Sodium Alginate Modification as a Promising Strategy for Inducing Oral Immune Tolerance to Fish Gelatin-Based Cell Scaffolds to Control Allergenic Risks.

The allergenic risks of fish gelatin-based cell scaffolds remain an unresolved issue. The study proposed a sodium alginate modification strategy to solve allergenic risks by inducing oral immune tolerance. The strategy significantly reduced antibody production, mast cell degranulation, inflammatory mediator release, and Th2 immune response. Peptidomics and bioinformatics prediction revealed that the strategy protected CD4+ T cell epitopes (FTGASGLPG) and CD8+ T cell epitopes (e.g., GATGPTGL). Additionally, intestinal barrier function, integrity, and Th1/Th2/Th17/Treg immune balance were restor…

Modified Guilu Erxian Glue restores immune tolerance in aplastic anemia by reprogramming T cell differentiation via the miR-146a/STAT1/SOCS1 axis.

OBJECTIVE: Two traditional Chinese medicine (TCM) formulas are Guilu Erxian Glue (GEG) and Danggui Buxue Tang (DBT). Their combination, Modified Guilu Erxian Glue (MGEG), is used to treat aplastic anemia (AA). This study aims to clarify how MGEG restores immune tolerance in AA mice. Specifically, we studied whether MGEG can regulate the differentiation and maturation of T cell lineage by regulating the transcription of miR-146a to rebuild the stable state of bone marrow immune microenvironment, inhibit the immune response mediated by T helper (Th)1 cells, and enhance the immunosuppressive func…

Effects of an infant formula containing a whey protein concentrate on feeding tolerance and markers of intestinal immune defense in Chinese infants.

BACKGROUND: Human milk (HM) bioactive components can have immune modulatory functions, impact the gut microbiome, and may result in functional benefits when added to infant formula (IF). In this single-arm, prospective, intervention study, we tested the effectiveness of an IF with a whey protein concentrate co-enriched in α-lactalbumin, milk fat globule membrane (MFGM), and Sn-2 palmitate resulting in protein and lipid profiles observed in HM. The outcomes tested were feeding tolerance, Bifidobacteria abundance, and intestinal and immune health of Chinese infants. METHODS: Predominantly formu

HEPLISAV-B Breaks Immune Tolerance and Induces HBV Control via CD4 T Cell-Dependent Mechanisms in a Chronic Hepatitis B Mouse Model.

BACKGROUND: Chronic hepatitis B virus (HBV) infection (CHB) affects nearly 300 million individuals globally and remains incurable with current antiviral therapies, which suppress viral replication but rarely achieve functional cure defined by sustained loss of hepatitis B surface antigen (HBsAg). CHB is characterized by profound virus-induced immune tolerance that limits the efficacy of conventional therapeutic vaccination strategies. OBJECTIVE: To evaluate the therapeutic efficacy and immunological mechanisms of HEPLISAV-B, a CpG-1018-adjuvanted HBsAg vaccine, in breaking immune tolerance an…

A model based on chitinase 3-like protein for expecting liver severity of hepatitis B virus infections in the immune tolerance phase.

BACKGROUND: The question of whether to treat patients with chronic hepatitis B (CHB) during the immune tolerance (IT) period is a matter of ongoing debate, as it is difficult to discern different levels of liver disease severity. We created and assessed a novel diagnostic model for identifying significant liver tissue damage in individuals with CHB in IT phase. METHODS: From November 2018 to December 2022, a cross-sectional study of 311 patients with chronic hepatitis B virus infection (HBV DNA > 30 IU/mL) at Ningbo No. 2 Hospital, Ningbo, China, who underwent liver biopsy, including 44 patie…

Invariant Natural Killer T Cells Regulate Conventional Dendritic Cell Maturation to Re-establish Immune Tolerance to Rheumatoid Arthritis in DBA/1 Mice.

Rheumatoid arthritis (RA) is a type of autoimmune disease that results in immune disorder and excessive inflammatory response due to a reduction of self-tolerance. Invariant natural killer T (iNKT) cells can effectively alleviate clinical symptoms and hyper-inflammation in RA, but their mechanism of action is not well-defined. This study aims to investigate the mechanism of iNKT cell therapy for RA. We established a DBA/1 mouse model for RA and treated it with specific iNKT cells. A cytometric bead array was used to measure the amounts of cytokines in the serum. Flow cytometry was then employe

Eradication of FIX inhibitor in haemophilia B children using low-dose immune tolerance induction with rituximab-based immunosuppressive agent(s) in China.

INTRODUCTION: Development of haemophilia B inhibitors (HBI) results in the ineffectiveness of FIX replacement therapy. Inhibitor eradication by immune tolerance induction (ITI) is therefore necessary. In HBI, ITI even at high FIX dose is less effective and has a higher risk of severe complications. AIM: To characterize clinical features and outcome of ITI on HBI. METHODS: This retrospective study was conducted in Haemophilia Paediatric Comprehensive Care Centre of China. We used low-dose ITI (25-50 FIX IU/kg/three-times-weekly to every-other-day) with domestic prothrombin complex concentrate…

Use of Polymeric Nanoparticle Platform Targeting the Liver To Induce Treg-Mediated Antigen-Specific Immune Tolerance in a Pulmonary Allergen Sensitization Model.

