Insecticide resistance and malaria transmission: divergent effects on Anopheles vector competence.

The global reduction of malaria burden through the use of insecticides in the form of insecticide-treated nets (ITNs) or indoor residual spraying has been challenged with insecticide resistance among species of Anopheles mosquitoes. The challenge is due to resistance mechanisms such as target site mutation, metabolic resistance, cuticular thickening, and behavioral resistance. Despite few studies reporting significant influence of insecticide exposure on vector competence-a mosquito's ability to infect, harbour, and transmit Plasmodium - the relationship between insecticide exposure in mosquit…

Policy-practice gaps and barriers to the effective management of insecticide resistance in Tanzania.

BACKGROUND: Malaria control gains are stagnating or reversing, in part due to widespread insecticide and drug resistance, as well as increasing financial and implementation challenges. This study assessed the national strategies for insecticide resistance monitoring and management in Tanzania to identify key barriers and misalignments between policy and actual practice. METHODS: We initially reviewed technical documents from Tanzania's National Malaria Control Programme (NMCP) produced between 2014 and 2024, along with World Health Organization (WHO) guidelines and reports on insecticide resi

Discovery of Novel Isoxazoline Derivatives Containing Pyrazolamide Fragment as Insecticidal Candidates.

Lepidopteran species cause significant harm to agricultural yields and food quality. In this study, a series of innovative isoxazoline derivatives incorporating pyrazolamide moieties were carefully designed and synthesized as potential insecticidal agents. Among these, compound F16 demonstrated an LC50 value of 0.01 mg/L against Plutella xylostella, surpassing that of the lead compound fluxametamide (LC50 = 0.15 mg/L). Furthermore, F16 exhibited broad-spectrum insecticidal activity against Pyrausta nubilalis, Spodoptera frugiperda, Chilo suppressalis, Aphis craccivora, and Sogatella furcifera.

Extract of Tangerine Peel as a Botanical Insecticide Candidate for Smallholder Potato Cultivation.

BACKGROUND: Contemporary agriculture heavily relies on synthetic chemicals to ensure high yields and food security; however, their overuse has led to health issues and the development of pesticide resistance in pests. Researchers are now exploring natural, eco-friendly alternatives for pest control. METHODS: This study evaluated two ethanol-based formulations (1.25% and 2.50%, v/v) derived from the tangerine peel (Citrus reticulata L. var. Clementina) against conventional chemical treatments and an untreated control group in the cultivation of potatoes (Solanum tuberosum L. var. Capiro). A ra

Monitoring of DNA structural changes after incorporation of the phenylpyrazole insecticide fipronil.

The use of insecticides presents a risk to the environment because they can accumulate in the water, soil, air, and organisms, endangering human and animal health. It is therefore essential to investigate the effects of different groups of insecticides on individual biomacromolecules such as DNA. We studied fipronil, which belongs to the group of phenylpyrazole insecticides. The interaction of fipronil with calf thymus DNA was investigated using spectroscopic methods (absorption and fluorescence spectroscopy) complemented with infrared spectroscopy and viscosity measurement. Fluorescence emiss…

Implementation of malaria vector surveillance and insecticide resistance monitoring interventions in Nigeria.

Malaria vector surveillance is required to determine disease transmission dynamics, vector insecticide susceptibility status, suitable control strategies and impact of control interventions. However, capacity and resources for vector surveillance and insecticide resistance monitoring is often inadequate in most countries at risk of vector-borne diseases. Collaborations and linkages between malaria control policy makers and existing research institutions generating vector surveillance research data are often weak, thereby hindering the availability of data for decision-making. A national vector

Structure and bioactivity of an insecticidal trans-defensin from assassin bug venom.

Disulfide-rich peptides such as defensins play diverse roles in immunity and ion channel modulation, as well as constituting the bioactive components of many animal venoms. We investigated the structure and bioactivity of U-RDTX-Pp19, a peptide previously discovered in venom of the assassin bug Pristhesancus plagipennis. Recombinant Pp19 (rPp19) was found to possess insecticidal activity when injected into Drosophila melanogaster. A bioinformatic search revealed that domains homologous to Pp19 are produced by assassin bugs and diverse other arthropods. rPp19 co-eluted with native Pp19 isolated

Elucidating the insecticidal mechanisms of zein nanoparticles on Anticarsia gemmatalis (Lepidoptera: Erebidae).

Previous research suggested that positively charged zein nanoparticles [(+)ZNP] were toxic to neonates of Anticarsia gemmatalis Hübner and deleterious to noctuid pests. However, specific modes of action for ZNP have not been elucidated. Diet overlay bioassays attempted to rule out the hypothesis that A. gemmatalis mortality was caused by surface charges from component surfactants. Overlay bioassays indicated that negatively charged zein nanoparticles [(-)ZNP] and its anionic surfactant, sodium dodecyl sulfate (SDS), exhibited no toxic effects when compared to the untreated check. Nonionic zein

Strong Positive Selection in Aedes aegypti and the Rapid Evolution of Insecticide Resistance.

Aedes aegypti vectors the pathogens that cause dengue, yellow fever, Zika virus, and chikungunya and is a serious threat to public health in tropical regions. Decades of work has illuminated many aspects of Ae. aegypti's biology and global population structure and has identified insecticide resistance genes; however, the size and repetitive nature of the Ae. aegypti genome have limited our ability to detect positive selection in this mosquito. Combining new whole genome sequences from Colombia with publicly available data from Africa and the Americas, we identify multiple strong candidate sele

Implications of environmentally shaped microbial communities for insecticide resistance in Sitobion miscanthi.

Insect-associated bacteria play an important role in the resistance to pesticides, yet bacterial community compositions in wild insect host populations and the environmental factors that shape them are mostly elusive. In this study, Sitobion miscanthi (Takahashi) populations were collected from major wheat growing regions in China. Following high-throughput sequencing of 16S rRNA gene fragments, association analyses were performed within the bacterial community associated with S. miscanthi, as well as with population resistance levels to four commonly used pesticides and different environmenta
