Differential expression of the 27 kDa cathepsin L-like cysteine protease in developmental stages of Spirometra erinacei.

The 27 kDa cathepsin L-like cysteine protease of Spirometra erinacei plerocercoid is known to play an important function in tissue penetration, nutrient uptake and immune modulation in human sparganosis. In the present study, the expression of this enzyme was examined at different developmental stages of S. erinacei including immature egg, coracidium, plerocercoid in tadpole and rat, and adult. Proteolytic activity against carboxybenzoyl-phenylalanyl-arginyl-7-amino-4-methylcoumarin was detected in the extracts of coracidia and plerocercoid while no activity was observed in those of immature e

Mercury and selenium distribution in key tissues and early life stages of Yellow Perch (Perca flavescens).

Whereas early life stages are usually considered as particularly sensitive to both organic and inorganic contaminants, field studies assessing contaminant bioaccumulation in these stages are scarce. Selenium (Se) is thought to counteract Hg toxic effects when it is found at Se:Hg molar ratios above 1. However, the variation of this ratio in key fish tissues of different early life stages is mostly unknown. The present study therefore aimed to assess Hg and Se content in gravid female tissues (gonads, muscle, liver, gut, and brain) and different life stages (egg masses, newly hatched larvae (NH

Transient receptor potential channel activation targets multiple stages of the schistosome parasite.

Schistosomiasis, a neglected tropical disease caused by infection with parasitic blood flukes of the genus Schistosoma, affects more than 250 million people worldwide. Currently, patients rely on a single medicine, praziquantel (PZQ), discovered half a century ago. To combat drug resistance and reach elimination of the schistosome parasite, alternative drugs to PZQ are required that treat and prevent disease. Here, we describe a drug candidate, M5493, with potent activity against multiple parasite life cycle stages. The target is a parasite transient receptor potential ion channel, distinct fr…

Genetic evidence for the microphallid trematode Maritrema gratiosum in the Northeast Pacific and an overview of its global distribution.

The trematode Maritrema gratiosum Nicoll, 1907 has a heteroxenous life cycle comprising a marine gastropod as its first intermediate host, a barnacle as its second intermediate host, and a bird as its definitive host. Globally, the species is widely distributed from subtropical waters of the South China Sea to arctic waters of the Barents Sea. In European waters (namely, western Europe and the Barents Sea), the complete life cycle of M. gratiosum is known. Elsewhere in its range, however, its life cycle remains incompletely understood. Clarifying its distribution and resolving unknown host ass

Oocyst: knowns and unknowns about the lengthiest life stage of the malaria parasite.

The oocyst is the longest life stage of Plasmodium, the causative agent of malaria, one of the most persistent and devastating infectious diseases of humankind. Following ingestion during blood feeding, parasites reproduce sexually and traverse the mosquito midgut epithelium to differentiate into oocysts on the basal lamina, where they undergo prolonged development, ultimately giving rise to thousands of sporozoites capable of infecting a new human host. Oocyst formation represents a severe population bottleneck, resulting in the lowest parasite numbers observed across the parasite life cycle.

A Novel Plasmodium berghei Protein, LSMP, Regulates Malaria Liver Stage Maturation.

Malaria parasites undergo extensive replication in hepatocytes before initiating the pathogenic blood stage of infection. Although this clinically silent phase is essential for parasite expansion, the molecular mechanisms regulating late liver stage maturation remain incompletely understood. Here, we characterize a previously unannotated protein (PBANKA_0620800), which we designate liver stage maturation protein (LSMP). LSMP was expressed during schizont, sporozoite, and liver stages and localized predominantly to the parasite cytosol. Using reverse genetic approaches, we generated LSMP KO par

Life Cycle and Mass Occurrence of Paracolopha morrisoni (Hemiptera: Aphididae) in South Korea.

Paracolopha morrisoni (Baker, 1919) (Aphididae: Eriosomatinae) is a gall-forming aphid that alternates between Zelkova serrata (Thunb.) Makino (Ulmaceae) and bamboo hosts in East Asia. Here, we provide field-based documentation of its seasonal biology and host alternation in South Korea, supported by DNA barcoding. Subterranean colonies on the roots of Pseudosasa japonica (Siebold & Zucc. ex Steud.) Makino ex Nakai (Poaceae), together with observations of migratory alates and larviposition, confirmed this bamboo as a secondary host. Phyllostachys heterocycla (Carrière) Matsum. (Poaceae) was id

Comparative Toxicity and Residual Activity of Botanical and Synthetic Insecticides Against Multiple Life Stages of Aleurodicus dispersus (Hemiptera: Aleyrodidae).

The spiraling whitefly, Aleurodicus dispersus Russell (Hemiptera: Aleyrodidae) is a highly polyphagous and invasive pest that poses serious challenges to crop protection, particularly in tropical and subtropical agroecosystems. Given the rising concern over pesticide resistance and environmental risks associated with conventional insecticides, this study was conducted under controlled laboratory conditions to compare the efficacy of selected botanicals, fish oil rosin soap (FORS), organic salt, and synthetic insecticides against multiple life stages of A. dispersus. An ISWI-related chromatin remodeller regulates stage-specific gene expression in Toxoplasma gondii.

