycarenus hyalinipennis Costa.

Many phytochemicals can affect the growth and development of plants and insects which can be used as biological control agents. In this study, different concentrations of crude, hexane, chloroform, butanol, and aqueous extracts of Euphorbia nivulia Buch.-Ham., an endemic plant of the Cholistan desert in South Punjab of Pakistan, were analysed for their chemical constituents. Their various concentrations were also tested for their phytotoxic and insecticidal potential against duckweed, Lemna minor L., and the dusky cotton bug, Oxycarenus hyalinipennis Costa. various polyphenols, i.e., quercetin

Physiology of crapemyrtle bark scale, Acanthococcus lagerstroemiae (Kuwana), associated with seasonally altered cold tolerance.

Temperature is one of the most important abiotic factors influencing the adaptation and diversification of insects. Diverse and complex physiological mechanisms have evolved to help insects adapt to seasonal changes in temperature and prevent cold injury. Although the mechanisms of seasonal adaptation to low temperatures have been studied for insects in different taxa, none of these mechanisms have been investigated in scale insects in the superfamily Coccoidea. The crapemyrtle bark scale, Acanthococcus lagerstroemiae (Kuwana) (Hemiptera: Eriococcidae), is a newly introduced scale pest of crap…

From urban edge to forest core: how non-native species restructure ecological networks.

The Permanent Preservation Area (PPA) of Morro da Glória, Brazil, is crucial for conserving biodiversity. Nevertheless, its location near an urban area makes it vulnerable to invasion by non-native species. This study reports, for the first time, an ecological interaction involving multiple species within this PPA, including the invasive ant Tetramorium bicarinatum, the whitefly Aleurothrixus floccosus, the non-native fungus Penicillium coffeae, and two host plants: the exotic Citrus sinensis and the native Eugenia uniflora. This interaction has not previously been documented in the literature

Stable nutritional endosymbiosis across cryptic diversity of a leafhopper species complex.

BACKGROUND: Ancient nutritional symbioses underpin the ecological success of many sap-feeding insects. In 'true hoppers' - the hemipteran suborder Auchenorrhyncha, obligate bacterial partners provide essential amino acids lacking in plant phloem diets. However, the stability and persistence of such associations across the diversity of hoppers are poorly understood, and investigations are often complicated by insufficiently resolved host identity. RESULTS: Here, we combined multitarget amplicon sequencing, metagenomics, and microscopy to assess the compositional and functional diversity of the…

A co-delivery nanosystem of dsRNA and nitenpyram for insecticide resistance management in Nilaparvata lugens.

Nilaparvata lugens has evolved high resistance to neonicotinoid insecticides such as nitenpyram (NTP), driven by detoxification-associated genes. RNA interference (RNAi) can suppress these genes and restore insecticide susceptibility, but delivering double-stranded RNA (dsRNA) in the field remains a major challenge because naked dsRNA degrades within hours. To overcome this barrier, a layered double hydroxide (LDH)-based nanocarrier for co-delivery of dsRNA and NTP (dsRNA@NTP@LDH) was developed. The nanoplatform was synthesized by one-step co-precipitation and electrostatic dsRNA adsorption, t

Summer-form susceptible, winter-form resistant: differential susceptibility of Cacopsylla chinensis nymphs to Beauveria bassiana.

INTRODUCTION: The pear psylla Cacopsylla chinensis is a destructive pear pest whose nymphs exhibit seasonal polymorphism between summer-form and winter-form morphs. Whether the two morphs differ in susceptibility to Beauveria bassiana and the underlying mechanisms have not been characterized. METHODS: Bioassays at three spore concentrations (2.5×106, 2.5×107, and 2.5×108 spores/mL) were conducted to compare fungal virulence against fourth-instar nymphs of both morphs. Hemocyte counts and transcriptome sequencing were performed at 24 h and 36 h post-infection, respectively. qRT-PCR and enzymat

Wolbachia-mediated viral transmission enhancement in insect vectors.

The white-backed planthopper (Sogatella furcifera) serves as the vector for the southern rice black-streaked dwarf virus (SRBSDV), with varying transmission efficiencies across populations. This research identifies Wolbachia, a common insect symbiont, as a key facilitator in breaching the salivary gland barrier for SRBSDV, revealing a mechanism by which the virus exploits an insect symbiont for transmission. Through field and laboratory investigations, it was observed that high-transmission (HT) planthopper populations contained high levels of Wolbachia, while low-transmission (LT) populations

High susceptibility of Diaphorina citri first instar nymphs to Bacillus thuringiensis-derived pesticidal proteins from multiple structural groups.

The Asian citrus psyllid, Diaphorina citri, is the primary vector of Candidatus Liberibacter asiaticus, the causative agent of citrus greening disease, which continues to threaten citrus production worldwide. Current management relies heavily on chemical insecticides, despite increasing concerns regarding resistance and environmental impacts. Because pathogen acquisition is most efficient during the nymphal stage, management strategies targeting immature psyllids may improve disease suppression. Although bacterial pesticidal proteins (BPP) have generally shown limited activity against D. citri

The fungicide jinggangmycin stimulates fecundity of Nilaparvata lugens Stål via ILP/Foxo signaling.

The brown planthopper (BPH), Nilaparvata lugens is a typical pesticide-induced resurgent rice pest. A previous study showed that a fungicide, jinggangmycin (JGM)-treated rice led to markedly increased sugar content and (Insulin-like Peptide 2) ILP2 in response to sugar-mediated TOR signaling and stimulated fecundity in BPH. However, the role of the other ILPs in response to types of carbohydrate compounds remained poorly understood. The present studies showed that JGM treatment led to strongly increased glucose content and slightly increased sucrose and trehalose contents in rice plants. The 5