Nanoparticles (NPs) can be used to accomplish antigen-specific immune tolerance in allergic and autoimmune disease. The available options for custom-designing tolerogenic NPs include the use of nanocarriers that introduce antigens into natural tolerogenic environments, such as the liver, where antigen presentation promotes tolerance to self- or foreign antigens. Here, we demonstrate the engineering of a biodegradable polymeric poly(lactic- co-glycolic acid) (PLGA) nanocarrier for the selective delivery of the murine allergen, ovalbumin (OVA), to the liver. This was accomplished by developing a…

Evidence for contribution of CD4+ CD25+ regulatory T cells in maintaining immune tolerance to human factor IX following perinatal adenovirus vector delivery.

Following fetal or neonatal gene transfer in mice and other species immune tolerance of the transgenic protein is frequently observed; however the underlying mechanisms remain largely undefined. In this study fetal and neonatal BALB/c mice received adenovirus vector to deliver human factor IX (hFIX) cDNA. The long-term tolerance of hFIX was robust in the face of immune challenge with hFIX protein and adjuvant but was eliminated by simultaneous administration of anti-CD25+ antibody. Naive irradiated BALB/c mice which had received lymphocytes from donors immunised with hFIX developed anti-hFIX a…

Altered or Impaired Immune Response to Hepatitis B Vaccine in WNIN/GR-Ob Rat: An Obese Rat Model with Impaired Glucose Tolerance.

Obesity is shown to increase the incidence and severity of infectious diseases and individuals seem to exhibit poor antibody response to vaccination due to several inherent immune defects. With the increasing prevalence of impaired glucose tolerance (IGT) seen in obese individuals, the present study was aimed to investigate the basal immune response and immune response upon Hepatitis B vaccination (HBV) in an obese rat model WNIN/GR-Ob with impaired glucose tolerance (IGT). Decreased proportions of splenic CD4(+) T helper cells and CD3(+) T cells were observed in obese animals compared to lean

From discovery to precision tolerance: Re-imagining patient-matched regulatory T cells in shaping of a roadmap for personalized immune re-balancing.

The recognition of FOXP3 as the master regulator of regulatory T cells (Tregs) established the genetic and functional basis for peripheral immune tolerance. Contemporary advances in single-cell genomics and epigenetic mapping have further refined this landscape, revealing distinct, tissue-adapted Treg subsets and identifying the Treg-specific demethylated region (TSDR) as the critical molecular marker for durable lineage stability. The transition toward personalized Treg medicine has become feasible through an integrated pipeline that maps individual cellular signatures and employs precision e…

Peripheral Blood Mononuclear Cell Transcriptomics in Porcine Reproductive and Respiratory Syndrome: A Window into Innate Immune Resistance and Tolerance to Viral Disease.

Peripheral blood mononuclear cells (PBMCs) are the most immunologically active and readily accessible fraction of whole blood, and they initiate host immune responses following infection and vaccination. Since PBMC transcriptomes capture diverse host-pathogen interactions, they offer a promising tool to uncover the genetic drivers of viral resistance and tolerance. As a detailed case study of this principle, we examine porcine reproductive and respiratory syndrome (PRRS), which remains one of the most economically important viral diseases of swine worldwide. Following PRRS virus (PRRSV) exposu

Analysis of Specific IgG4 Antibody and sIgG4/sIgE Antibody Ratio as Marker of Immune Tolerance to IgE-Mediated Response in Allergic Children.

BACKGROUND: The diagnosis and treatment of allergic diseases remain challenging because the results of allergy testing do not always correlate with clinical manifestations. For instance, although a specific IgE (sIgE) test for a particular allergen may be positive, some patients remain asymptomatic due to the development of immune tolerance. Recently, specific IgG4 (sIgG4) has been suggested as a potential indicator of immune tolerance. However, data in children are limited. Therefore, this study aimed to evaluate the clinical usefulness of sIgG4 testing and its potential role in assessing imm…

iPSC-derived cells lack immune tolerance to autologous NK-cells due to imbalance in ligands for activating and inhibitory NK-cell receptors.

BACKGROUND: Dozens of transplants generated from pluripotent stem cells are currently in clinical trials. The creation of patient-specific iPSCs makes personalized therapy possible due to their main advantage of immunotolerance. However, some reports have claimed recently that aberrant gene expression followed by proteome alterations and neoantigen formation can result in iPSCs recognition by autologous T-cells. Meanwhile, the possibility of NK-cell activation has not been previously considered. This study focused on the comparison of autologous and allogeneic immune response to iPSC-derived c