ATP-dependent chromatin remodellers are specialized multiprotein machines that organize the genome in eukaryotic cells and regulate its accessibility by repositioning, ejecting or modifying nucleosomes. However, their role in Toxoplasma gondii is poorly understood. Here we show that T. gondii has evolved two divergent proteins within the imitation switch (ISWI) family: TgSNF2h and TgSNF2L. TgSNF2h specifically forms a core complex with the transcription factor AP2VIII-2 and the scaffold protein TgRFTS. Depletion of TgRFTS phenocopies the knockdown of TgSNF2h, restricting access to chromatin an

Plasmodium falciparum PP1 phosphatase is a key regulator of malaria parasite transmission.

UNLABELLED: For the successful transmission of malaria parasites from humans to mosquitoes, Plasmodium falciparum gametocytes must remain in the bloodstream long enough to be taken up by a mosquito. Once ingested, they are then activated into gametes to continue the parasite life cycle in the mosquito midgut. Both persistence of gametocytes in the blood and their activation into gametes are tightly regulated by phospho-signaling. While the serine-threonine phosphatase PfPP1 is an essential enzyme for asexual blood stage parasite proliferation, its role during transmission of sexual stages rema

The Human Blood Fluke, Schistosoma mansoni, Harbors Bacteria Throughout the Parasite's Life Cycle.

While symbiotic relationships between invertebrates and bacteria have been extensively described, studies of microbial communities inhabiting parasitic worms remain scarce. Exploring the microbiota associated with helminths responsible for major infectious diseases will inform on parasite biology, host-pathogen interactions, and disease pathophysiology. We investigated the presence of microorganisms inhabiting tissues of the human parasite Schistosoma mansoni. In situ hybridization using a pan-bacterial 16S rRNA gene probe revealed bacteria colonizing key developmental stages that were success

The branched chain amino acids (BCAAs) modulate the development of the intra-cellular stages of Trypanosoma cruzi.

The life cycle of Trypanosoma cruzi, the etiological agent of Chagas disease, involves different forms of the parasite, which alternates between insect and vertebrate hosts. One critical process in the parasite's life cycle is metacyclogenesis, in which the replicative non-infective forms present in the insect midgut differentiate into non-dividing vertebrate-infective forms. It is known that proline (Pro) is important for this process and that leucine (Leu) and isoleucine (Ile) can act as inhibitors of metacyclogenesis. In this study, we investigated further the role of branched-chain amino a

Genetic Ablation of a Female-Specific Apetala 2 Transcription Factor Blocks Oocyst Shedding in Cryptosporidium parvum.

The apicomplexan parasite Cryptosporidium is a leading global cause of diarrheal disease, and the infection poses a particularly grave threat to young children and those with weakened immune function. Infection occurs by ingestion of meiotic spores called oocysts, and transmission relies on fecal shedding of new oocysts. The entire life cycle thus occurs in a single host and features asexual as well as sexual forms of replication. Here, we identify and locus tag two Apetala 2-type (AP2) transcription factors and demonstrate that they are exclusively expressed in male and female gametes, respec

Life cycle assessment of waste-to-hydrogen systems for fuel cell electric buses in Glasgow, Scotland.

Waste-to-hydrogen (WtH) technologies are proposed as a dual-purpose method for simultaneous non-fossil-fuel based hydrogen production and sustainable waste management. This work applied the life cycle assessment approach to evaluate the carbon saving potential of two main WtH technologies (gasification and fermentation) in comparison to the conventional hydrogen production method of steam methane reforming (SMR) powering fuel cell electric buses in Glasgow. It was shown that WtH technologies could reduce CO2-eq emissions per kg H2 by 50-69% as compared to SMR. Gasification treating municipal s

Schistosoma mansoni larvae in vitro cultures using Biomphalaria glabrata extracts.

Schistosomiasis is one of the most prevalent waterborne parasitic diseases affecting humans. In natural conditions, snails are necessary for maintenance of its lifecycle and also required as intermediate hosts to maintain the lifecycle in laboratory settings. In the present study, the location of S. mansoni larvae in Biomphalaria glabrata snails after infection (inoculation of miracidia) was investigated. Larvae were found located in the head-foot (HF) area of B. glabrata snails at 10 days post-infection (DPI), then their location was predominantly changed to the hepatopancreas and ovotestis (

Eukaryotic Initiation Factor 2α Kinases Regulate Virulence Functions, Stage Conversion, and the Stress Response in Entamoeba invadens.

Entamoeba histolytica is a protozoan parasite that causes amoebic dysentery and liver abscess. This pathogen possesses a two-stage life cycle consisting of an environmentally stable cyst and a pathogenic amoeboid trophozoite. Since infection is acquired by ingestion of cysts from contaminated food and water, this parasite is prevalent in underdeveloped countries. A reptilian pathogen, Entamoeba invadens, which can encyst in culture, has long served as a surrogate to study stage conversion. In the host, Entamoeba species must manage stress, including nutrient deprivation and host immune pressur

Redistribution of FLAgellar Member 8 during the trypanosome life cycle: Consequences for cell fate prediction.

The single flagellum of African trypanosomes is essential in multiple aspects of the parasites' development. The FLAgellar Member 8 protein (FLAM8), localised to the tip of the flagellum in cultured insect forms of Trypanosoma brucei, was identified as a marker of the locking event that controls flagellum length. Here, we investigated whether FLAM8 could also reflect the flagellum maturation state in other parasite cycle stages. We observed that FLAM8 distribution extended along the entire flagellar cytoskeleton in mammalian-infective forms. Then, a rapid FLAM8 concentration to the distal tip
