[11C]Carbon dioxide was prepared from [11C]cyanide. First, [11C]CH4 was produced by the (p, alpha) reaction on a N2/H2 mixture and was converted to [11C]CN- by reaction with ammonia over platinum at 1273 K. The [11C]CN- was adsorbed on a cobalt(II, III) oxide/ceramic furnace material at room temperature and was subsequently converted to [11C]CO2 by heating the cobalt(II, III) oxide to 948 K over a 10 min period and collecting the [11C]CO2 in a trap at 77 K. Specific activities as high as 600 mCi/mumol at end of bombardment (EOB) were obtained in a 15 microA irradiation for 10 min.

A quantitative method has been developed for valproic acid and twelve of its metabolites using capillary gas chromatography--mass spectrometry with selected-ion monitoring. The method is applicable to serum or urine and all metabolites are measured in a single run. Ions selected for quantitative purposes were the characteristic (M-57)+ ions of the tert.-butyldimethylsilyl (tBDMS) derivatives. The 4-hydroxyvalproic acid was measured as the gamma-lactone. Tin is an essential ingredient of most technetium-99m radiopharmaceutical preparations but its in-vivo distribution and long-term fate are not well understood. beta-Lactamase from Bacillus licheniformis 749/c was used as a marker for ELISA. Conjugates of the beta-lactamase with the capsule antigen of the plague causative agent and with the monoclonal antibodies to it were prepared by glutaric aldehyde "linking". The conjugates preserved high immunospecific and beta-lactamase activity. High sensitivity of the modification of ELISA allowed detecting up to 8 ng/ml of the capsule antigen of the plague causative agent. The enzymatic activity of the beta-lactamase was determined microiodometrically which provided visual recording of the results of ELISA.

A new and novel method for determination of serum acetaminophen is described. The assay, which can be completed in less than 5 min, is based on the enzymatic hydrolysis of acetaminophen, with subsequent colorimetric detection of the aminophenol so produced. In this method, we use the Amicon MPS-1 centrifugal ultrafiltration device and the YMB membrane in measuring free testosterone in serum. Two independent assays are combined: total testosterone and the ultrafiltrable fraction of added [3H]testosterone. The unbound fraction is determined in 0.15-0.5 mL ultrafiltrates of 0.6 to 1 mL of variably diluted serum that has been equilibrated with [3H]testosterone at 37 degrees C. We have devloped an enzyme-linked immunosorbent assay for determining choriomammotropin (human placental lactogen) in serum. Unlabeled hormone competes with choriomammotropin-beta-galactosidase conjugate for antibody bound to polystyrene tubes. The entire assay can be performed in 2.5 h with good precision. The coefficient of variation for one sample with a mean concentration of 5.6 mg/L, assayed 10 times on the same day, was 5.7%. A number of 3-keto bile acids were synthesized by the selective oxidation of bile acid methyl esters with silver carbonate-Celite in refluxing toluene. The pure 3-keto bile acids were isolated simply by filtering the reaction mixture and concentrating the filtrate. The relation of the bile acid structure to the oxidation rate is also discussed.

A sensitive method for the quantitative determination of piperazine is described. The method is precise and responds linearly from 25 g to 500 mug and above of the material. The procedure is based on the formation of a complex of piperazine with reineckate in neutral or acid medium. The complex can be separated by centrifugation. It is then dissolved in acetone and estimated at 530 nm in a colorimeter. Piperazine present in trichloroacetic acid extractrs of biological samples can also be estimated by this method.

Erythrocytes of a patient with hereditary erythroblastic multinuclearity with a positive acidified serum test (HEMPAS) were tested with many seed extracts and various other reagents. Serological evidence of membrane abnormality was confirmed. Various anti-H reagents reacted relatively poorly with HEMPAS cells. HEMPAS cells have both enhanced i and depressed H antigens. A brief note on a 'new' anti-H lectin (Cytisus glabrescens) is provided.
